The purpose of this study is as follows: (1) To know the method of single bud breeding; (2) To know the influence of Rootone-F growth regulator on growth of single bud cassava seeds; And (3) to know the effective concentration of growth regulating substances in the cultivation of single bud cassava. The study was conducted for 15 days at the Laboratory of the Faculty of Agriculture of SOERJO Ngawi University. Materials used in this study include Cassava, Rootone-F, Aquades and Planting Media (Sand, Compost, Soil). While the tools used in this study include saws / cutting knives, glass mineral water, pipettes, measuring cups, beaker glass, spatula and handsprayer (spray tool). Research findings: (1) Single bud system is a plant hatching system using 1 bud eye. Cassava stalks that are more than 6 months old in pieces and then taken the eyes of his buds that are in each segment of cassava stems; (2) Plant growth regulators in plants are non-nutrient organic compounds, which in small amounts may support, inhibit and alter the physiological processes of plants; (3) The plant growth regulator in the plant consists of five groups namely Auxin, Giberelin, Cytokinin, Ethylene and Inhibitor with distinctive features and different effects on physiological processes; (4) The use of rootone-f on a single bud breeding system is used to help accelerate the growth of the root system. The growth regulator substances contained by rootone-f ie IAA, IBA, and NAA work on root meristem tissue to form a new root system; (5) Single bud breeding system in this cassava includes a new technology and innovation in agricultural world in Indonesia. keywords: growth regulators, cassava plants (manihot esculenta), single bud method
