Basil leaves (Ocimum x africanum Lour.) and binahong leaves (Andredera cordifolia (Ten.) steenis) are known to have antibacterial activity that can inhibit acne-causing bacteria. The purpose of this study was to formulate an acne patch preparation combining ethanol extracts of basil leaves and binahong leaves and to determine the physical evaluation and antibacterial activity of the acne patch preparation. This study was analytical, using ethanol extracts of basil and binahong leaves obtained through maceration with 70% ethanol. Acne patches were formulated in four variations of extract concentrations, namely F0 (0%), FI (20% basil, 15% binahong), FII (25% basil, 12.5% binahong), and FIII (30% basil, 10% binahong). Antibacterial activity tests were carried out using the well diffusion method, using mediklin as a positive control. The results showed that the acne patch preparation, a combination of ethanol extracts of basil and binahong leaves, met the organoleptic test requirements, pH, moisture content, weight uniformity, thickness, and folding strength. The antibacterial activity test showed a very strong antibacterial activity against S. epidermidis bacteria, with F0 (0.00 mm) as a negative control, FI (26.2 mm), FII (27.3 mm), FIII (27.4 mm), and K+ (medicinal) of 29.6 mm. Activity against C. acnes bacteria was also classified as very strong, with inhibition zones of F0 (0.00 mm), FI (26.3 mm), FII (27.3 mm), FIII (27.5 mm), and K+ (28.1 mm). It is recommended that future researchers conduct further research on physical evaluation tests using more adequate tools and materials. Further in vivo testing is also recommended to determine the biological effects of the acne patch before application to humans.
