Abstract

Concentration of protein which found in freshmilk, sweet ketchup and salt ketchup have been done byusing hollow fiber polysulfone membrane with differentmolecular weight cut - off (MWCO). Membranes usedin these experiments have MWCO 66,000, 87,000 and 149,000 Dalton. Experimental were conducted bycontinues flow system and parameters observed in these experimental were protein, sugar and salt, respectively.Protein infresh milk rejected by membrane above 98 %,but all of sugar pass through to the permeate side. Forsweet ketchup feed sample, rejection coefficient towardsprotein was smaller than feed fresh milk. The higherrejection coefficientfor sweet ketchup was 48 % for themembrane with MWCO 66.000 Dalton. Rejectioncoefficient of membrane towards protein for salt ketchup sample was smaller than sweet ketchup sample. For saltketchup, the higher rejection coefficient towards proteinwas 27 % for the membrane with MWCO 66,000Dalton. Rejection coefficient of membrane towardsNaCl for feeding solution sweet ketchup and saltketchup were in the rlJ.1Jgefrom 8.74 to 23.96 %.Key words : Hollow fiber membrane, ultrafiltration, protein,milk, ketchup.

Abstract

Fermentation broths of TSC 18 and TSC 22 fungi isolates obtained from Taxus sumatrana plant were investigated for their in vitro anti breast cancer activity against T47D cell line. The isolates were further identified by PCR and sequencing methods. In the fermentation experiments, the growth curves were constructed by plate count method and the pH change were recorded, while glucose and protein dynamics of the broth were determined by Nelson-Somogy and Lowry methods respectively. Anti-cancer activity was assessed by SRB (Sulforhodamine B)method. The investigation indicates the fermentation broth both isolates possessed the anticancer activity with lese of 42.6 ppm for TSC 18 and 55.7 ppm for TSC 22. Cisplatin which is used as a reference compound gave ICso of 15.9 ppm . The PCR and tree view program proved that TSC 18 is Phomopsis sp. strain MAFF-238472 and TSC 22 is Coprinopsis cinerrea strain NBRC30628.Keyword: Anticance, Isolates TSC 18 and TSC 22 Taxus Sumatrana.

Abstract

Antibiotic UK-3A contains a 9-membered dilactone ring. It had been isolated as a minor component from the mycelium of 5trepyomyces sp. 51701.The antibiotic was hypothesized to be potential to inhibit the growth of leukemia cancer cell line of P388 and KB with ICso 38 and 20 Dg/mL, respectively. To understand the effect of lipophilicity increase of the analogues on their anticancer activities based on QSAR parameter (Log P) and binding energy to BcL-xL protein. To produce analogues of UK-3A, 3hydroxypicolinyl serine methyl ester (A) was synthesized from 3-hydroxypicolinic acid and L-serine methyl ester. The product was then esterified by pentanoic (1), hexanoic (2), heptanoic (3), and octanoic (4). The final products were confirmed with IHand 13C FT-NMR and FTIR spectra, and also MS spectra. Then they were tested against P388 Murine Leukemia cells. The result of bioassay showed lipophilicity increase of3hydroxypicolinyl serine methyl octhyl ester (PSMOE) correlated positively with their anticancer activity increase, withICso 15.4mg/mL against P388 cell lines.Keywords: Anticancer, Streptomyces sp 517-02, UK-3A, Analog UK-3A and P388

Abstract

In-house cocoa flour RMs instead of CRMs was prepared and checked of its homogeneity and stability. Due to CRMs are limited availability in terms of matrices of concern and costly (high production cost) making in-house cocaflour RMs is alternatively used for daily quality control in the laboratory. The analysis method for reliable checking of homogeneity and stability of cadmium content in the in-house cocoaflour RMs was studied. The effects of magnesium nitrate and lanthanum/magnesium nitrate as chemical modifiers in the determination of cadmium in cocoaflour by GFAAS were studied as well. Detection limit of method was estimated to be 0.07 ug L'(calcuiaied as 3Sy;/slope of calibration curve) and calibration curve of working range was linear. The results analysis for candidate of in-house cocoaflour RMs reveals the consistence results and no variability between and within-bottle observedat 95% limit of confidence and stabile during the period of time observed.

Abstract

Has been done research to know anticancerpotencial activity from fragrant screw pine leaf extractapplies method Brine Shrimp Lethality Test ( BSLT).Extract is made by the way of macerate to apply threekinds of solvent, that is butanol, ethyl acetate, and etherpetroleum. Toxicity test is done by using prawn larvaArtemia salina Leach which age 48 hours. Toxic effecteach extract is identified with presentase death of prawnlarva applies probit analysis (LC50). Active extract thenis tested its the phytochemistry content and compoundbioa1(fi! suggested applies GC-MS. Result of his (itsshowing ethyl acetate extract to have the character oftoxic ( LC50 : 288,4 ppm). Abstract

Optimum condition of carbamate are carbofurane and carbaryl recidues pesticide analysis by height performance liquid crhomatography (HPLC) has been done. The optimum condition are isocratic mobile please on acetonitrile/H20 :45/55 (v/v) and wave lenght of ultra violet (UV) detector has optimum absorbance at 275 nm. Performance of HPLC verification has minimum detection are 60 pbb for carbofurane and 40 ppb for carbaryl. Linierity and work range are 0.06 to 0.15 ppb for carbofurane with r2 value of 0.9702 and 0.04 to 0.15 ppb for carbaryl with r2 value of 0.09874. Precision of HPLC was showed by relative standard deviation (RSD), for carbofurane are 0.30 % of retention time (Rt) and 7.2 % of area. Whereas for carbaryl, the RSD are 0.20 % for Rt and 4.63 % for area. Homogeneity of targets in tomato matric as well as carbaofurane and carbaryl are homogen by analysis of varians (ONOVA) method which. is showed by F-value <  F-critis. Homogeneity anlysis by National Acreditation of Comeetee (KAN) method also homogen with criteria I. Keywords: Homogeneity, Pesticide, Carbamate, Carbofurane,Carbaryl, Chromatographs.

Abstract

Experiments were conducted to find optimum medium and process conditions based on shake-flask and laboratory fermentor scales to anticipate industrial scale for the production of glucoamylase. Membrane filtration and ammonium sulfate precipitation were tested for the enzyme concentration and isolation respectively. The glucoamylase enzyme properties were determined and preference to various starch substrates were checked. Immobilization within commercial agar and alginate were done and their shelf-life were observed. The application of free glucoamylase in saccharification of the various starches were also investigated after being liquefied with commercial alpha-amylase.Keywords : rhizopus oryzae, glucoamylase, isolation, immobilization.

Abstract

Mistletoes are used traditionally in Indonesiaas medicinal plant, one of this as anticancer. Cancer isone of degenerative diseases could be triggered by toomuch free radicals in the body, whereas antioxidantcould reduced the risk of degenerative diseases.Mistletoes reposted content flavonoids which known have antioxidant activity. Mistletoe potency as drugmaterial should be studied sa the utilization ofmistletoe could be developed. Antioxidant activitytest using "DPPH free radical scavenger" methodand toxicity test using BSLT (Brine Shrimp LethalityTest) method had been conducted on water andethanol extracts of mistletoe Dendropthoe pentandra(L.) Miq.) that grown on various trees (star fruit,mango, cananga, 'duku, sour-sop, kepel, mahkotadewa, and tea). The result showed that ethanol andwater extracte-ofD, petandra on all host plants haveantioxidant activity (IC50 value between 6.4 - 51.8ug/mL). The result of toxicity test showed that ethanolextract of mistletoe on cananga, starfruit, kepel andmahkota dewa host plants have toxicity effect againstA. salina larvae with LCso value below 1000 ug/mL,nevertheless ethanol extract on others host plants andwater extract didn It give toxicity effect against A.salina larvae (LC50 > 1000 ug/mL).Key words: mistletoe, D. petandra, DPPH, antioxidant,toxicity, degenerative disea.
