Abstract

ABSTRAKPenelitian ini bertujuan untuk mengisolasi bakteri asam laktat (BAL) pada feses rusa sambar.  Sampel yang digunakan yaitu feses segar yang berasal dari 6 ekor rusa sambar yang ada di Taman Rusa Sibreh, Aceh Besar. Isolasi bakteri BAL dilakukan dengan menggunakan metode Carter (1976). Koloni BAL yang tumbuh diamati morfologi koloninya secara makroskopis dan mikroskopis. Selanjutnya dilakukan uji katalase dan pewarnaan spora dengan metode Schaeffer and Fulton. Data yang diperoleh dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa bakteri yang diisolasi termasuk kedalam kelompok bakteri Gram positif, katalase postif dan ada juga yang negatif, serta tidak berspora. Berdasarkan hasil penelitian dapat disimpulkan bahwa bakteri asam laktat (BAL) genus Lactobacillus dapat diisolasi pada feses rusa sambar di Taman Rusa Sibreh, Aceh Besar.Kata kunci: Rusa sambar, Bakteri Asam Laktat, LactobacillusABSTRACKThis study aim to isolate lactic acid bacteria (BAL) from sambar deer. The samples used in this study. Study were fresh feces from 6 sambar deers in Taman Rusa Sibreh, Aceh Besar. Isolation of BAL was done based on Carter method (1976). The BAL colonies were observed, macroscopically and microscopically. Then, katalase test and spore with Schaeffer and Fulton technique were done. The data were analyzed descriptively. The results showed that isolated bacteria belonged to Gram positive bacterial groups, catalase postif and there was also a negative, and not spore. Based on the results of this study it can be concluded that lactic acid bacteria (BAL) of Lactobacillus genus can be isolated from sambar deers feces.Keywords: Sambar deer, Lactic acid bacteria (BAL), Lactobacillus

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui daya hambat ekstrak biji buah pala (Myristicafragrans Houtt) terhadap pertumbuhan bakteri Escherichia coli. Biji pala yang digunakan dalam penelitian ini diperoleh dari pasar tradisional Darussalam, Banda Aceh. Proses ekstrasi biji buah pala menggunakan pelarut Etanol 96% dengan tingkat konsentrasi yang berbeda yaitu 1%, 10%,20%.  Menggunakan kertas cakram kosong (Blank disk) yang telah dicelupkan ke dalam ekstrak biji buah pala masing-masing pengulangan dan antibiotik ampisilin yang diletakkan pada media MHA. Pengujian daya hambat dilakukan dengan metode difusi Kirby-Bauer menggunakan kertas cakram pada media Mueller hinton agar (MHA) dengan 5 perlakuan dan 3 pengulangan. Media agar diinkubasi pada suhu 36-37ºC selama 24 jam. Hasil penelitian ini menunjukkan bahwa bahwa ektsrak  biji  buah  pala  memiliki  daya  hambat  yang  lemah  terhadap  pertumbuhan  bakteri Escherichia coli.  Ekstrak biji buah pala pada konsentrasi 1% memiliki daya hambat sebesar 9,73 mm, konsentrasi 10% sebesar 8,5 mm, konsentrasi 20% sebesar 8 mm dan kontrol positif sebesar20,83 mm.Kata kunci : antibakteri, ekstrak biji buah pala, Escherichia coli.. ABSTRACTThis study aims to determine the inhibitory power of nutmeg extract (Myristica fragransHoutt) on the growth of Escherichia coli bacteria. The nutmeg seed used in this study was obtained from the traditional market of Darussalam, Banda Aceh. Extraction process of nutmeg seed using96% ethanol solvent with different concentration level that is 1%, 10%, 20%. Using blank disc paper (Blank disk) which has been dipped into nutmeg extract of each repeating and ampicillin antibiotic placed on MHA media. Inhibitory testing was performed by the Kirby-Bauer diffusion method using paper discs on Mueller hinton agar medium (MHA) with 5 treatments and 3 repetitions. Media to be incubated at 36-37ºC for 24 hours. The results of this study indicate that the nutmeg nut extraction has a weak inhibitory effect on the growth of Escherichia coli bacteria. The extract  of nutmeg  seed  at concentration  of 1%  has inhibitory power  of 9.73  mm,  10%concentration of 8.5 mm, 20% concentration of 8 mm and positive control of 20.83 mm.Keywords : antibacterial, nutmeg ekstact, Escherichia coli.

Abstract

Tujuan dari penelitian ini adalah untuk mengisolasi bakteri Shigella sp dari sampel feses sapi yang berada di BPTU-HPT Indrapuri. Penelitian ini dilakukan dengan metode Cross-Sectional. Sampel feses diambil dari 85 ekor sapi, selanjutnya dilakukan isolasi bakteri Shigella sp menggunakan metode Carter. Isolasi Shigella sp dilakukan dengan menginokulasi feses sapi yang diambil menggunakan swab steril pada media Nutrient Broth (NB). Kemudian dilakukan penanaman pada media Eosin Methylene Blue Agar (EMBA). Dari 85 sampel 11 diantaranya diduga bakteri Shigella sp. Kemudian dilakukan perbanyakan bakteri pada media Nutrient Agar (NA). Selanjutnya dilakukan penanaman pada media Salmonella Shigella Agar (SSA) dan diinkubasikan pada suhu 37˚C selama 18-24 jam. Kemudian diambil 1 koloni yang terpisah untuk dilakukan perwarnaan Gram. Hasil penelitian menunjukkan tidak ada yang positif bakteri Shigella sp. Kesimpulannya tidak dapat diisolasi bakteri Shigella sp pada feses sapi aceh di BPTU-HPT Indrapuri. (The purpose of this study was to isolate Shigella sp bacteria from cow feces sample collected from BPTU-HPT Indrapuri. This research was conducted by Cross-Sectional method. The fecal samples were taken from 85 cows, followed by isolation of Shigella sp bacteria using Carter method, Shigella sp isolation was done by inoculating cow feces using sterile swab on Nutrient Broth (NB) medium. Then inoculated on Eosin Methylene Blue Agar (EMBA) medium. 11 out of 85 samples suspected shigella sp then was multiplied on Nutrient Agar (NA) medium followed by planted on Salmonella Shigella agar media (SSA) and incubated at 37˚C for 18-24 hours. A separate colony was then collected for Gram staining. The results showed no positive bacteria Shigella sp. The Conclusion is can not be isolated Shigella sp bacteria on the feces of aceh cattle in BPTU-HPT Indrapuri .)

Abstract

This study aims to evaluate the slaughter of qurban animals in Banda Aceh City and some of Aceh Besar in 2017. This research was conducted in the form of data analysis from questionnaire results of qurban slaughtering animals. The data analyzed was data obtained from the results of questionnaires and observations about some aspects which classified into four categories, namely qurban animals health aspects, aspects of shelter and qurban animals slaughter equipment, the process of slaughtering qurban animals, and fourth was the aspect of cutting meat or carcass. The data were analyzed descriptively. The results showed that 83.28% of the health aspects of qurban animals in Banda Aceh city and some of Aceh Besar can be said very good. As many as 84.12% of the people in Banda Aceh city and some of Aceh Besar have done the shelter and qurban animal slaughtering equipment. 76.56% of the people in Banda Aceh city and some of Aceh Besar have done the slaughtering of qurban animals in accordance with slaughtering requirements, and 88.88% have done carcass or carcass handled properly. The conclusion stated that the slaughtering of qurban animals in Banda Aceh City and some of Aceh Besar in 2017 has fulfilled the aspect of sanitation and animal welfare

Abstract

ABSTRAKSusu memiliki sumber gizi yang paling sempurna. Nilai gizinya yang lengkap menyebabkan susu dapat menjadi media yang sangat baik bagi mikroorganisme untuk pertumbuhan dan perkembangannya. Penelitian ini bertujuan untuk mengetahui jumlah cemaran Staphylococcus aureus pada susu kambing peranakan etawa (PE) di peternakan Adoe A Kabupaten Aceh Besar. Penelitian ini dilaksanakan di Laboratorium Kesmavet Fakultas Kedokteran Hewan Universitas Syiah Kuala, Darussalam Banda Aceh. Penelitian ini menggunakan 10 sampel susu kambing peranakan etawa. Pengambilan sampel dilakukan secara acak dan diencerkan dengan pengenceran desimal dari 101 sampai 106. Selanjutnya dilakukan penghitungan jumlah koloni dengan menggunakan Total Plate Count (TPC) untuk mengetahui jumlah cemaran Staphylococcus aureus. Hasil penelitian menunjukkan bahwa terdapat 30% sampel tercemar Staphylocoucus aureus. Nilai TPC tertinggi diperoleh dari sampel susu K3 yaitu 3,0 x 102 cfu/ml, dan nilai TPC terendah diperoleh dari sampel susu K1 yaitu 1,6 x 102 cfu/ml. Sehingga dapat disimpulkan bahwa nilai TPC yang ditemukan melebihi batas standar SNI untuk bakteri Staphylocoucus aureus pada susu yaitu 1 x 102 cfu/ml.ABSTRACTMilk has the most perfect source of nutrition its complete nutritional value causes milk to be an excellent for microorganisms for growth and development. This study aims to determine the amount of Staphylococcus aureus contamination in Etawa crossbreed’s milk on Adoe A farms in Aceh Besar Regency. This study was carried out at the Veterinary Public Health Laboratory, Faculty of Veterinary Medicine, Syiah Kuala University, Banda Aceh. This study use 10 samples of Etawa Crossbreed’s milk. Sampling was carried out randomly and diluted with a decimal dilution from 101 to 106. Furthermore, the number of colonies was counted using the Total Plate Count (TPC) to determine the amount of staphylococcus aureus contamination. The results of this study showed that 30% of the samples were contaminated with Staphylocoucus aureus. The highest TPC value was obtained from the K3 milk sample, which was 3.0 x 102 cfu/ml, and the lowest TPC value was obtained from the K1 milk sample, which was 1.6 x 102 cfu/ml. So it can be concluded that the TPC value found exceeds the SNI standard limit for Staphylocoucus aureus bacteria in milk, which is 1 x 102 cfu/ml.

Abstract

ABSTRAK          Telur itik merupakan salah satu produk hasil pengawetan telur dengan cara pengasinan. Penelitian ini bertujuan mengetahui pengaruh penambahan air perasan jeruk purut (Citrus hystrix) pada proses pembuatan telur asin terhadap jumlah total cemaran bakteri. Metode penelitian yang digunakan adalah metode rancangan acak lengkap (RAL) yang terdiri atas 4 perlakuan dan 3 kali ulangan. Perlakuan yang diberikan adalah penambahan larutan perasan jeruk purut dengan konsentrasi 0%, 10%, 20% dan 30% ke dalam media adonan pengasinan telur itik. Pengamatan jumlah total bakteri dilakukan setelah proses pengasinan telur dilakukan yaitu setelah 14 hari masa penyimpanan telur. Pengamatan jumlah total bakteri dilakukan dengan  metode Total Plate Count (TPC) menggunakan media Plate count agar (PCA).  Data jumlah bakteri yang diperoleh di analisis secara statistik menggunakan Analysis Of Variance (ANOVA). Hasil penelitian menunjukkan bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 0%, 10%, 20% dan 30% ke dalam adonan pengasin telur selama proses pembuatan telur asin dapat menurunkan jumlah total bakteri secara signifikan. Hasil uji lanjut menggunakan uji Duncan diketahui bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 30% secara nyata dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan dibandingkan dengan konsentrasi 0%, 10%, 20%. Sedangkan pembahan larutan perasan jeruk purut dengan konsentrasi 10% sampai dengan 20% tidak dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan. Berdasarkan penelitian ini dapat disimpulkan bahwa penambahan larutan perasan jeruk purut dengan konsentrasi 30% ke dalam adonan pengasin telur dapat menurunkan jumlah total bakteri pada telur asin yang dihasilkan. ABSTRACT            Duck egg is a product of egg preservation by salting.This study aims to determine the effect of adding citrus hystrix to the process of making salted eggs to the total amount of bacterial contamination. The research method used is a complete randomized design method (RAL) consisting of 4 treatments and 3 replays. The treatment given is the addition of a solution of orange juice purut with a concentration of 0%, 10%, 20% and 30% into the medium of duck egg marinating dough. Observation of the total number of microbes is done after the process of salting eggs is done after 14 days of egg acidification. Observation of the total number of microbes is done by the Method of Total Plate Count (TPC) using media Plate count agar (PCA) Data on the number of microbes obtained in statistical Analysis Of Variance (ANOVA). The results showed that the addition of a solution of orange juice purut with concentrations of 0%, 10%, 20% and 30% into the egg marinating dough during the process of making salted eggs can significantly decrease the total number of microbes. The results of further tests using Duncan's test found that the addition of a solution of orange juice purut with a concentration of 30% can significantly decrease the total number of microbes in the salted eggs produced compared to the concentration of 0%, 10%, 20%. While the deity of orange juice solution purut with a concentration of 10% to 20% can not decrease the total number of microbes in salted eggs produced. Based on this research, it can be concluded that the addition of a solution of orange juice purut with a concentration of 30% into the egg marinating dough can decrease the total number of microbes in the resulting salted eggs.

Abstract

ABSTRAK Ikan lele adalah ikan air sungai yang mengandung nutrisi yang tinggi. Kandungan gizi pada ikan lele yaitu lemak (4,8 %), mineral (1,2 %), protein (17,7 %) dan air (76 %). Ikan lele mudah membusuk sehingga perlu dilakukan pengolahan yang baik untuk menjaga kualitas ikan. Salah satu cara pengolahan untuk mencegah ikan membusuk yaitu dengan diasapkan. Penelitian ini bertujuan untuk membuktikan cemaran Escherichia coli pada ikan lele asap yang ada di Kecamatan Simpang Kiri Kota Subulussalam memenuhi Standar Nasional Indonesia (SNI) atau tidak. Sampel penelitian ini adalah ikan lele asap dari lima penjual. Dua ekor ikan lele asap dari setiap penjual sehingga total ikan berjumlah 10 ekor ikan lele asap. Perhitungan jumlah Escherichia coli menggunakan metode TPC (Total Plate Count). Sampel ikan dari setiap penjual ditimbang sebanyak 5g/ sampel kemudian dilakukan pengenceran menggunakan larutan Buffer Pepton Water (BPW) konsentrasi 0,1% dan diperiksa menggunakan uji TPC menggunakan media Eosin Methylen Blue Agar (EMBA). Data hasil pengamatan dianalisis secara deskriptif. Hasil penelitian menunjukkan seluruh sampel ikan dari lima pedagang didapatkan hasil positif pada media. Semua sampel ikan dari lima pedagang ikan lele asap di Pasar Tradisional Kecamatan Simpang Kiri Kota Subulussalam menunjukkan hasil positif tercemar bakteri Escherichia coli dan belum memenuhi SNI sehingga perlu untuk ditinjau kembali pada proses pengolahan serta cara penjualan. Kata kunci : Clariasis gariepinus, Escherichia coli, Kota Subulussalam, analisis deskriptif. ABSTRACT Catfish is a river water fish that contains high nutrients. The nutritional content of catfish is fat (4.8%), minerals (1.2%), protein (17.7%) and water (76%). Catfish is easy to rot so it needs good processing to maintain the quality of the fish. One way to prevent fish from rotting is by smoking it. This study aims to prove that Escherichia coli contamination in smoked catfish in Simpang Kiri District, Subulussalam City meets the Indonesian National Standard (INS) or not. The sample of this research is catfish as soon as possible from the five merchants. Two catfish as fast as possible from each merchants so that the total fish can catch 10 catfish as fast as possible. The calculation of the number of Escherichia coli uses the TPC (Total Plate Count) method. Fish samples from each merchant were weighed as much as 5g / sample then dilution was carried out using a 0.1% concentration of Buffer Pepton Water (BPW) solution and the cost using the TPC test using Eosin Methylene Blue Agar (EMBA) media. The data from the observations were descriptive. The results showed that all fish samples from five merchants had positive results on the media. All fish samples from smoked catfish merchants in the Traditional Market of Simpang Kiri Sub-district, Subulussalam City showed positive results contaminated with the Escherichia coli bacteria and were not Indonesian Nasional Standard so it is necessary to review the processing and sales methods.
