ABILITY OF BACTERIA DEGRADING HYDROCARSBN COMPOUNDS IN WASTE OIL FROM WORKSHOP SOIL

Abstract

ABSTRAKAbstrak: Limbah oli mengandung logam berat (besi, timah, kadmium, mangan) yang bersifat non biodegradable dan senyawa yang bersifat toksik seperti: poly chlorinated biphenyls (PCBs) dan polycyclic aromatic hydrocarbons (PAHs). Hidrokarbon dari limbah oli dapat menyebabkan berbagai resiko penyakit, sehingga upaya untuk memulihan lingkungan dengan bioremediasi dengan menggunakan mikroorganisme. Degradasi hidrokaron dapat dilakukan dengan bantuan mikroorganisme salah satunya adalah bakteri. Penelitian ini bertujuan untuk mengetahui karakteristik dan potensi bakteri dalam mendegradasi hidrokarbon. Sampel menggunakan tanah bengkel yang tercemar limbah oli bekas. Isolasi dilakukan dengan metode pour plate pada media NA (Nutrient Agar) dan diperoleh sebanyak 15 isolat bakteri, terdapat 3 isolat bakteri gram negatif dan 12 isolat bakteri gram positif, keseluruhan isolat bakteri berasal dari genus Pseudomonas dan genus Bacillus. Berdasarkan uji kemampuan degradasi oli kedua jenis isolat ini mampu tumbuh pada setiap konsentrasi oli 1%, 2% dan 3%, isolat dengan kode SB 13 (Pseudomonas) menunjukkan nilai absorbansi yang paling baik pada media NB (Nutrient Broth) dengan konsentrasi 1% sedangkan isolat SB14 (Bacillus) menunjukkan hasil absorbansi tertinggi pada konsentrasi 1%, nilai absorbansi tertinggi pada isolat SB 13 yaitu 1.650, nilai absorbansi tertinggi pada isolat SB 14 yaitu 1.357.Kata kunci : Degradasi hidrokarbon, limbah oli, bioremediasiABSTRACTAbstract: Waste oil contains heavy metals (iron, tin, cadmium, manganese) which are non-biodegradable and toxic compounds such as poly chlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarSBns (PAHs). Hydrocarbons from waste oil can cause various disease risks, so efforts to restore the environment by bioremediation using microorganisms. Hydrocarbon degradation can be done with the help of microorganisms, one of which is bacteria. This study aims to determine the characteristics and potential of bacteria in degrading hydrocarbons. The sample used workshop soil contaminated with used oil waste. Isolation was carried out by pour plate method on NA (Nutrient Agar) media and obtained as many as 15 bacterial isolates, there were 3 isolates of gram negative bacteria and 12 isolates of gram positive bacteria, all of the bacterial isolates came from the genus Pseudomonas and genus Bacillus. Based on the oil degradation potential test, these two isolates were able to grow at each oil concentration of 1%, 2% and 3%. isolate SB 14 (Bacillus) showed the highest absorbance at a concentration of 1%, the highest absorbance value in isolate SB 13 was 1,650, the highest absorbance value was at isolate SB 14 was 1,357.Keywords : Hydrocarbon degradation, waste oil, bioremediation
