Abstract

This study was aimed to produce recombinant growth hormone (rGH) from giant grouper (Epinephelus lanceolatus), giant gouramy (Osphronemus gouramy) and common carp (Cyprinus carpio) and compare their bioactivity potential by means of inducing the growth hormone of juvenile Nile tilapia (Oreochromis niloticus) as the model. DNA fragment encoding mature GH protein of giant grouper (El-mGH), giant gouramy (Og-mGH) and common carp (Cc-mGH) was amplified by PCR method. The purified PCR products were ligated to pCold-1 to generate pCold/El-mGH, pCold/OgmGH, and pCold/Cc-mGH protein expression vector, respectively. Each of the expression vectors was transformed into the Escherichia coli BL21. E. coli BL21 was cultured using 2xYT medium and protein production was induced by cold shock at 15±1oC for overnight. The inclusion bodies of E. coli transformants containing protein expression vector were isolated by sonication method, and rGH production was analyzed by SDS-PAGE. Juvenile of Nile tilapia of average body weight of 12.41±3.28 g was intramuscularly injected once a week for 4 weeks with 1 μg inclusion body containing rGH per gram fish body weight. The result showed that rGH in molecular weight of about 25 kDa was obtained. Fish injected with rGH of El-mGH, Cc-mGH and Og-mGH grew 20.94%, 18.09%, and 16.99% faster, respectively, compared with the control. This result indicated that the three rGH produced in E. coli possessed biological activity when tested on Nile tilapia and further research is needed to find its effect on the growth of other aquaculture fish species.

Abstract

The Cyca-DAB1*05 allele of major histocompatibility complex class II genes is recently suggested to have a link with the European common carp strain resistant to koi herpesvirus (KHV). In this study, a set of specific primers for Cyca-DAB1*05 was designed and applied as a marker to identify broodstocks of majalaya common carp strain subsequently used as a candidate resistant to KHV infection. From a total of 23 broodstock subjected to PCR analysis, two female and male fish, both having (P) and no Cyca-DAB1*05 (N), were selected and then diallelly mated. Disease resistance of progenies from 10 crosses was determined by a survival analysis in pond rearing and a laboratory challenge-test using cohabitation method. The results have revealed that the average survivals of PxP progenies for pond rearing and KHV challenge test were 86% and 100% higher (P < 0.05) respectively compared to that of NxN fish. Survival rate of PxN/NxP progenies was significantly lower (P < 0.05) than that of PxP fish. Furthermore, PCR analysis showed that almost 91% progenies of PxP crosses seemed to have a KHV resistant gene marker. Thus, this study suggests that the marker is associated with the KHV resistance in majalaya common carp strain, and farming of PxP progenies can be useful to increase common carp production.

Abstract

Eicosapentaenoic acid (EPA, 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3) have important nutritional benefits in humans. EPA and DHA are mainly derived from fish, but the decline in the stocks of major marine capture fishes could result in these fatty acids being consumed less. Farmed fish could serve as promising sources of EPA and DHA, but they need these fatty acids in their diets. Generation of fish strains that are capable of synthesizing enough amounts of EPA/DHA from the conversion of α-linolenic acid (LNA, 18:3n-3) rich oils can supply a new EPA/DHA source. This may be achieved by over-expression of genes encoding enzymes involved in HUFA biosynthesis. In aquaculture, the successful of this technique would open the possibility to reduce the enrichment of live food with fish oils for marine fish larvae, and to completely substitute fish oils with plant oils without reducing the quality of flesh in terms of EPA and DHA contents. Here, three genes, i.e. Δ6-desaturase-like (OmΔ6FAD), Δ5-desaturase-like (OmΔ5FAD) and elongase-like (MELO) encoding EPA/DHA metabolic enzymes derived from masu salmon (Oncorhynchus masou) were individually transferred into zebrafish (Danio rerio) as a model to increase its ability for synthesizing EPA and DHA. Fatty acid analysis showed that EPA content in whole body of the second transgenic fish generation over-expressing OmΔ6FAD gene was 1.4 fold and that of DHA was 2.1 fold higher (P<0.05) than those in non-transgenic fish. The EPA content in whole body of transgenic fish over-expressing OmΔ5FAD gene was 1.21-fold, and that of DHA was 1.24-fold higher (P<0.05) than those in nontransgenic fish. The same patterns were obtained in transgenic fish over-expressing MELO gene. EPA content was increased by 1.30-fold and DHA content by 1.33-fold higher (P<0.05) than those in non-transgenic fish. The results of studies demonstrated that fatty acid content of fish can be enhanced by over-expressing gene encoding enzymes involved in fatty acid biosynthesis, and perhaps this could be applied to tailor farmed fish as even better sources of valuable human food.

Abstract

Vaccination is a practical step in preventing diseases caused by koi herpes virus (KHV) in koi fish (Cyprinus carpio haematopterus). We have developed two DNA vaccines for KHV named as GP-25 and GP-11 from two local isolates coded as ORF25 and ORF81, respectively. Although both vaccines have been reported to increase survival rates, the evaluation of koi fish immune responses at the molecular level has not been done post-vaccinations. The aim of this research was to determine the effects of koi herpesvirus DNA vaccine on the immune-modulation of koi fish at mRNA level. This recent research used the best vaccine doses of both vaccines determined from our previous study: 7.5 and 12.5 µg per 100 g fish of GP-11, and 12.5 µg per 100 g fish of GP-25. The immune gene expression was analyzed using the RT-qPCR method from the fish liver at 0, 1, 7, 14, and 28 days post-vaccination (dpv). The results showed that, in the vaccinated fish, the immune genes viz. tumor necrosis factor a (TNFa), interleukin-1b (IL1b), interferon-g (IFNg), Mx1, immunoglobulin Mu chain (IgM), and major histocompatibility complex (MHC) class I and class II were induced to significant extents. The higher dose vaccination using the GP-11 vaccine showed higher immune gene expression than that of the lower dose. Furthermore, the GP-25 vaccine had induced lower immune responses than the GP-11 vaccine when using the same dose of vaccination, but relatively the same when the half-dose of GP-11 vaccine was used. In conclusion, the GP-11 and GP-25 vaccine provided the immune-modulatory effects on the koi fish immune response after vaccination.

Abstract

Javaen barb is a native fish in Indonesian inland water with economic value and the potential to be developed as cultured fish resources and ornamental commodities. In the development of aquaculture, there are still problems, such as the low adaptability of the larvae, so that their survival is also low. This study was conducted to evaluate the effect of melatonin supplementation on the productivity of Javaen barb fish juveniles in culture containers. This study was conducted experimentally using a completely randomized design consisting of four treatments with different doses of melatonin supplementation, i.e., A) control (without melatonin supplementation), B) 0.2 mg/100 g of feed, C) 0.4 mg/100 g feed, and D) 0.6 mg/100 g feed, each treatment was repeated three times. Javaen barb juveniles were kept in an aquarium measuring 30×36×60 cm with a water level of 40 cm, consisting of 12 units. Each aquarium was stocked with 150 individuals and given an aeration system with the same air pressure intensity. Feeding was carried out ad-libitum with a frequency of three times daily for 180 days of rearing. The results showed that melatonin supplementation of as much as 0.6 mg/100 g of feed was able to increase the growth of Javaen barb fish juvenile with the highest survival rate (81.33 ± 0.54 %) and feed conversion ratio (2.61 ± 0.14). Melatonin supplementation of 0.6 mg/100 g of feed had higher total leukocyte (3.41±0.73×104 cells/mm3) and hemoglobin (5.07±0.12 g%) values and provided the best production performance in Javaen barb juveniles.

Abstract

The study investigated the influence of the different LED light spectrums on the maturation of the gonads in female synodontis broodstock and examined the maturation-related gene expression levels. Female synodontis were exposed to four different LED light spectra, specifically: white, blue, green, and red, for a continuous period of 120 days rearing. This study employed a completely randomized design, consisting of four treatments and five individual fish as replication for each treatment. An analysis was conducted on the gonadosomatic index (GSI), hepatosomatic index (HSI), gonad histology, estradiol levels, kiss2 and gnrh2 mRNA expression levels. The utilization of blue LED light treatment is highly effective in enhancing the reproductive parameters in female synodontis fish. The results show that brain and gonad kiss2 mRNA expression levels are not significantly different (P<0.05), while gnrh2 mRNA expression levels were significantly different (P<0.05) and had the highest expression in the gonads. The results suggest that light exposure can induce changes in the expression levels of kiss2 and gnrh2 mRNA, as well as control reproduction.

Abstract

Neon tetra, Paracheirodon innesi, is endemic to rivers and streams in southeastern Colombia, eastern Peru, and western Brazil and is commercially traded as aquarium fish in the world. In Indonesia, neon tetras were mass produced in Bojongsari District, Depok, West Java as the centre of neon tetra fish production. Understanding their genetic variation is useful for implementing their selective breeding programs, environmental restoration, and estimating genetic contributions in stocks. The current study aimed to investigate the patterns of morphometric and molecular diversity using randomly amplified polymorphic DNA-polymerase chain reaction techniques among farmed broodstocks of P. innesi in Indonesia. Three populations, namely: the Bojongsari, Curug, and Pondok Petir derived from the Bojongsari District, Depok, west Java, were used in the study. Thirty live fish from each location were analyzed based on 25 truss morphometric characters. Sixty fresh fish samples were obtained for DNA analysis using the RAPD-PCR technique, which uses three random primers. Principal component analysis (PCA) was performed to distinguish morphometric variations among populations. Morphological and molecular analysis displayed a similar result that Bojongsari and Pondok Petir neon tetra fish had high similarities, while Curug neon tetra was distinguished from others. The closest genetic distance was between the Pondok Petir and Curug populations (0.4088), while the farthest genetic distance was between the Curug and Bojongsari populations (0.4138). The results will be useful in developing breeding programs to improve broodstock quality.
