Optimization of Lactose and Avicel PH 101 as Fillers in Bilimbi Leaf Extract Tablets as Candidate of Antidiabetic Drug

Abstract

Oral hypoglycemic medications for diabetes can have significant side effects. Bilimbi leaves extract is an antidiabetic agent formulated in tablet form with a combination of lactose and Avicel PH 101. The study aimed to determine the compounds in the extract, determine the optimal proportion of lactose and Avicel PH 101, and determine the optimal antidiabetic activity of bilimbi leaves extract tablets in alloxan-induced mice. The research method was carried out experimentally with maceration extraction using 70% ethanol solvent and continued with phytochemical screening. Optimization of bilimbi leaves extract tablets combined with lactose and Avicel PH 101 used the Simplex Lattice Design (SLC) method with responses of weight uniformity, hardness, friability, and disintegration time. Verification of the optimal formula used a one-sample t-test. Antidiabetic analysis was measured using ANOVA and indicated by the percentage of reduction in blood sugar levels in each group. The results showed that bilimbi leaves extract contains flavonoids, tannins, and saponins. The optimum formula composition is lactose 78.375mg and avicel PH 101 167.925mg. The results of the optimum formula verification have a sig>0.05 in each response, so the data is valid because there is no significant difference with the Simplex Lattice Design prediction. The results of the antidiabetic test in mice showed that bilimbi leaf extract tablets have potential as an antidiabetic agent because there was a reduction in blood glucose levels after alloxan induction.
