Antioxidant Activity Evaluation of Extracts from Different Accessions of Red Betel (Piper ornatum) Leaves in East Java Province

Abstract

As a natural antioxidant, the red betel leaves (Piper ornatum) exhibit several biological functions. The purpose of this study is to assess the phytochemical profile and antioxidant activity of several red betel leaves accessions that come from different parts of East Java Province, including Jombang (JB), Blitar (BL), Batu (BT), Trenggalek (TG), and Probolinggo (PB). The secondary metabolite content and antioxidant activity of the five accessions were evaluated qualitatively using phytochemical screening, and their antioxidant activity was measured using the DPPH assay at a single concentration of 500 ppm following maceration extraction with 70% ethanol. Alkaloids, flavonoids, phenols, saponins, and tannins were found in all accessions of red betel leaves (Piper ornatum) ethanol extracts from the East Java Province.  The highest antioxidant activity of red betel leaves ethanol extract was obtained from the Blitar (BL), Batu (BT), and Jombang (JB) accessions, with inhibition percentages of 98.13 ± 0.52, 95.64 ± 0.90, and 95.18 ± 0.17%, respectively. Meanwhile, lower antioxidant activity was observed in the ethanol extracts of red betel leaves from the Probolinggo (PB) and Trenggalek (TG) accessions, with inhibition percentages of 91.54 ± 0.41% and 91.16 ± 1.18%, respectively. These differences were statistically significant (p < 0.05, one-way ANOVA, Tukey). The antioxidant activity correlates with the phenolic and flavonoid content of the plant, supporting the potential of red betel leaves as a natural antioxidant source.
