Abstract

ABSTRAKSenyawa metabolit sekunder merupakan senyawa yang diproduksi suatu makhluk hidup untuk mempertahankan kelangsungan hidup, sebagai bentuk upaya mempertahankan diri dari predator alaminya. Ikan buntal adalah satu organisme laut yang memiliki potensi dalam menghasilkan metabolit sekunder. Tujuan dari penelitian ini untuk menganalisis perbandingan larutan eluen dan fraksi yang optimal untuk menentukan senyawa metabolit sekunder flavonoid pada daging ikan buntal Arothron manilensis. Metode penelitian dilakukan menggunakan metode Skrining Fitokimia dan Kromatografi Lapis Tipis (KLT) untuk mendeteksi senyawa metabolit sekunder. Hasil penelitian menunjukkan bahwa penentuan eluen, kepolaran eluen maupun pereaksi yang digunakan sangat memengaruhi nampaknya profil senyawa metabolit sekunder. Pada fraksi etil asetat senyawa metabolit sekunder flavonoid terlihat paling kuat sedangkan pada fraksi n-hexana senyawa flavonoid terlihat lemah. Kandungan senyawa metabolit sekunder pada ikan buntal Arothron manilensis mengandung komponen senyawa flavonoid. Kandungan senyawa metabolit sekunder flavonoid pada ikan buntal A. manilensis menunjukkan nilai Rf 0.5-0.975 yang memiliki potensi sebagai antioksidan. Diperlukan kembali pengujian High Performance Liquid Chromatography (HPLC), untuk memisahkan molekul senyawa metabolit sekunder dalam waktu minimum.Kata Kunci: Arothron manilensis, Flavonoid, KLT, Skrining Fitokimia.ABSTRACTSecondary metabolites are compounds that are produced by living things to maintain life, as a form of effort to defend themselves from natural predators. Pufferfish are the only marine organisms that have the potential to produce secondary metabolites. The purpose of this research is to analyze the comparisonof eluent solutions and optimal fractions to determine the secondary metabolite compounds of flavonoids in the flesh of the pufferfish Arothron manilensis. The research method was carried out using Phytochemical Screening and Thin Layer Chromatography (TLC) methods to detect secondary metabolites. Research results showed that the determination of the eluent, the polarity of the eluent, and the reagents used greatly affect the appearance of the secondary metabolite compound profile. In the ethyl acetate fraction, the secondary metabolite flavonoid compound is the most prominent, while in the n-hexane fraction, the flavonoid compounds are not visible. The content of secondary metabolites in puffer fish (Arothron manilensis) contains flavonoid compounds. The content of flavonoid secondary metabolites in puffer fish A. manilensis showed an Rf value of 0.5-0.975 which has potential as an antioxidant. High Performance Liquid Chromatography (HPLC) testing is needed again, to separate the secondary metabolite compound molecules in a minimum time.Keywords: Arothron manilensis, Flavonoids, Phytochemical Screening, TLC
