Sembukan leaves (Paederia foetida L.) are medicinal plants. This plant contains secondary metabolites, namely flavonoids, which have antidiabetic activity. This study was conducted to determine the effect of purification on total flavonoids levels and antidiabetic activity of sembukan leaf extract and to determine the correlation between the two. The sample was in the form of sembukan leaves that were extracted and purified to remove ballast substances such as chlorophyll and odor-causing components. Determination of total flavonoids levels was carried out by UV-Vis spectrophotometry using AlCl3 reagent and antidiabetic activity was tested based on the extract’s ability to reduce glucose levels and expressed as EC50 values. Based on the results of the study, the purified extract of sembukan leaves (ETDS) had a higher total fkavonoid content of 36,1209 mgQE/gram compared to the crude extract of sembukan leaves (EKDS) of 12,2018 mgQE/gram. The purification process affects the antidiabetic activity of the sembukan leaf extract with the EC50 value of the purified sembukan leaf extract (ETDS) of 15,3548 ppm and the crude sembukan leaf extract (EKDS) of 20,0568 ppm. Statistical analysis shows that there is a significant difference between the crude sembukan leaf extract (EKDS) and the purified sembukan leaf extract (ETDS) (sig.<0,05) and there is a very strong negative correlation (r = -1,000) between the total flavonoid content and the EC50 value. These result indicate that the higher the total flavonoid content of the sembukan leaf extract, the smaller the EC50 value, as an indicator that its antidiabetic activy is stronger. These results indicate that the purification process can increase the flavonoid content and the potential antidiabetic activity of the sembukan leaf extract
