Abstract

The objective of this research was to study the growth kinetic of Lactic Acid Bacteria    (isolate T5) originated from tempoyak. The results revealed that isolate T5 yielded a maximum OD value of  1,434 g/l, and biomass concentration of 2,553 g/g, while optimal growth kinetic of  isolate T5 were  9 hours. Abstract

Cassava (Manihot esculenta) is the third primary food in Indonesia after rice and corn. Cassava doesn’t have distinctive harvest period, therefore the harvesting period can be varied and resulting differences physical and chemical properties. Production rate and physical and chemical properties will vary due to soil fertility of cassava plantation. The purpose of this study is to analysis physical and chemical properties at Gunung Agung village Sekampung Udik district and Sinar Baru village Marga Tiga district, at Lampung Timur regency with different harvesting periods. The method used in this study was a descriptive method with two factors : harvesting age and plant location. The results show that Kasetsart variety with ten month of harvesting age at location A (Gunung Agung village Sekampung Udik) had better physical and chemical properties, and these can be seen from the fresh cassava water content (60,67%), cassava density (1,15 g/mL), starch content (35,93%), starch yield (18,94%), starch moisture (8,17%), amilose content (18,03%) and amylopectin content (81,97%) and enzymaticly conversion of starch to glucose (64,92%). Different plan locations and harvesting periods resulted in different physical and chemical properties. These indicated that Kasetsart variety grown at location A is more suitable to be converted to bioethanol compared to those of at location B. Abstract

Improving of antioxidant status can be achieved by consuming antioxidant containing-food.  Consumption of natural food that contains antioxidant will give positive effects for health if it is consumed according to regular on optimum and effective dosage.  Isoflavones are the active compound in soybean.  By measuring immunohistochemical technique, it was proven increasing the profile of copper, zinc-superoxide dismutase (Cu,Zn-SOD) in the liver, kidney and testis of rats that were given isoflavones by oral administration.  As phenolic antioxidants, isoflavones have a potential role for protection of cells in those tissues because of the capability of isoflavone as scavenger free radicals. Abstract

The effects of the addition of mixed inoculums consisted of Saccharomyces cerevisiae, Lactobacillus lactis and Acetobacter aceti on the chemical and microbial changes during cacao fermentation were investigated. The fermentation conditions were designed as natural fermentation (without any inoculum addition), fermentation added with mixed inoculums at zero fermentation time, and fermentation added with S. cerevisiae, L. lactis, and A. aceti at the zero, first, and second day of fermentation, respectively. The fermentation was conducted in the sweat box of 5 kg for 5 days and sampling was done at every 12 hours. The result showed that the addition of mixed inoculums at zero time speeded the optimum fermentation time. Microbial inoculums added at the first or second day of fermentation are not recommended. Abstract

The objective of this research was to optimize the product of Shiroyutaka sweet potato instant rice processing.  This research was done in three steps of optimation process.  The first step was the addition of water with the ratio of 1:1.5, 1:2, and 1:3, the second step was the temperature of added water, and the third step was the binder compound addition (0, 5, and 10%).  The results indicated that the optimal characteristic of instant rice was obtained by addition of 5% tapioca, addition of hot water to form a dough, forming, cooking in rice cooker until it gets cooked, drying in the form of plate, and milling. The product had the composition of 6.68% water, 82,78% of total carbohydrate, 1,73% of  protein, 1,38% of fat, 5,84% of fiber, and 1,59% of ash. Abstract

The purposes of this work were to find the effects of ethanol-PKO ratio and reaction time on the yield and antibacterial activities of palm kernel oil ethanolysis products.  The treatments were ratios of ethanolyisis mediums and duration of agitation. The medium treatments were  ethanol 96% (NaOH 1%; w/w of oil) solution and PKO at ratio of 0,4; 0,7; 1,0; 1,3 and 1,6 (v/w) and the duration of agitation on a shaker at 300 rpm for 4, 6, 8, and 10 minutes.  Antibacterial activities were tested using Staphylococcus aureus and Escherichia coli. The results showed ethanol-PKO ratio at 1,6 (v/w) effectively increased the yield of ethanolysis products for 4 up to 10 min, while the treatments at ratio of 0,4; 0,7; 1,0 (v/w) did not affect significantly, and at ratio of 1,3 (v/w) was optimum for 4 min. Ethanolysis products and their fractions for all treatments had antibacterial activities toward two bacteria (E. coli and S. aureus) with the average diameter of zone inhibition was 5,38 mm/15 mg.  The optimum ethanol-PKO ratio and time of reaction were 1,3 (v/w) and 10 minutes to produce ethanolysis products, primarily for fraction 1 (uper layer after fractionation). Abstract

This work was conducted to develop a new measuring system for nondestructive dry matter prediction in sawo fruit using short wavelength near infrared (SW-NIR) spectroscopy. In this research, a number of 100 sawo fruits were used as samples. Spectra were acquired using a portable spectrometer (VIS-NIR USB4000, The Ocean Optics, USA) with 100 ms integration time and 50 scans for number of scanning. Dry matter was measured using oven drying. The calibration and validation model was developed using the partial least squares (PLS) regression method. The result showed that the best calibration model could be developed for original spectra in the wavelength range of  700-990 nm with F= 8, r = 0.92, SEC = 0.68 and  SEP = 0.86. Abstract

The aim of this work was to obtain the minimal concentration of chitosan which possess antibacterial characteristic. The concentrations of chitosan that used were 0%; 0,2%; 0,4%; 0,6%; and 0,8% (w/v), whereas bacteria cultures used to examine antibacterial activity of chitosan were Escherichia coli, Bacillus subtilis and Staphylococcus aureus. Well method (gel diffusion) was utilized to test the chitosan antibacterial activity.  The result was analyzed using descriptive method and presented in the graphic form.  The result showed the activity of chitosan restraint as antibacterial.  The test of chitosan antibacterial activity utilizes gel diffusion (well) method. The data analyzed descriptively and presented in the graphic form.  The result  showed the activity of chitosan restraint as anti bacteria against Staphylococcus aureus, Bacillus subtilis or Escherichia coli with the highest restraint diameter was noticed at the addition of 0,2% chitosan, which were 20,27 mm/mg chitosan, 24,50 mm/mg chitosan and 31,53 mg chitosan, respectively.  The lowest restraint diameter was found at the addition of 0,8% chitosan, which were 6,82 mm/mg chitosan, 12,14 mm/mg chitosan and 14,23 mm/mg chitosan, respectively. Abstract

The objective of this research was to measure antibacterial activities of ethanolysis products that produced through ethanolysis reaction of mixture of palm kernel oil (PKO) and Morinda seeds oil (25:1 and 50:1, w/w) in ethanol solution contained 1% NaOH (v/w based on oil weight) at ratio of 5 : 6 (b/v) and shaken on a shaker (300 rpm, room temp.) for 3, 6, 9 and 12 minutes. Fractionation of ethanolysis products using absolute ethanol (1:3; v/v) and centrifuged at 4.000 rpm for 30 min, yielding Fraction 1 (upper layer) and Fraction 2 (lower layer).  Generally, all ethanolysis products showed antibacterial activities against four test bacteria (E. coli, S. aureus, B. cereus and S. enteritidis) at range of d values (mm) from 0,93 (B. cereus; 3 min) up to 3,87 (E. coli; 9 min). Fraction 1 showed the highest antibacterial activity with d values (mm) from 1,20 (B. cereus; 9 min) up to 5,63 (E. coli; 9 min), while Fraction 2 for all treatments had no antibacterial activity, relativelly.  Ethanolysis products and its fraction (Fraction 1) were resulted from ethanolysis reaction for 6 and 9 min showed higher antibacterial activities than those for 3 and 12 min.
