Abstract

This study aims to investigate the effect of Biotin vitamin on the growth of in vitro shoot explants from Barangan and Cavendish banana varieties and to determine the optimal Biotin concentration. The research employed tissue culture techniques using Murashige and Skoog (MS) medium and a Completely Randomized Design (CRD) with a total of 144 culture bottles. The results indicate that the addition of Biotin significantly influences the growth of both varieties. The Cavendish variety exhibited a better response than Barangan, with a Biotin concentration of 2.5 ppm yielding the best results in terms of shoot number, shoot length, and faster shoot formation time. Meanwhile, for the Barangan variety, a concentration of 0.5 ppm accelerated shoot formation, while a concentration of 2 ppm resulted in the highest shoot number and length. The conclusion of this study suggests that higher Biotin concentrations can enhance the growth of shoot explants, particularly in the Cavendish variety. These findings may serve as a basis for the more efficient development of banana seedlings through in vitro methods. Further research is necessary to evaluate the efficiency during the acclimatization stage in the field.

Abstract

Soybeans are a plant that has great opportunities and promising prospects to be cultivated, but there are often problems in this cultivation such as the problem of seed setback. The gradual process of degradation of seed quality and vitality, known as seed deterioration, cannot return to its original state or be restored. Soybean seeds that have expired so far are no longer used, because their short shelf life makes them only usable for consumption. Using invigoration techniques is one method to control seed setback. Matriconditioning is a treatment that utilizes moist solid media, such as charcoal, husks, and sawdust. This research aims to investigate the effect of husk charcoal on the viability and vigor of soybean seeds, as well as its effects on soybean growth and yield (Glycine max (L). Merril). This research lasted about 4 months, starting in November 2022, , and was carried out at the Seed Technology Laboratory and Teaching and Research Farm, Faculty of Agriculture, University of Jambi and was carried out in two stages. This research was carried out simultaneously, namely Phase I of the research was carried out in the laboratory, while Phase II was carried out in the field. The results of the study showed that invigoration treatment with matriconditioning had a significant effect on germination power, maximum growth potential, normal germination dry weight, and growth speed. The treatment of planting media has a real effect on the weight of planting seeds and the number of pods per plant. Matriconditioning treatment had no noticeable effect on plant height, dry weight of the plant, root nodules, seed weight per plant, and number of pods per plant. Matriconditioning treatment has a real effect on the leaf area index.

Abstract

Peanuts are a legume commodity that ranks second after soybeans and have the potential to be a strategic commodity because demand is quite large every year, both for food, feed and industry. Ultisol soil is soil that is less fertile and unproductive for most plants. Organic matter can improve soil conditions and provide nutrients for plants. Biochar is a soil amendment material made from various crop residues that has potential and has a high C/N ratio or is difficult to decompose. The aim of this research is to determine the effect of rice husk biochar combined with chicken manure on improving the fertility of Ultisol, on peanut yield. , and determine the best dose of rice husk biochar combined with chicken manure for peanut yield on Ultisol soil. The research was carried out at the experimental field of the Agricultural Training Center Jl. Jambi – Palembang, KM 16 Pondok Meja Village, Mestong District, Muaro Jambi Regency, from May to August 2022. Research usingRandomized Block Design (RAK), with 9 treatments, namely chicken manure (P0B0 = Without biochar and chicken manure, P1B0 = 10 tons ha-1 chicken manure, P2B0 = 15 tons ha-1 chicken manure manure, P0B1 = 5 ton ha-1 biochar, P0B2 = 10 tons ha-1 biochar, P1B1 = 10 tons ha-1 chicken manure + 5 tons ha-1 biochar, P1B2 = 10 tons ha-1 chicken manure + 10 tons ha- 1 biochar, P2B1 = 15 tons ha-1 chicken manure + 5 tons ha-1 biochar, P2B2 = 15 tons ha-1 chicken manure + 10 tons ha-1 biochar) and 3 groups as replications, so that in the research There are 27 experimental plots. The observation variables in this research were plant height, number of leaves per plant, flowering age, number of pods per plant, pod weight per plant, dry pod weight per plot, weight of 100 seeds, plant yield per hectare, and plant P and K nutrient uptake. To determine the effect of treatment, a 5% level of analysis of variance (ANOVA) was carried out. The results of the research show that the application of rice husk biochar combined with chicken manure affects the fertility of ultisol, namely by increasing the soil pH to close to neutral, the elements of total N, total P, total K and available P so that it influences the variables of plant height, number of leaves, number of pods per plants, plant P and K uptake, dry weight of pods per plant, dry weight of pods per plot, weight of 100 seeds, and plant yield per hectare. Keywords: Peanuts, Ultisol, Biochar, Chicken Manure.
