Abstract

Coleus amboinicus is one plant with a good antioxidant content. The concentration of antioxidant from C. amboinicus could be increased by using a treatment fermented plant extract (FPE) containing paitan grass. C. amboinicus is potential to be used as raw material to produce herbal tea such as green tea and black tea because of its high antioxidant content. The objective of this study was to analyze the total phenolics, flavonoids, and antioxidant activity from tea product of C. amboinicus. Antioxidant activity was analyzed using one method, that Ferric Reducing Antioxidant Power (FRAP). The results showed that C. amboinicus treated with FPE paitan grass had the highest total phenolic, flavonoid, and antioxidant activity content than control. The green tea produced using FPE paitan grass had the highest content of total phenolic and flavonoid i.e., 25,596 Gallic Acid Equivalent (GAE) mg/g of dry weight (DW) and 5,632 Catechin Equivalent (CTE) mg/g of DW, respectively. Antioxidant activity analyzed by using FRAP method also showed that the highest antioxidant was found in green tea treated using FPE paitan grass. It can be concluded that green tea C. amboinicus treated by FPE paitan grass is a good source of phenolic, flavonoid, and antioxidant.

Abstract

The objective of this study was to analyze the total phenolics, total flavonoids, andantioxidant activities of Amaranthus spinosus L leaves. During cultivation, these plantswere treated with 2 treatments, biocontrol Effective Microorganism 5 (EM5) garlic andcontrol (water). The A. spinosus L leaves were dried using freeze dryer and extracted bydissolving n-hexane, dicloromethane (DCM) and methanol. The results showed thatA.spinosus L leaves treated with biocontrol had the highest total phenolic, totalflavonoid and antioxidant activity content compare to the control. The total phenolic,total flavonoid, 2,2 diphenyl-1-picrylhydrazyl (DPPH) and Nitric Oxide (NO) radicalscavenging were the highest on DCM extract with each value of 74,656 mg Gallic AcidEquivalent (GAE)/g of dry weight (DW), 22,337 mg Catechin Equivalent (CE)/g DW,0,373 mg/mL and 0,025 mg/mL. n-Hexane extract had the highest antioxidant activityon Ferric Thiocyanate ( FTC) test (27,615 %), meanwhile methanol extract indicated thehighest antioxidant activity on FRAP test (1,022 mmol Fe +2 /g DW). From this study,it can be concluded that A. spinosus L leaves with biocontrol EffectiveMicroorganism 5 (EM5) garlic can increase the phenolic, flavonoids and antioxidantactivitiy contents of A. spinosus L plant.

Abstract

Beef is potential as a medium for microbial growth such as coliform bacteria since it has a high nutrients content. Coliform bacteria in a given amount could be used as a hygienic indicator and a sign for the presence of pathogenic bacteria. One species of coliform bacteria that often contaminate meat is Escherichia coli. Escherichia coli is a normal flora in gastrointestinal tract, but some strains of them are pathogens which cause acute diarrhea. The aim of this study was to calculate the number of coliformbacteria and to detect the presence of Escherichia coli in beef sold in traditional and modern markets in Pekanbaru using the Most Probable Number method. The results showed that all samples were found to be contaminated by Escherichia coli andcoliform bacteria. The number of coliform bacteria in all samples exceeded the maximum threshold of bacterial contamination according to Indonesian National Standard No. 7388:2009, i.e 1x102 MPN/g. The number of coliform bacteria in samplessold at traditional and modern markets were in the range of 0.11 x10 7 MPN/g to >24x10 7 MPN/g and 0.35 x10 8 MPN/g to >24x10 8 MPN/g, respectively.

Abstract

Dumai water samples are estimated to contain cellulolytic microorganisms because of farm waste. Samples 930 from Sei Pakning and 931 from Rupat Strait were water samples containing cellulolytic bacteria taken from Dumai. In this study the production of cellulase enzymes from cellulolytic bacteria in samples of 930 and 931 used CMC as a substrate. Cellulase is an enzyme that catalyses the hidrolisis of β-1-4-glycosidic bond of cellulose. All of the isolates produced cellulase and were able to degrade cellulose. The results indicated that 1% substrate gave the highest activity of Cellulase in the 931-4A (Rupat strait) (1.186 ± 0.319) x 10 -3 U/mL, whereas the highest specific enzyme activity of the bacterial isolates was 930-1C (8.438 ± 0.109) x 10 -3 U/mg.

Abstract

Coliform bacteria are often referred as indicator organisms to indicate the potential presence of disease-causing bacteria. Escherichia coli is a type of Coliform bacteria. Most Escherichia coli are harmless and commonly found in the intestines of humans and animals. However, some strains can cause illness. Coliform bacteria and Escherichia coli contamination in chicken could be detected using Most Probable Number (MPN) multi-step assay which consists of presumptive, confirmed, and completed test. The aim of this study was to determine the number of Coliform bacteria contamination and to detect Escherichia coli in 15 chicken meat samples collected from 3 traditional markets and 3 modern markets in Pekanbaru. Estimated numbers ofColiform bacteria in samples were varied from 2.1x10 4 MPN/g to >11x10 MPN/g. We found that chicken samples were contaminated by Escherichia coli, which showed colour changes and bubles production in EC broth and the appearence of dark centered colony with metallic shine on EMB Agar.

Abstract

Cellulolytic bacteria are  bacteria that can degrade cellulose into smaller polysaccharides called cellodextrins or completely into glucose units. Glucose is used as a carbon source for bacterial growth. The aims  of this study were to isolate cellulolytic bacteria from estuary water of Siak watershed in  Bengkalis  area and to determine its  cellulolytic activity and specific activity. A total of 13 isolates of cellulolytic bacteria have  been isolated using commercial cellulose substrate containing 1% w/v carboxymethyl cellulose (CMC). We found that 11 of the 13 isolates were Gram negative bacteria, while others were Gram positive. The isolate of C3 from Pelabuhan Roro showed the highest cellulolytic activity [(1.701 ±  0.166)  x 10-3 U/mL]  which was  not significantly different (p≤0.05) with cellulolytic activity of the  A3  isolate  [(1.605 ± 0.041) x 10-3 U/mL]. The C3 isolate also had the highest Cellulolytic specific activity [ (31.037  ± 4026) x 10-3 U/mg] and it was significant (p≥0.05) different with other isolates

Abstract

Coleus  amboinicus  is a plant with good antioxidant content . Antioxidants are very important  in preventing various diseases caused by free radicals.   The high antioxidant content of plant  C. amboinicus  potentially  used as raw material for making herbal tea products, such as  green tea and black tea. The objective of this research was to analyze the  total phenolics, flavonoids and antioxidant activity of green tea and black tea prepared from  C. amboinicus  plants with two different treatment, fermented plant extract (FPE) spice and control (water).  The results showed that the total phenolic and flavonoid content  of each  treatment  was different. The total phenolic content of control treatment and FPE spice green tea and black tea products  were  16,660 ± 0.251 mg AGE/g DW; 10.510 ± 0.737 mg AGE/g DW and 14,219 ± 0,198 mg AGE/g DW; 8.062 ± 0.334 mg AGE / g DW. Were  the total flavonoids treatment control and FPE spice green tea and black tea is 3.522 ± 0.077 mg CTE/g DW; 2.141 ± 0.104  mg CTE/g DW and 2.278 ± 0.057 mg CTE/g DW; 2.077 ± 0.044 mg CTE/g DW.  The content  of antioxidant activity by FTC method is highest shown  in the control treatme nt compared with ETT RR good green tea products and  black tea.  It can be concluded that green tea product of  C. amboinicus  is better  source of flavonoids, phenolics and antioxidant activity compared with the black tea product.
