Abstract

The objective of this research was to know the growth and survival rate of redclaw crayfish (Cherax quadricarinatus) reared in recirculation system with density 20, 30, 40 and 50 m-2. Lobster with 6.02 ± 0.13 cm length and 6.23 ± 0.51 gram initial body weights were cultured in 60 x 30 x 40 cm aquarium and compiled in recirculation system, for 42 days. Result of research showed that there are statistically difference at growth rate daily weight, coefficient of variances and feed efficiency (p0.05). From this research it can be concluded that the best density for freshwater crayfish (Cherax quadricarinatus) was 50 m-2.Keywords: density, growth, survival rate, redclaw crayfish,  Cherax quadricarinatus ABSTRAKPenelitian ini bertujuan untuk mengetahui pertumbuhan dan kelangsungan hidup lobster capit merah (Cherax quadricarinatus) yang dipelihara pada sistem resirkulasi dengan kepadatan 20, 30, 40 dan 50 ekor/m2. Benih lobster yang digunakan memiliki panjang awal rata-rata 6,02 ± 0,13 cm dan berat 6,23 ± 0,51 gram, dipelihara pada akuarium dengan ukuran 60 x 30 x 40 cm yang diisi air setinggi 20 cm dan disusun dalam sistem resirkulasi, selama 42 hari. Hasil penelitian menunjukkan bahwa terdapat perbedaan nyata pada laju pertumbuhan bobot harian, koefisien keragaman dan efisiensi pakan (p0,05). Dari hasil penelitian ini, dapat disimpulkan bahwa padat penebaran yang dapat memberikan hasil maksimum adalah 50 ekor/m2.Kata kunci: padat penebaran, pertumbuhan, kelangsungan hidup, lobster capit merah, Cherax quadricarinatus

Abstract

Probiotic bacteria has been widely used as biocontrol agents in tiger shrimp hatcheries.  Vibrio SKT-b is one of the probiotic bacteria candidates that could suppressed the growth of pathogenic bacteria Vibrio harveyi and could increase survival rate of tiger shrimp larva. This experiment was carried out to study the effects of probiotic bacteria SKT-b Vibrio addition at different doses on survival and growth rate of tiger shrimp larva.  Experiment was conducted with five treatments and three replications, consisted of SKT-b Vibrio probiotic bacteria addition at the doses of 103 CFU/ml, 104 CFU/ml, 105 CFU/ml, and 106 CFU/ml and control (0 CFU/ml).  Results showed that optimum dose of probiotic bacteria for tiger shrimp was 104 CFU/ml with a survival rate of 94.67%. However, the addition of probiotic bacteria at this particular dose did not significantly increase shrimp growth rate as compared with control. Key words: Probiotic bacteria, SKT-b Vibrio, doses, tiger shrimp larva   ABSTRAK Bakteri probiotik telah banyak digunakan sebagai agen biokontrol dalam pembenihan udang windu.  Vibrio SKT-b merupakan salah satu jenis bakteri kandidat probiotik yang telah diuji dapat menekan pertumbuhan bakteri patogen Vibrio harveyi dan dapat meningkatkan kelangsungan hidup larva udang windu.  Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian bakteri probiotik Vibrio SKT-b dengan dosis yang berbeda terhadap kelangsungan hidup dan pertumbuhan larva udang windu. Penelitian ini dilakukan dalam 5 perlakuan dengan masing-masing 3 ulangan, yaitu penambahan bakteri probiotik Vibrio SKT-b dengan dosis 103 CFU/ml, 104 CFU/ml, 105 CFU/ml, dan 106 CFU/ml dan kontrol (0 CFU/ml).  Hasil penelitian menunjukkan bahwa dosis optimal untuk larva udang windu adalah 104 CFU/ml dengan nilai kelangsungan hidup 94,67%. Namun, pemberian bakteri probiotik tersebut belum menghasilkan pertumbuhan yang berbeda nyata dengan kontrol. Kata kunci: Bakteri probiotik, Vibrio SKT-b, dosis, larva udang windu

Abstract

Motile aeromonad septicaemia (MAS) is caused by the bacterium Aeromonas hydrophila. The use of plant (natural materials) such as meniran and garlic can be as an alternative way to inhibit the activity of A. hydrophila by their active substances which have potency as an antibacterial and immunostimulant. The aim of this study was to analyze the effectiveness of using a mixture of meniran and garlic in feed to control of A. hydrophila in catfish. There were split in two kind of doses namely, preventive (garlic:meniran=5 ppt:20 ppt) and curative (garlic:meniran=10 ppt:40 ppt). The preventive treatment was given for two weeks before challenging test. The curative treatment was performed on 2th-8th day after challenging test.  Challenging test was carried out by intramuscularly injecting of 0,1 mL A. hydrophila (108cfu/ml)  into the fish. The results indicated that preventive treatment with a mixture of extracts 5 ppt meniran and 20 ppt garlic was more effective in preventing infection of A. hydrophila than curative treatment.  Keywords: Phyllanthus niruri, Allium sativum, catfish, Aeromonas hydrophila.  ABSTRAK  Penyakit MAS (motile aeromonad septicaemia) disebabkan oleh bakteri Aeromonas hydrophila. Bahan alami seperti meniran dan bawang putih dapat digunakan sebagai alternatif untuk menghambat aktivitas bakteri ini.  Zat aktif yang dimiliki bahan ini berpotensi sebagai antibakteri dan immunostimulan. Tujuan dari penelitian ini adalah untuk menganalisis efektivitas penggunaan campuran meniran dan bawang putih dalam pakan untuk pengendalian penyakit MAS pada ikan lele. Perlakuan dibagi menjadi dua dosis, yaitu pencegahan (bawang putih:meniran=5 ppt:20 ppt) dan pengobatan (bawang putih:meniran=10 ppt:40 ppt). Perlakuan pencegahan diberikan selama seminggu sebelum uji tantang. Perlakuan pengobatan dilakukan pada hari ke 2 hingga hari ke-8 setelah uji tantang. Uji tantang dilakukan dengan menyuntikkan secara intramuskuler 0,1 ml A. hydrophila (108cfu/ml) ke ikan lele. Hasil penelitian menunjukkan bahwa perlakuan pencegahan dengan campuran ekstrak meniran 5 ppt dan bawang putih 20 ppt efektif dalam mencegah infeksi A. hydrophila dibandingkan dengan perlakuan pengobatan dengan campuran ekstrak meniran 10 ppt dan 40 ppt bawang putih.   Kata kunci: meniran, bawang putih, lele, Aeromonas hydrophila.

Abstract

This research was purposed to determine the effective dose of meniran and garlic flour in feed by repelleting method for preventive and curative efficacy to the infection of Aeromonas hydrophila in catfish. The treatment tested were eight different dosages of garlic and meniran meals (2:1). Those four dosage used to preventive treatment namely A (0.1%), B (1.1%), C (2.1%), and D (3.1%), the other four used to curative treatment namely E (0.2%), F (2.2%), G (4.2%), and H (6.2%). The preventive treatments were given for two weeks before challenging test. The curative treatment was performed on 2nd–8th day after challenging test. Challenging test was carried out by intramuscularly injecting of 0.1 mL A. hydrophila (108cfu/mL) into the fish. Parameters observed were feeding response, growth rate, survival rate, clinical symptoms, wound healing, and organs morphology. The result showed that the survival rate of preventive treatment was 40±40% for A and B, 60±20% for C, and 20±20% for D. The survival rate of curative treatment was 33.33±11.55% for E, 46.67±11.55% for F, 33.33±11.55% for G, and 26.67±11.55% for H. The results indicated that the preventive treatment was more effective than the curative treatment, presumably because the immunostimulatory activities on preventive was better than curative. Abstract

The use of antibiotics for controlling of luminous vibriosis caused by Vibrio harveyi is restricted now, because it induces antibiotic-resistant bacteria and leave residue in shrimp’s body. An alternative solution that can be done to treat the disease is by using applications of probiotic, prebiotic, and synbiotic. The aim of this research was to examine the effect of probiotic, prebiotic, and synbiotic on the survival rate and growth of Pacific white shrimp against V. harveyi infection. Feed as a treatment was supplemented with probiotic 1%, prebiotic 2%, and probiotic 1%+prebiotic 2% (synbiotic). Shrimps feed without supplementation of probiotic and prebiotic was used as a control treatment. The shrimps were maintainedin the aquarium (60×30×35 cm3) with a density of 40 shrimps/40 L and an average weight of 0.4±0.1 g. After 30 days of feeding treatment, the shrimp was challenged by immersion method with V. harveyi solution containing 106 CFU/mL. The results showed that before challenge, synbiotic feed treated shrimp has a growth rate (5.89%), feed conversionrate (1.21), and a high survival rate (80%). After challenge, survival rate (83.33%) of shrimp fed diet supplemented with synbiotic was higher than prebiotic (51.67%) and positive control (31.67%). Abstract

Motile Aeromonad Septicaemia (MAS) caused by Aeromonas hydrophila induced serious epidemics of disease in catfish Clarias sp. The purposed of this research was to determine optimum durations of feed application of mixed powders Phyllanthus niruri and Allium sativum to prevent MAS. Experimental design that used was Complete Random Design which consist of three treatments and three replications. This research tested on three different of durations of feed application of P. niruri and A. sativum, namely A (powder of P. niruri and A. sativum feed application during 21 days and infected with A. hydrophila 0,1 mL IP), B (powder of P. niruri and A. sativum feed application during 14 days and injected IP with A. hydrophila 0,1 mL), C (powder of P. niruri and A. sativum feed application during seven days and injected IP with A. hydrophila 0,1 mL), and control, namely K- (without P. niruri and A. sativum feed application and injected IP with PBS 0,1 mL) and K+ (without P. niruri and A. sativum powder application and injected IP with A. hydrophila 0,1 mL). The treatments was given for 21 days before challenging test, at 22th day test in vivo carried out by injecting A. hydrophila (108 CFU/mL) into the fish by intramuscular and observed for 10 days. The highest value of survival rate that consist in treatment K- was 100±0.00%, treatment A was 93.3±11.55%, and treatment B was 73.33±30.55%. While treatment K+ and C have the same survival rate that is, 26.67±11.55%. The application P. niruri and A. sativum powder during 21 days was optimum for preventing MAS in catfish.Keywords: Phyllanthus niruri, Allium sativum, Aeromonas hydrophila, Clarias sp.

Abstract

ABSTRACT This study aimed to determine the dose of garlic and shatterstone herb mixed into the feed in 11-day old catfish juvenile (body length 1.53±0.26 cm, body weight of 40±16 mg). The study was divided into two steps, the first step was dose determination and the second step was the dose testing. The treatment was carried out for 21 days, then the challenge test was conducted by immersion method for 60 minutes in A. hydrophila density of 104 cfu/mL. Parameters of observation were survival, feed intake, relative growth rate, morphology of the liver, and water quality. The result showed that garlic at 25 g/kg and shatterstone herb at 5 g/kg provided higher fish viability (81.11±3.85%) compared with the control (23.00±5.77%). Feed intake and relative growth rate were not different among the treatments. Whereas the liver condition in positive control was paler than other treatments. Thus, prevention of A. hydrophila infection on catfish juvenile was effectively achieved by feeding with diet supplemented by garlic 25 g/kg and shatterstone herb 5 g/kg. Keywords: garlic, shatterstone herb, Aeromonas hydrophila, juvenile, catfish  ABSTRAK  Penelitian ini bertujuan untuk menentukan dosis bawang putih dan meniran yang dicampur ke pakan dalam bentuk tepung pada benih lele berumur 11 hari (berukuran panjang 1,53±0,26 cm dan bobot 40±16 mg). Penelitian ini terbagi menjadi dua tahap, pertama adalah tahap penentuan dosis (dosis 20 ppt bawang putih+5 ppt meniran dan 25 ppt bawang putih+5 ppt meniran) dan kedua adalah tahap pengujian dosis. Perlakuan diberikan selama 21 hari, kemudian uji tantang dilakukan dengan metode perendaman selama 60 menit dengan kepadatan bakteri A. hydrophila 104 cfu/mL. Parameter yang diamati adalah kelangsungan hidup, jumlah konsumsi pakan, pertumbuhan relatif, morfologi organ hati, dan kualitas air. Hasil penelitian membuktikan bahwa perlakuan bawang putih 25 g/kg dan meniran 5 g/kg memberikan kelangsungan hidup benih lele sebesar 81,11±3,85%, sedangkan kontrol positif 23,00±5,77%. Jumlah konsumsi pakan dan pertumbuhan relatif tidak berbeda antarperlakuan, sedangkan organ hati berwarna pucat pada kontrol positif. Pencegahan infeksi bakteri A. hydrophila dengan pakan campuran bawang putih 25 g/kg dan meniran 5 g/kg efektif dilakukan pada benih ikan lele. Kata kunci: bawang putih, meniran, Aeromonas hydrophila, benih, ikan lele

Abstract

 ABSTRACTProbiotic bacteria SKT-b Vibrio that belong to heterotrophic bacteria have been tested can suppress the growth of pathogenic Vibrio harveyi in tiger shrimp larvae. One of the most important energy sources of heterotrophic bacteria is organic carbon such as sucrose and glucose found in molasses. The objective of this experiment was to study the effect of molasses addition on the survival and growth rate of tiger shrimp Penaeus monodon larvae treated with SKT-b Vibrio probiotic bacteria. This experiment was done both in vitro and in vivo. In vitro tests performed by growing bacteria in media containing molasses, fish meal and premix. Whereas in vivo tests performed with the addition of molasses to shrimp larva culture media inoculated with SKT-b Vibrio bacteria which included five treatments: the control treatment without bacteria and molasses addition (K), the addition of bacteria without molasses (B), the addition of molasses 1 ppm and bacteria (MB1), the addition of molasses 3 ppm and bacteria (MB3), and the addition of molasses 5 ppm and bacteria (MB5). The result of in vitro tests showed the SKT-b Vibrio bacteria can grow on molasses media with a population reached 0,41 × 108 CFU/ml. In vivo test results showed that treatment MB3 gave the highest survival (93.3%) and growth weight (35.94%), but were not significantly different from control result in the survival and growth weight that respectively were 83.3% and 30.38%.Keywords: probiotic bacteria, SKT-b Vibrio, molasses, tiger shrimp larvae ABSTRAKBakteri probiotik Vibrio SKT-b yang tergolong bakteri heterotrof telah diuji dapat menekan pertumbuhan Vibrio harveyi yang bersifat patogen pada larva udang windu. Salah satu sumber energi yang paling penting bagi bakteri heterotrof adalah karbon organik seperti sukrosa dan glukosa yang terdapat pada molase. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh penambahan molase terhadap kelangsungan hidup dan pertumbuhan larva udang windu Penaeus monodon Fab. yang diberi bakteri probiotik Vibrio SKT-b. Penelitian dilakukan secara in vitro dan in vivo. In vitro yaitu menumbuhkan bakteri pada media yang mengandung molase, tepung ikan dan premix. In vivo yaitu penambahan molase pada media pemeliharaan udang yang diberi bakteri probiotik Vibrio SKT-b, yang meliputi lima perlakuan yakni: perlakuan kontrol tanpa penambahan bakteri maupun molase (K), penambahan bakteri tanpa molase (B), penambahan molase 1 ppm dan bakteri (MB1), penambahan molase 3 ppm dan bakteri (MB3), penambahan molase 5 ppm dan bakteri (MB5). Hasil uji in vitro menunjukkan bakteri Vibrio SKT-b dapat tumbuh pada media molase dengan populasi mencapai 0,41×108 CFU/mL. Hasil uji in vivo menunjukkan perlakuan MB3 menghasilkan kelangsungan hidup (93,3%) dan pertumbuhan bobot (35,94%) tertinggi, namun tidak berbeda nyata dengan kontrol yang menghasilkan kelangsungan hidup dan pertumbuhan bobot berturut-turut 83,3% dan 30,38%.Kata kunci: bakteri probiotik, Vibrio SKT-b, molase, larva udang windu

Abstract

ABSTRACTWhite spot syndrome virus (WSSV) is the most dangerous shrimp pathogen and give a very bad impact to the shrimp industry in many countries, including Indonesia. Furthermore, the vaccine usage is less effective since the local WSSV suspected to have some mutation and have differences in sequence from the one that used for recombinant vaccine production so far. Therefore, it is necessary to produce recombinant vaccine for immunization of local shrimp according to WSSV isolates from Indonesia. Viral protein 19 (VP19) is an important WSSV envelope proteins because of its involvement in the systemic infection of shrimp, so it can be developed as a recombinant vaccine. The objectives of this study are to obtain VP19-encoding gene of WSSV from Situbondo, Indonesia. This research conducted through several stages, DNA genome isolation of Penaeus monodon infected with WSSV, amplification of VP19-encoding gene, isolation of VP19-encoding gene, ligation of VP19-encoding gene into pGEM-T Easy vector, transformation to Escherichia coli, recombinant vector screening, vector isolation using mini preparation technique, vector verification, and sequencing. The result of cloning shows that the size of VP19-encoding gene from Situbondo isolates is similar to VP19-encoding gene from the abroad one, which is 366 bp. However, the sequence result shows that there are some differences on bases and amino acid. However, the result of sequencing shows there are difference of five base and three amino acid arranged VP19 sequence the Situbondo isolate with isolate from several countries. The similiarity of the Situbondo isolate sequence with the other isolate (from Indonesian and other countries) was only 98%. This difference indicates the importance of making a vaccine from a local WSSV genes especially Situbondo isolate.Keywords: white spot syndrome virus, VP19, Penaeus monodonABSTRAKWhite spot syndrome virus (WSSV) merupakan patogen yang paling serius menyerang udang windu dan telah menghancurkan industri udang windu di berbagai negara, termasuk di Indonesia. Penggunaan vaksin WSSV yang didesain berdasarkan isolat WSSV luar negeri kurang efektif karena diduga WSSV yang menyerang udang Indonesia telah mengalami mutasi dan memiliki perbedaan sekuen. Untuk itu, penggunaan isolat WSSV asal Indonesia, khususnya Situbondo sebagai cetakan dalam memproduksi vaksin rekombinan untuk imunisasi udang di Indonesia sangat perlu dilakukan. Viral protein-19 (VP19) merupakan protein pembungkus WSSV yang penting karena keterlibatannya dalam infeksi sistemik pada udang, sehingga dapat dikembangkan sebagai vaksin rekombinan. Tujuan penelitian ini adalah untuk mendapatkan gen penyandi protein permukaan VP19 virus WSSV isolat Situbondo. Tahapan penelitian yang dilakukan pada penelitian ini adalah isolasi DNA genom udang windu yang terinfeksi WSSV, amplifikasi gen penyandi protein VP19, isolasi gen penyandi protein VP19, ligasi gen penyandi protein VP19 ke dalam plasmid pGEM-T Easy, transformasi plasmid, skrining plasmid rekombinan, isolasi plasmid dengan teknik mini preparation, verifikasi plasmid dan pengurutan. Hasil pengklonan menunjukkan bahwa ukuran gen penyandi protein VP19 isolat Situbondo sama dengan ukuran gen penyandi protein VP19 isolat asing yaitu 366 pb. Namun hasil pengurutan menunjukkan adanya perbedaan lima basa dan tiga asam amino penyusun sekuen VP19 isolat Situbondo dengan isolat dari beberapa negara lain. Kemiripan sekuen isolat Situbondo dengan isolat lain (dari Indonesia sendiri maupun dari negara-negara lain) hanya sebesar 98%. Abstract

ABSTRACT This study aimed to analyze the genetic diversity of bacteria associated in bioflocs using 16S-rRNA polymerase chain reaction (PCR) with ARDRA technique. A total of 38 dominant bacterial isolates was obtained from bioflocs samples and of these isolates, 16S-rRNA gene was then isolated and amplified using PCR. The 16S-rRNA gene of the isolates was then cut using HaeIII (5’-GG↓CC) and HhaI (5’-GCG↓C) restriction enzymes resulting an ARDRA pattern which was further used as the binary data for the construction of phylogenetics tree that was used to estimate the group of bacteria. The result with HaeIII cut restriction enzyme from biofloc-associated bacteria gave 11 ARDRA patterns, while with the restriction enzyme HhaI gave eight ARDRA patterns. Phylogenetics of bacterial populations from biofloc-based cultivation system water consisted of at least 13 different bacterial species. Result of sequencing from two gene sample 16S-rRNA were identified as Microbacterium foliorumand and Pseudomonas putida. Keywords: bacterial diversity, ARDRA, biofloc, phylogeny  ABSTRAK Penelitian ini bertujuan untuk menganalisis keragaman genetika bakteri bioflok menggunakan metode polymerase chain reaction (PCR) 16S-rRNA dengan teknik ARDRA. Sebanyak 38 isolat bakteri dominan yang diperoleh diamplifikasi gen 16S-rRNAnya dengan PCR, kemudian dipotong dengan enzim restriksi HaeIII (5’-GG↓CC) dan HhaI (5’-GCG↓C). Pola ARDRA ini dijadikan data biner sebagai input untuk konstruksi pohon filogenetika yang dapat digunakan untuk memerkirakan jenis bakteri yang ada. Gen 16S-rRNA hasil PCR setelah dipotong dengan enzim restriksi HaeIII didapatkan 11 pola ARDRA, sedangkan dengan enzim restriksi HhaI menghasilkan delapan pola ARDRA. Berdasarkan pohon filogenetika, diketahui populasi bakteri pada air sistem budidaya bioflok sedikitnya terdiri atas 13 jenis bakteri. Berdasarkan sekuensing dari dua sampel gen 16S-rRNA teridentifikasi jenis bakteri Microbacterium foliorum dan Pseudomonas putida. Kata kunci: keragaman bakteri, ARDRA, bioflok, filogenetika
