Abstract

The residue of 17a-methyltestosterone (MT) in fish has been a concern related to food and the environment safety. This study aimed to determine sex ratio of Nile tilapia treated by MT immersion at 36 °C water temperature, and measure the level of testosterone in fish body. Three hundreds of ten-day-old post hatched larvae was immersed in one liter of water containing 2 mg MT, at 26 °C and 36 °C. Immersion was performed for 2 and 4 hours. The results showed that immersion at 36 °C and increasing immersion time increased male percentage. Higher male percentage was obtained in treatment 4 hours immersion at 36 °C (92.5% male) and the effective treatmen 4 hours immersion at room temperature. By ELISA method, testosterone level in MT treated fish decreased sharply to 88.2% at one month after immersion, and being the same as control without MT immersion. Thus, masculization in tilapia could be performed by MT immersion at 36 °C water temperature for 4 hours, and it is most likely that no MT residual in fish after one month post immersion. Keywords: Nile tilapia, sex reversal, temperature, 17a-methyltestosterone

Abstract

African catfish (Clarias gariepinus) is one of the economically valuable aquaculture fish species in Indonesia. This research was aimed to produce F0 transgenic catfish carrying masou salmon Δ6-desaturase-like (OmΔ6FAD) gene. The Δ6-desaturase enzyme is involved in highly unsaturated fatty acid biosynthesis. Transgenic catfish was produced by sperm-mediated gene transfer using electroporation method. In this study, as the first step, sperms were electroporated with three different OmΔ6FAD concentration (25, 50, and 100 µg mL-1) to have the highest sperm viability after electroporation (125 V/cm, pulse frequency 5 times, pulse length 30 millisecond, pulse interval 0.1 second). The highest sperm viability and sperm carrying OmΔ6FAD were obtain at 100 µg mL-1. This concentration was then used to produce F0 transgenic catfish in the second step. Sperm motility, sperm viability, fertilization rate, hatching rate, and larval survival at 14 days after hatching were the same as the controls (p>0.05). Genomic DNA was extracted from caudal fin and then used as template to identify transgenic F0 by PCR method using specific primer for OmΔ6FAD gene. The PCR result showed that 53.84% of F0 carried OmΔ6FAD gene. The result of fatty acid analysis showed that EPA and DHA contents of F0 transgenic fish and non-transgenic fish were similar. Keywords: catfish, Δ6-desaturase-like gene, fatty acids, electroporation

Abstract

The aim of the present study was to investigate the growth performance of Nile tilapia immersed in 17α-methyltestosterone (MT) and recombinant giant grouper growth hormone (rElGH), and fed a rElGH-enriched diet. A total of 200 tilapia larvae aged 10 days and 14 days post hatching was immersed in 1 liter of 30 g/L saline water for 3 minutes, then in freshwater containing either MT, MT+rElGH, or rElGH for 4 hours. The MT dose was 2.0 mg/L and rElGH was 2.5 mg/L. As control was treatment without MT and rElGH. Fish were maintained in 250-L aquariums for 6 weeks and then moved to net cages for 8 weeks of rearing. The rElGH-enriched diet was fed to the 6-week-old fish for a month, and then fed a non-enriched diet. Each treatment was triplicates. The result showed that the daily growth rate (DGR), biomass gain (BM), survival (SR) and feed (artemia nauplii) consumption during maintenance in the aquariums in the rElGH and MT+rElGH treatments were higher (P<0.05) than in the control. The higher DGR and BM at net cage rearing and lower feed conversion ratio (P<0.05) were also found in MT and MT+rElGH treatments. None of the treatments showed any difference in survival (P>0.05). Profits estimation in the MT and MT+rElGH treatments were 63.38% and 57.91% higher than the control. The present study concluded that higher Nile tilapia farming performance could be obtanied by MT immersion in the larval phase and feeding juvenile on the rElGH-enriched diet.

Abstract

Transfection and electroporation method shave a high possibility to apply towards transgenic production of small eggs size fish species.  This study aimed to examine the potential of transfection and electroporation methods to use for transferring a foreign gene into betta fish (Betta splendens) embryos using green fluorescent protein (GFP) gene as a model.  Fish were spawned naturally in the ratio of male: female was 1:1, then a total of 200 eggs were taken for each treatment.  Transfection was performed for 30 minutes (room temperature of about 25 °C) at two-cell stage of embryos using transfast reagent.  Transfection reaction consisted of 0.75 µL transfast reagent, 0.25 µL GFP expression vector (DNA concentration: 50 µg/µL) and 99 µL NaCl solution (concentration: 0,95%).  Electroporation was performed using 125 volt cm-1, 3 times pulse frequency at one second interval and pulse length of 7 micro seconds.  A volume of 800 µL GFP expression vector solution (DNA concentration: 50 µg/ µL) in PBS was used for electroporation.  The successful of foreign gene transfer was determined by PCR method with GFP specific primers.  The results showed that hatching rate of eggs in transfection treatment was 67.08%, while the electroporation was 72.09%.  Survival of larvae in transfection treatment was 73.00%, while the electroporation was 75.00%.  The results of PCR analysis showed that transfection method allowed 65% of the survived fish carrying GFP gene, whereas the electroporation method was 70%.  Thus, foreign gene transfer in betta fish can be conducted using the transfection and electroporation methods.
