Abstract

Endophytic microbe is a definition which is given to all microorganisms that during a more or less long period of their life, colonize symptomnlessly the living tissues of their hosts. The endophytes can produce useful substances such as oligosaccharide degrading enzyme, antimicrobial substances and plant growth regulator. Quisqualis indica is one of tropical herb that is widely known for traditional medicine. There is no information about endophytic microbes of this plant. The aim of this study are to isolate the endophytic microbes from Q. indica, and screen on their ability in producing antimicrobial substances. Isolation is conducted by direct seed plating method using PDA and NA medium. Liquid fermentation method is taken place using supernatant F4 and PDY medium, then followed by bioassay of the supernatant on three test microbes: Staphylococcus aureus, Escherichia coli and Candida albicans. Result of isolation is 25 bacterial isolates and 12 fungal isolates. Result of bioassay showed that there are some isolates that have ability in producing antimicrobial substances. There are 1 bacterial isolate and 2 fungal isolates that showed wide spectrum activity of antimicrobial substance.

Abstract

Infeksi Ganoderma pada tanaman kelapa sawit merupakan permasalahan utama dalam pengendalian penyakit pada perkebunan kelapa sawit di Indonesia. Sampai saat ini infeksi Ganoderma baru diketahui setelah muncul tubuh buahnya pada batang tanaman kelapa sawit, dan pada saat itu tanaman kelapa sawit sudah tak bisa dipertahankan lagi dan tingkat penyebarannya sudah meluas. Penelitian ini bertujuan untuk mengidentifikasi spesies Ganoderma yang banyak menyerang tanaman kelapa sawit di Indonesia baik dari segi morfologinya maupun dengan pendekatan biologi molekuler, yaitu berdasarkan pada peta genetik (sequence) 18S rRNA dan ITS (Internal Transcribe Spacer). Hasil penelitian menunjukkan bahwa Ganoderma yang banyak menyerang tanaman kelapa sawit di Indonesia adalah Ganoderma boninense. Dari hasil karakterisasi telah didapatkan peta genetik spesifik (specific sequences) ITS dari Ganoderma boninense dibandingkan dengan spesies Ganoderma yang lain, sehingga peta genetik spesifik ini dapat dijadikan dasar untuk mendeteksi secara dini adanya serangan Ganoderma boninense pada areal perkebunan kelapa sawit di Indonesia. Pendeteksian secara dini terhadap serangan Ganoderma ini akan sangat bermanfaat untuk menentukan proses penanganannya dan pada akhirnya diharapkan akan dapat meminimalisasi tingkat kerugian yang terjadi (1).

Abstract

Some endophytic microbes were isolated from medicinal plant Artemisia annua collected of Tawangmangu and Cibinong. Microbe isolate were made on corn meal malt agur supplemented with chloramphenicol and nutrient agar supplemented with nystatin from tissue subjected after the triple surface sterilization procedure. The potencies of these isolates to produce-terpene compounds were screenned by phytochemistry analysis, thin layer chromatography (tlc) and high performance liquid chromatography (hplc) after fermentation in four different medium. The results showed that 6 isolates from 17 microbe isolates can produce terpene compounds.

Abstract

Caesalpinia major Dandy ex Excell is one of the traditional medicinal plants from Nusa Tenggara Timur Province (Flores island, Ruteng) which the local people called as “Dekar". The root of this plant is used as anthelmintic, anti rheumatic, back-ache and also as tonic. Chemical structure elucidation of isolated compound from C. major caesaldekarin A has been done and it's based on ultraviolet, infrared, nuclear nagnetic resonance and mass spectrometry: To know the correlation between proton and proton, proton and carbon, carbon and carbon, NMR two dimensional (2D) have been also interpreted by the data of 'H-'H COSY, 13C-'H COSY, COLOC, HMBC and INADEQUATE. The stereochemistry of the relative structure of caesaldekarin A is determined by 2D NMR NOESY.

Abstract

Generally guajava is known as a plant, which all of its parts are used as adstringent. But research results shown that this plant has some efficacies, for instance us analgesic, antibacterial, antidiabetic, antispasmodic, anti inflamation etc. Recently the people also use the ripe guajava fruit juice, especially the red one, as a remedy for of dengue infection, which its scientific background is not known yet. Therefore many research activities should be done in order to know what efficacies are shown by any part of guajava plant.

Abstract

Kaempferia angustifolia Rosc. (Zingiberaceae) dan K. rotunda L. di Jawa dikenal sebugai Kunci pepet dan Kunir putih. Kedua macam tumbuhan tersebut biasanya tumbuh di daerah hutan sekunder di Jawa. Rhizoma dari K. angustifolia berbentuk serbuk kering atau tunbuhan segar biasa dijual di pasar tradisional di Indonesia, dan dikenal sebagai pencegah dan pengobatan kanker secara tradisional. Ekstrak khloroform dari rhyzoma K. Angustifolia menunjukkan efek antioksidan (scavenging) yang signifikan terhadap radikal bebas 1,7-difenil2 pikril hidrasil (DPPH). (IC50 = 370 ug/mL). Dua macam senyawa dari ekstrak khloroforin telah diisolasi dan diidentifikasi. Senyawa 1, suatu khalkon yang telah dikenal, yaitu, 2'hidroksi-4', 4', 6'-trimetoksi khalkon adalah senyawa yang aktif (IC5, = 97 ug/mL). Senyuwu 2, suatu sikloheksan epoksid yang telah dikenal, (+)-krotepoksida, tidak aktif (IC;, = 690 ugi mL) Struktur senyawa 1 dan 2 diidentifikasi berdasarkan data spektroskopik, MS dan 2DNMR (HMQC dan HMBC) dan dibandingkan dengan data yang telah dipublikasikan.

Abstract

The modern products of biotechnology are, vaccine, interferon, interleukin, monoclonal antibody, new rice varieties, antibiotics, biological pesticides such as bacteria Bacillus thuringiensis which are active against specific insect species and their application through transgenic plants. There is a great difference in the products if compared with those of the conventional biotechnology, such as soy sauce, fish sauce, yoghurt, ethanol and tempe. Nowadays HIV/AIDS, pneumonia virus SARS (severe acute respiratory syndrome), genetic disorder, cloning, biological weapon such as anthrax from Bacillus anthraxis, are the kinds of several diseases, discoveries and conditions which look for development and clarification through biotechnology. It was shown in this paper some new biotechnology products, technology scope closely related to biotechnology and also the comparation of national priorities and strategy for developing of biotechnology in developed and developing countries.
