Abstrak- Kegiatan budidaya rumput laut di wilayah pesisir berpotensi memengaruhi kondisi ekosistem dasar perairan, khususnya komunitas makrozoobentos yang berperan sebagai bioindikator kualitas lingkungan. Penelitian ini bertujuan untuk menganalisis kepadatan dan keanekaragaman makrozoobentos pada lokasi budidaya rumput laut di Desa Tesabela, Kecamatan Pantai Baru, Kabupaten Rote Ndao. Penelitian dilaksanakan pada bulan Juli 2025 menggunakan metode kuadran transek pada tiga stasiun pengamatan dengan total 27 titik pengambilan sampel. Parameter yang dianalisis meliputi kepadatan populasi dan indeks keanekaragaman Shannon–Wiener (H’), serta didukung oleh pengukuran suhu, salinitas, dan pH perairan. Hasil penelitian menunjukkan bahwa makrozoobentos yang ditemukan terdiri atas 13 spesies dari dua filum, yaitu Mollusca dan Echinodermata. Kepadatan makrozoobentos berkisar antara 12,56–14,67 ind/m², dengan nilai tertinggi pada Stasiun II (14,67 ind/m²), diikuti Stasiun I (12,89 ind/m²), dan terendah pada Stasiun III (12,56 ind/m²). Nilai indeks keanekaragaman berkisar antara 0,364–0,368, yang tergolong rendah dan mengindikasikan komunitas makrozoobentos berada dalam kondisi tertekan. Kondisi tersebut diduga dipengaruhi oleh aktivitas budidaya rumput laut dan tekanan antropogenik di sekitar lokasi penelitian. Kata Kunci: Makrozoobentos, Kepadatan, Keanekaragaman, Budidaya Rumput Laut Abstract- Seaweed cultivation activities in coastal areas have the potential to affect the condition of the aquatic ecosystem, especially the macrozoobentos community which acts as a bioindicator of environmental quality. This study aims to analyze the density and diversity of macrozoobentos at the seaweed cultivation location in Tesabela Village, Pantai Baru District, Rote Ndao Regency. The research was carried out in July 2025 using the transect quadrant method at three observation stations with a total of 27 sampling points. The analyzed parameters included population density and the Shannon–Wiener diversity index (H'), and were supported by measurements of water temperature, salinity, and pH. The results showed that the macrozoobentos found consisted of 13 species of two phylum, namely Mollusca and Echinoderma. The density of macrozoobentos ranged from 12.56–14.67 ind/m², with the highest value at Station II (14.67 ind/m²), followed by Station I (12.89 ind/m²), and lowest at Station III (12.56 ind/m²). The diversity index value ranged from 0.364–0.368, which was relatively low and indicated that the macrozoobentos community was under stress. These conditions are thought to be influenced by seaweed cultivation activities and anthropogenic pressures around the research site. Keywords: macrozoobentos, density, diversity, seaweed cultivation
