Abstract

Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis. Indonesia is one of the developing countries with high TB patients in the world. The most common detection used for TB diagnosis is sputum smear examination. Smear negative pulmonary TB still have a risk of infection and can develop into active. The GeneXpert molecular examination is a rapid diagnosis of TB by the real-time PCR method. The purpose of this study was to assess the validity of GeneXpert in smear-negative pulmonary TB compared to Lowenstein Jensen's culture. The design of this diagnostic test study is a cross-sectional study. The study was conducted on 40 people with smear-negative pulmonary TB suspect patients in Dr. Doris Sylvanus Regional Hospital. Sputum examination was performed with GeneXpert and compared with Lowenstein Jensen culture. The results of GeneXpert validity for diagnosing smear-negative pulmonary TB suspect are sensitivity 81.8%, specificity 96.5%, positive predictive value 90%, negative predictive value 93.3%, and accuracy 92.5%. It was concluded that GeneXpert has sensitivity, specificity, positive predictive value, negative predictive value, and high accuracy as a diagnostic tool in smear-negative pulmonary TB suspects.

Abstract

Background: Tuberculosis (TB) is a chronic infectious disease caused by the bacterium Mycobacterium tuberculosis. The sputum Acid-Fast Bacillus (AFB) examination is a widely used diagnostic test, but it is difficult to control a minimum of 5,000 germs / ml of sputum to get a positive result. The GeneXpert MTB / RIF examination is an automatic machine with easy and fast use that uses the principle of real-time nested PCR and molecular technology for the examination of Mycobacterium tuberculosis (MTB) and rifampicin resistance (RR). Purpose: To determine the relationship between the results of sputum AFB examination and the results of rifampicin resistance GeneXpert MTB / RIF in pulmonary TB patients at Doris Sylvanus Hospital, Palangka Raya for the period 2018-2019. Methods: This study used an analytic observational method with a cohort study design. The subjects of this study were all pulmonary TB patients who had been examined for sputum AFB and GeneXpert MTB / RIF at Doris Sylvanus Regional Hospital for the period 2018-2019. Data analysis used SPSS program with Chi Square test (p <0.05). Results: The results of statistical tests showed that the Sputum BTA result variable had a significant relationship with the GeneXpert MTB / RIF outcome variable. So  the result is p = 0.009. Conclusion: There is a significant relationship between the results of sputum AFB examination and the results of rifampicin resistance of GeneXpert MTB / RIF in pulmonary TB patients at Doris Sylvanus Hospital, Palangka Raya for the period 2018-2019.

Abstract

The Dayak tribe of Kalimantan is known as the root of Saluang Belum (Lavanga sarmentosa) a traditional medicinal plant that has properties to increase stamina, and sexual arousal and is used as an alternative medicine to increase male fertility. L.sarmentosa is consumed by drinking the boiled water of the plant's roots. The aim of this study is to determine the effect infusion roots of Saluang Belum (L.sarmentosa) in increasing the spermatids as seen in the tubulus seminiferus histological of mice (Mus musculus). The root will be extracted by the infusion method with different doses for each group, namely 200 mg/KgBB, 400 mg/KgBB, and 600 mg/KgBB, and negative controls (aquades) which was carried out for 15 days to mice. On the day 16, the right testis was taken and histological preparations were made with hematoxylin-eosin staining and observed under a microscope at 400x magnification in five fields of view. Compounds contained in the infusion of L.sarmentosa are alkaloids, saponins, triterpenoids, flavonoids, phenolics, and steroids. The number of spermatids in the tubulus seminiferus histological of mice increased statistically significantly from the doses of 200 mg/KgBB, 400 mg/KgBB, and 600 mg/KBBg. Conclusion: Infusion of the roots L.sarmentosa can increase the number of spermatids seen in the tubulus seminiferus histological of mice.

Abstract

The antigen rapid diagnostic test is used for screening COVID-19 patients and is easy to do anywhere and saves time. The sensitivity of various brands of rapid antigen test kits can vary, so medical personnel need to pay close attention to them. Testing for the SARS-CoV2 antigen kit needs to be done diagnostically. This study aims to determine the specificity, sensitivity, and accuracy of the Haelgen antigen rapid test through diagnostic tests. The study design was cross-sectional, with a total sample of 60 people who took swabs from the nasopharynx and oropharynx. The sample is placed in the viral transport medium (VTM). Samples were examined by RT-PCR and, at the same time, examined using an antigen rapid test. The results of the analysis were carried out by calculating the sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), and accuracy of the SARS-Cov2 antigen test compared to standard RT-PCR using a diagnostic test formula. The most common characteristics of COVID-19 patients were female and 20–29 years old (28.3%). The results of the Haelgen rapid antigen diagnostic test showed a sensitivity of 92.31%, a specificity of 100%, a positive predictive value of 100%, a negative predictive value of 87.5%, and an accuracy of 95%. The sensitivity, specificity, and accuracy of the Haelgen rapid antigen are suitable for testing for COVID-19.

Abstract

Background: HAIs are infections that are obtained from hospitals or other health care facilities, such as health centers. According to the Ministry of Health, in Indonesia it reaches 15.74%, much higher than in developed countries, which ranges from 4.8 to 15.5%. According to the Indonesian Ministry of Health, the average is 8.1%. At the Puskesmas, the Emergency Unit (ER) is the place most at risk for HAIs, for patients undergoing treatment using less sterile equipment, such as minor surgical equipment. Object: To determine the presence or absence of bacteria and to identify the types of bacteria in minor surgical medical devices in the ER Pahandut Public Health Center, Palangka Raya City. Methods: This study used a descriptive quantitative design method, to determine the presence or absence of bacteria in sterilized minor surgical medical devices. Results: The identification results found gram-positive bacteria in 3 samples of minor surgical instruments, namely 2 samples identified as Staphylococcus aureus and 1 sample identified as Streptococcus sp. Conclusion : There are bacterial contaminants in minor surgical medical devices used in the ER Pahandut Public Health Center, Palangka Raya City.

Abstract

Anemia saat ini merupakan salah satu dari 6 masalah gizi yang ada di Indonesia terutama pada remaja putri. Anemia pada remaja putri berdampak buruk terhadap kesehatan dan reproduksi, menurunnya prestasi belajar, serta tidak tercapainya tinggi badan maksimal. Penelitian ini bertujuan untuk mengetahui perbedaan kadar hemoglobin remaja putri antara sebelum dan sesudah pemberian tablet tambah darah di SMP Islam Nurul Ihsan dan MTs Darul Ulum tahun 2023. Metode penelitian yang digunakan adalah quasi eksperiment dengan dengan pendekatan one grop pretest-postest design, pengambilan sampel secara proportional random sampling. Pengumpulan data dilakukan dengan wawancara dan pemeriksaan kadar hemoglobin menggunakan alat point of care testing (POCT). Populasi dari penelitian ini adalah semua siswi kelas 7 yang ada di SMP Islam Nurul Ihsan dan MTs Darul Ulum kecamatan Pahandut kota Palangka Raya yang berjumlah 129 siswi dan didapat sebanyak 62 sampel. Hasil uji Wilcoxon didapat perbedaan antara kadar hemoglobin sebelum dengan sesudah pemberian tablet tambah darah dengan nilai p sebesar 0,001.

Abstract

Antioksidan dalam mendetoksifikasi radikal bebas akan ikut teroksidasi sehingga untuk mencukupi kebutuhan tubuh diperlukan antioksidan eksogen. Antioksidan eksogen dapat diperoleh dari tanaman herbal salah satunya Bajakah Kalalawit. Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb.) adalah salah satu jenis bajakah yang terdapat di Kelurahan Marang, Kalimantan Tengah yang berpotensi sebagai antioksidan. Penelitian ini bertujuan untuk menganalisis aktivitas antioksidan dan kekuatan antioksidan Bajakah Kalalawit (Uncaria gambir (W. Hunter) Roxb.). Batang Bajakah Kalalawit diekstrak dengan metode maserasi 3x24 jam menggunakan pelarut etanol 96%. Setelah didapatkan ekstrak kental kemudian dianalisis kandungan senyawa metabolit sekunder dengan uji fitokimia. Uji aktivitas antioksidan dibuat dengan konsentrasi ekstrak 20ppm, 40ppm, 60ppm, 80ppm, dan 100ppm dengan metode DPPH (2,2-diphenyl-1-picrylhydrazyl). Hasil penelitian menunjukkan senyawa yang terdapat dalam ekstrak etanol Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb) adalah terpenoid, flavonoid, fenolik, steroid, saponin, alkaloid, dan tanin. Ekstrak etanol Bajakah Kalalawit memiliki aktivitas antioksidan yang sangat kuat dengan nilai IC50 8,69ppm.

Abstract

Hiperglikemia kronis menyebabkan terjadinya peningkatan reaksi glikasi  glukosa dengan protein. Produk akhir glikasi yaitu AGEs (Advanced Glycation End Products) berperan dalam komplikasi diabetes. Hasil skrining fitokimia yang telah dilakukan sebelumnya menunjukkan bahwa batang Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb.) mengandung flavonoid, terpenoid, tanin, alkaloid yang diduga berpotensi sebagai antiglikasi. Penelitian ini bertujuan untuk mengetahui potensi antiglikasi ekstrak etanol batang Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb.). Metode: Jenis penelitian adalah true eksperimental. Model reaksi glikasi menggunakan BSA dan glukosa sebagai kelompok kontrol. Ekstrak etanol 96% batang Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb.) dibuat menjadi larutan konsentrasi 100, 200, 400, 600, 800 dan 1000ppm dan ditambahkan ke model reaksi glikasi, selanjutnya diukur absorbansi menggunakan spektrofotometer, dan dihitung nilai potensi antiglikasi. Hasil rerata potensi antiglikasi ekstrak Bajakah Kalalawit berdasarkan ANOVA berbeda signifikan (p<0.05) untuk setiap kenaikan konsentrasi ,yaitu konsentrasi 100, 200, 400, 600, 800 dan 1000ppm memiliki potensi 42,131%, 47,541%, 57,541%, 67,377%, 70,328%, 80,656% dengan nilai IC50 253,043ppm. Kesimpulan: Ekstrak etanol batang Bajakah Kalalawit (Uncaria gambir (W.Hunter) Roxb.) memiliki potensi antiglikasi dengan konsentrasi efektif 100ppm.

Abstract

Salah satu penyebab infertilitas adalah penurunan kualitas sperma yang disebabkan oleh stress oksidatif. Pemberian antioksidan eksogen yang didapatkan dari akar saluang belum (Luvunga sarmentosa) dapat menurunkan stres oksidatif. Penelitian terkait bioaktivitas Luvunga sarmentosa terhadap motilitas spermatozoa in vitro masih terbatas. Untuk mengetahui pengaruh fraksi metanol ekstrak akar saluang belum terhadap motilitas spermatozoa secara in vitro dan konsentrasi efektif untuk meningkatkan motilitas spermatozoa. Sampel yang digunakan adalah spermatozoa manusia in vitro sebanyak 15 sampel yang dicuci dan ditambahkan medium BWW (Biggers, Whitten, and Whittingham) kemudian diinkubasi pada suhu 37℃ selama 1 jam dengan fraksi methanol ekstrak akar Saluang Belum dengan konsentrasi 100, 500, 1000, 5000 ng/ml serta kelompok kontrol (BWW). Selanjutnya, dilakukan pemeriksaan di mikroskop untuk melihat gerak motilitas spermatozoa. Hasil penelitian menunjukkan bahwa fraksi metanol ekstrak akar Luvunga sarmentosa mengandung senyawa seperti alkaloid, amino acid, phenylpropanoid, dan terpenoid. Motilitas spermatozoa meningkat secara signifikan pada konsentrasi 100, 500, 1000 dan 5000 ng/mL dan kelompok kontrol. Fraksi metanol akar Luvunga sarmentosa efektif pada konsentrasi 100 ng/mL.. Fraksi metanol akar saluang belum Luvunga Sarmentosa meningkatkan motilitas spermatozoa. Fraksi metanol, saluang belum, motilitas spermatozoa.

Abstract

Akar Salung Belum (Luvunga sarmentosa) mengandung antioksidan yang dapat mencegah kerusakan sel akibat radikal bebas. Pengujian efek senyawa terhadap spermatozoa untuk menilai kemampuan viabilitas dapat dilakukan melalui analisis integritas membran dengan pendekatan in vitro. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol 96% Saluang Belum (Luvunga sarmentosa) terhadap integritas membran spermatozoa secara in vitro. Penelitian spermatozoa manusia in vitro yang dicuci dan ditambahkan medium BWW kemudian diinkubasi pada suhu 37℃ selama 1 jam dengan ekstrak etanol 96% akar saluang belum pada perlakuan konsentrasi, 100 ng/mL, 200 ng/mL, 400 ng/mL, 800 ng/mL serta kelompok kontrol (hanya BWW). Integritas membran dinilai pada masing-masing 100 uL yang telah ditambahkan 1 mL HOST (Hypo-osmotic swelling test) di inkubasi 37°C selama 30 menit dan diamati menggunakan mikroskop cahaya dengan perbesaran 400x untuk melihat integritas membran spermatozoa dari ekor yang menggembung. Hasil penelitian menunjukkan bahwa senyawa yang terdapat di dalam ekstrak etanol 96% akar Saluang Belum (Luvunga Sarmentosa) adalah terpenoid, flavonoid, steroid, fenolik dan alkaloid. Jumlah viabilitas sperma meningkat secara signifikan dari konsentrasi 100 ng/mL (68 ±3,99) , 200 ng/mL (72,2 ±4,26) , 400 ng/mL (77± 4,76),dan 800 ng/mL (80±4,87). Meningkat jika dibandingkan dengan kontrol (63±3,81). Ekstrak etanol 96% akar Saluang Belum (Luvunga sarmentosa) yang diberikan secara invitro memiliki pengaruh untuk meningkatkan integritas membran spermatozoa.
