Background: Methicillin-resistant Staphylococcus aureus (MRSA) is a major nosocomial pathogen that shows high resistance to many antibiotics, especially beta-lactams. Katuk leaves (Sauropus androgynus L.) contain bioactive compounds such as phenolics and flavonoids that may act as natural antibacterial agents. This study aimed to determine the total phenolic and flavonoid contents and to evaluate the antibacterial activity of ethanol extract of katuk leaves against S. aureus ATCC 25923 and MRSA ATCC 33591. Methods: This experimental laboratory study included sample preparation, maceration extraction with ethanol, total phenolic determination using the Folin-Ciocalteu method, total flavonoid determination using the AlCl3 colorimetric method, and antibacterial testing using the disk diffusion method. Data were analyzed descriptively as mean ± SD. Results: The katuk leaf extract contained 25.74 ± 1.05 mg GAE/g total phenolics and 14.45 ± 0.26 mg QE/g total flavonoids. Antibacterial testing showed weak inhibition against MRSA with a 2 mm zone of inhibition and against S. aureus with a 1 mm zone of inhibition. Conclusion: Ethanol extract of katuk leaves contains relatively high levels of phenolics and flavonoids, but at 10% (w/v) concentration it exhibited weak antibacterial activity against S. aureus and MRSA. Further studies using higher concentrations or combined antibacterial agents are needed to improve inhibitory activity.
