Abstract

ABSTRAKPenelitian ini bertujuan mengetahui pengaruh pemberian vitamin E terhadap kadar SOD serum tikus putih (Rattus norvegicus) diabetes melitus. Penelitian ini menggunakan tikus putih sebanyak 25 ekor dengan kriteria: sehat, bobot badan 150 – 200 gram; umur ± 3 – 4 bulan; dan jenis kelamin jantan. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) pola searah dengan 5 kelompok perlakuan. Masing-masing kelompok perlakuan terdiri atas lima ekor tikus. Kelompok tikus non-diabetes (KN), kelompok tikus diabetes induksi aloksan (KP), kelompok perlakuan yaitu tikus diabetes induksi aloksan dan diberi vitamin E dosis 50 IU/kgbb/hari (P1), 100 IU/kgbb/hari (P2), dan 150 IU/kgbb/hari (P3). Perlakuan vitamin E diberikan selama 28 hari. Pada hari ke-29 setelah perlakuan, dilakukan koleksi serum untuk pengukuran kadar SOD serum dengan spektrofotometer. Data yang diperoleh dianalisis dengan ANOVA satu arah dan uji lanjut Duncan. Hasil penelitian menunjukkan kadar SOD serum adalah 58,00 ± 7,48 U/ml, 36,50 ± 4,56  U/ml (KP), 42,55 ± 5,89 U/ml (P1), 50,44 ± 5,32 U/ml (P2), dan 55,20 ± 4,84 U/ml (P3). Hasil penelitian menunjukkan bahwa vitamin E dapat meningkatan kadar SOD serum secara signifikan (P0,05). Pemberian  vitamin E dengan dosis 150 IU/kg BB/hari menghasilkan peningkatan kadar SOD serum terbaik pada tikus putih diabetes melitus. Kesimpulan dari penelitian ini adalah pemberian vitamin E dapat meningkatkan kadar SOD serum tikus putih diabetes melitus.ABSTRACT            This study aims to know the effect of vitamin E administration to SOD serum level in diabetes mellitus induced white rats (Rattus norvegicus). This study used 25 white rat in criteria: healthy; body weight 150 – 200 gram; age ± 3 – 4 month; and male sex. The research design was used completely randomized design (CRD) with 4 unidirectional pattern groups treatments. Each group consisted of five rat. Group of non-diabetic rats (KN), group of alloxan-induced diabetic rats (KP), group of alloxan-induced diabetic rats which given with vitamin E 50 IU/kgBW/day (P1), 100 IU/kgBW/day (P2), and 150 IU/kgBW/day (P3). Treatment of vitamin E was given in 28 days. On the 29th day after treatment, serum collection was performed to quantify SOD serum level by spectrophotometers. The data obtained were analyzed by one-way ANOVA and continued with Duncan test. The result showed SOD serum level was 58.00 ± 7.48 U/ml, 36.50 ± 4.56 U/ml (KP), 42.55 ± 5.89 U/ml (P1), 50.44 ± 5.32 U/ml (P2), and 55.20 ± 4.84 U/ml (P3). The result showed that therapy of vitamin E could increase SOD serum level significantly (P0.05). Treatment dose of 150 IU/kgBW/day was the best increasing of SOD serum level in diabetes mellitus induced white rat. The conclusion of research showed that giving of vitamin E can increasing the SOD serum level in diabetes mellitus induced white rat.

Abstract

ABSTRAK             Penelitian ini bertujuan untuk mengetahui pengaruh pemberian campuran ampas tahu, jagung, dedak dan bungkil kedelai fermentasi dalam ransum komersial terhadap kualitas spermatozoa ayam kampung. Penelitian ini merupakan penelitian eksperimen menggunakan Rancangan Acak Kelompok (RAK) yang terdiri dari 4 perlakuan dengan 4 kali ulangan, kelompok perlakuan 1 (P0): ayam diberi 100% ransum komersial, perlakuan 2 (P1): ayam diberi 90% ransum komersial + 10% campuran ampas tahu, jagung, dedak dan bungkil kedelai  fermentasi, kelompok 3 (P2): ayam diberi 80% ransum komersial + 20% campuran ampas tahu, jagung, dedak dan bungkil kedelai  fermentasi dan kelompok 4 (P3) ayam diberi 70% ransum komersial + 30% campuran ampas tahu, jagung, dedak dan bungkil kedelai  fermentasi. Pemberian pakan dilakukan 2 kali sehari (pagi dan sore) dengan takaran 100gr/ekor/hari selama 30 hari. Pada hari ke 31 sampel semen dikoleksi dengan metode masase. Data yang diperoleh dianalisis dengan menggunakan analisis varian (ANAVA) yang dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan pemberian campuran ampas tahu, jagung, dedak dan bungkil kedelai yang difermentasi dalam ransum komersial berpengaruh secara nyata (P0,05) terhadap peningkatan konsentrasi spermatozoa dan penurunan persentase spermatozoa abnormal, namun tidak berpengaruh secara nyata (P0,05) terhadap persentase motilitas dan spermatozoa hidup ayam kampung. Rata-rata konsentrasi dan persentase spermatozoa abnormal pada kelompok P3 berbeda secara nyata (P0,05) dibandingkan dengan P0, namun tidak berbeda secara nyata (P0,05) dengan  P1 dan P2. Pemberian campuran ampas tahu, jagung, dedak dan bungkil kedelai fermentasi dalam ransum komersial dapat meningkatkan konsentrasi dan penurunan persentase spermatozoa abnormal, namun tidak berpengaruh terhadap persentase motilitas dan spermatozoa hidup ayam kampung.Kata kunci: Ayam Kampung, Kualitas Spermatozoa, Pakan Fermentasi                                                           ABSTRACTThis study aimed to determined the effect of tofu dregs, corn, bran and fermented soybean meal mixture addition in commercial rations towards the quality of Gallus domestica spermatozoa. This study used completely randomized design (CRD) consisting of 4 treatment group by four times repetition. First group (P0), chickens were fed with 100 % commercial rations, second group ( P1 ) were fed with 90 % commercial rations + 10 % mixture of tofu dregs, corn, bran and fermented soybean meal (TCBF), third group ( P2 ) were fed with 80 % commercial rations + 20 % of TCBF mixture, and fourth group ( P3 ) were fed with 70% commercial rations + 30 % of TCBF mixture. Feeding were twice a day (morning and evening) by 100g/each/day for 30 days. Semen sample were collected in 30th days by massage method. Acquired data were analyzed in Analysis of Variance (ANOVA) then proceed with Duncan test. The result showed that the mixture addition of tofu dregs, corn ,bran and fermented soybean meal in commercial rations significantly affect (P0,05) to spermatozoa concentration and decreased of abnormal spermatozoa, but not significantly affect (P0,05) to motility percentage and life spermatozoa in Gallus domesticus. Mean of spermatozoa abnormality and concentration in P3 was significantly different to P0, but not significantly different (P0,05) when compared to P1 and P2. Mixture addition of tofu dregs, corn, bran and fermented soybean meal in commercial rations was capable to increased spermatozoa concentration and reduced percentage of spermatozoa abnormality, but was not affect motility and life spermatozoa percentage of Gallus domesticus.    Keywords: Gallus domestica, Quality of spermatozoa, Fermented feed

Abstract

ABSTRAK Penelitian ini bertujuan mengetahui pengaruh pemberian formalin melalui intraperitoneal terhadap kadar hemoglobin dan jumlah eritrosit pada tikus putih (Rattus norvegicus) strain Wistar. Hewan yang digunakan dalam penilitian ini adalah 24 ekor tikus putih jantan. Rancangan yang digunakan adalah rancangan acak lengkap (RAL) pola searah dengan 4 kelompok perlakuan. Masing-masing kelompok perlakuan terdiri atas enam ekor tikus. Kelompok kontrol tikus hanya diberi pakan dan air minum tanpa perlakuan apapun, kelompok 1 tikus diinjeksi formalin dengan dosis 1 mg/Kg bb, kelompok 2 diinjeksi formalin dengan dosis 2,5 mg/Kg bb, kelompok 3 diinjeksi formalin dengan dosis 5 mg/Kg bb. Masing-masing kelompok diberi perlakuan selama 14 hari berturut-turut secara intraperitoneal. Selama penelitian tikus diberi pakan dan air minum secara adlibitum. Pada hari ke 15, sampel darah diambil melalui sinus orbitalis menggunakan pipet kapiler. Hasil analisis statistik menunjukkan rata-rata (±SD) kadar hemoglobin (g/dl) K0 12,00±0,72, K1 9,63±1,67, K2 9,83±1,30, dan K3 9,76±1,41. Rata-rata (±SD) jumlah eritrosit (106/mm3) K0 7,06±0,78, K1 5,31±1,46, K2  5,35±0,97, dan K3 4,98±0,59. Hasil uji ANOVA menunjukkan kelompok K0 berpengaruh nyata (P0,05) terhadap kelompok K1, K2, dan K3, sementara kelompok K1, K2, dan K3 tidak berpengaruh nyata (P0,05) antara kelompok perlakuan. Disimpulkan bahwa pemberian formalin secara intraperitoneal dapat menurunkan kadar hemoglobin dan jumlah eritrosit tikus putih strain Wistar.Kata kunci: formalin, kadar hemoglobin, jumlah eritrosit, Rattus norvegicusABSTRACTThe study aims to determine the effect of formalin administration via intraperitoneal on levels of haemoglobin and the number of erythrocyte in rats (Rattus norvegicus) Wistar strain. Animals used in this study were 24 male rat. The research design was used completely randomized design (CRD) with 4 unidirectional pattern groups treatments. Each group treatment consisted of six rat. Group control rat was given only fed and drinking without any treatment, group 1 rat injected formalin with dosage of  1 mg/Kg bw, group 2 rat  injected formalin with dosage of 2,5 mg/Kg bw, and group 3 rat injected formalin with dosage of 5 mg/Kg bw. Each group was given the specific treatment for 14 consecutive days intraperitoneally. During the study, rat were fed and drinking water in a adlibtum. On day 15, blood sample was taken through orbital sinus using a pipette capillary. The statistical analysis that showed mean (±SD) levels of haemoglobin (g/dl) K0 12,00±0,72, K1 9,63±1,67, K2 9,83±1,30, and K3 9,76±1,4. Mean (±SD) number of erythrocyte (106/mm3) K0 7,06±0,78, K1 5,31±1,46, K2  5,35±0,97, and K3 4,98±0,59. The results of ANOVA showed significant group K0 (P0,05) against groups K1, K2, and K3, while groups K1, K2, and K3 no significan effect (P0,05) in between group. It was concluded that the administration of formalin intraperitoneal can lower haemoglobin levels and the number of erythrocyte rats Wistar strain.Keywords:  formalin, haemoglobin levels, number of erythrocyte, Rattus norvegicus

Abstract

Penelitian ini bertujuan mengetahui pengaruh penambahan ekstrak kulit buah naga merah dalam media sitrat kuning telur terhadap daya tahan hidup spermatozoa sapi aceh setelah pendinginan. Semen ditampung dari 1 ekor pejantan umur 3 tahun sehat, menggunakan vagina buatan. Semen yang berkualitas baik dibagi ke dalam 5 kelompok perlakuan penambahan ekstrak kulit buah naga merah; 0,00 g/100 ml (P0); 0,2 g/100 ml (P1); 0,4 g/100 ml (P2); 0,6 g/100 ml (P3); dan 0,8 g/100 ml (P4) dalam media sitrat kuning telur dan disimpan dalam suhu 4. Pengamatan daya tahan hidup spermatozoa dilakukan setiap hari selama 5 hari. Data yang diperoleh dianalisa dengan analysis of varian(ANOVA) pola satu arah yang dilanjutkan dengan uji berganda Duncan. Hasil pengamatan daya tahan hidup spermatozoa setelah pendinginan pada perlakuan P0; P1; P2; P3; dan P4 secara berturut-turut 3,8; 4,6; 3,6; 1,0; dan 0,0 hari. Hasil penelitian ini menunjukkan bahwa penambahan ekstrak kulit buah naga merah dalam media sitrat kuning telur berpengaruh secara nyata (P0,05) terhadap daya tahan hidup spermatozoa. Penambahan ekstrak kulit buah naga merah 0,2 g/100 ml dalam media sitrat kuning telur menghasilkan daya tahan hidup spermatozoa sapi aceh lebih baik selama penyimpanan pada suhu 4. (This study aims to determine the effect of peels red dragon fruit extract addition into citrate-yolk medium on viability of aceh cattle sperm after cooling. Semen was collected from 1 healthy male aged 3 years, using artificial vagina. Good quality semen was divided into 5 groups; addition of peels red dragon fruit extract 0.0 g/100 ml (P0); 0.2 g/100 ml (P1); 0.4 g/100 ml (P2); 0.6 g/100 ml (P3); and 0.8 g/100 ml (P4) into citrate-yolk medium, and stored in a temperature of 4. Observation of sperm viability were done every day up to day 5. The data obtained was analyzed with the one-way analysis of variance (ANOVA), followed by multiple test Duncan. Results show sperm viability after cooling in the groups P0; P1; P2; P3; and P4 respectively was 3.8 days, 3.6 days, 4.6 days, 1.0 day, and 0.0 day. The results show that the addition of peels red dragon fruit extract in citrate-yolk medium affects significantly (P0.05) sperm viability. The addition 0.2 g/100 ml peels red dragon fruit extract into citrate-yolk medium during storage in a temperature of 4 results greater sperm viability.)

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui pengaruh vitamin E terhadap kadar MDA serum tikus putih (Rattus novergicus) diabetes melitus. Penelitian ini menggunakan rancangan acak lengkap (RAL). Sebanyak 25 ekor tikus dibagi secara acak menjadi 5 kelompok: kelompok kontrol negatif yaitu tikus non-diabetes (KN), kelompok kontrol positif yaitu tikus diabetes tanpa diberi vitamin E (KP), tikus diabetes yang diberi vitamin E dosis 50 IU/kgbb/hr (P1),100 IU/kgbb/hr (P2), dan 150 IU/kgbb/hr (P3) selama 28 hari. Selama penelitian tikus diberi pakan dan air minum secara ad libitum. Pada hari ke-29, dilakukan pengkoleksian serum darah untuk pemeriksaankadar MDA secara spektofotometri. Data yang diperoleh dianalisis menggunakan analisis varian (ANAVA) dan dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan rata-rata (±SD) kadar  MDA serum adalah 13,44 ± 3,15 µmol/l (KN), 22,18 ± 6,44 µmol/l (KP),19,01 ± 5,25 µmol/l (P1), 14,86 ± 4,11 µmol/l (P2), dan 12,25 ± 2,45 µmol/l (P3).Hasil penelitian menunjukkan bahwa pemberian vitamin E dapat menurunkan kadar MDA serum. Pemberian vitamin E dosis 150 IU/kgbb/hari lebih baik dibandingkan dengan dosis 100 IU/kgbb/hari dan 50 IU/kgbb/hari dalam menurunkan kadar MDA serum. Kesimpulan dari penelitian ini adalah pemberian vitamin E dapat menurunkan kadar MDA serum tikus putih diabetes melitus.ABSTRACT             The aim of this study were to determine the effect of vitamin E on serum MDA level in white rat (Rattus novergicus) diabetes mellitus. During the study, rat were fed with food and water in ad libitum. On the 29th day,the collection of blood serum was done to check the MDA level using the spectophotometer.Acquired data were analysed  by using Analysis of Variance (ANOVA) then proceed with Duncan test. The result showed the mean (±SD)of MDA serum level was 13,44 ± 3,15 µmol/l (KN), 22,18 ± 6,44 µmol/l (KP),19,01 ± 5,25 µmol/l (P1), 14,86 ± 4,11 µmol/l (P2), dan 12,25 ± 2,45 µmol/l (P3).The results of the study showed that fedding of vitamin E could reduce serum MDA levels.The fedding of vitamin E150 IU/kgbw/day was better than 100 IU/kgbw/day and 50 IU/kgbw/day in lowering serum MDA levels.
