Abstract

Busuk lunak yang disebabkan oleh Pectobacterium carotovorum (Syn. Erwinia carotovora subsp. carotovora) merupakan salah satu penyakit penting pada tanaman sawi putih. Bakteri ini menghasilkan enzim pektinase yang dapat menguraikan pektin pada dinding sel tanaman bagian lamela tengah. Salah satu alternatif pengendalian penyakit busuk lunak ialah menggunakan agens biokontrol dari filosfer. Penelitian ini bertujuan menyeleksi dan menguji bakteri filosfer dari daun kubis yang berpotensi menghambat penyakit busuk lunak P. carotovorum pada sawi putih. Bakteri filosfer diisolasi dari wilayah Cianjur, Tegal, dan Bogor. Bakteri filosfer diuji keamanan hayati berdasarkan pada reaksi hipersensitivitas dan kemampuan lisis pada agar-agar darah, serta diuji kemampuan penghambatannya terhadap P. carotovorum berdasarkan pada uji antagonis secara in vitro dan uji penghambatan penyakit secara in vivo. Bakteri filosfer dikarakterisasi berdasarkan pada morfologi koloni dan sifat Gram. Dua galur bakteri filosfer paling berpotensi mengendalikan penyakit busuk lunak pada sawi putih diidentifikasi sebagai Chryseobacterium sp.

Abstract

Probe DNA sebagai Kandidat Biosensor Clavibacter michiganensis subsp. michiganensis pada Tanaman Tomat Penelitian dilakukan untuk menguji teknik deteksi gen menggunakan DNA probe spesifik untuk mendeteksi gen tomA pada Clavibacter michiganensis subsp. michiganensis, bakteri penyebab kanker pada tomat. Probe dirancang menggunakan program Primer3Plus, dilabel dengan molekul nonradioaktif digoxigenin (DIG) dan digunakan pada metode hibridisasi dengan teknik dot blot. Sampel uji terdiri atas dua jenis, yaitu sampel DNA genom yang berasal dari kultur bakteri murni dan dari benih tomat yang diinfeksi buatan dengan C. michiganensis subsp. michiganensis. Sampel yang berasal dari kultur murni bakteri menunjukkan hasil hibridisasi positif pada semua tingkat konsentrasi DNA; sedangkan sampel yang berasal dari biji tomat hanya menunjukkan reaksi positif pada konsentrasi 10, 8, 6, dan 4 µg µL-1. Studi ini menyimpulkan bahwa probe yang dirancang berpotensi untuk digunakan dalam pengembangan metode deteksi C. michiganensis subsp. michiganensis pada biji tomat berbasis biosensor dan bersifat cukup spesifik karena tidak ada reaksi silang dengan kelompok bakteri bukan target.

Abstract

Cendawan akar putih (Rigidoporus microporus) merupakan salah satu patogen penting yang dapat menyebabkan kerusakan di pertanaman karet. Kemampuan hidup yang tinggi sebagai saprob pada tunggul tanaman dan sisa-sisa akar yang mati menjadikan bagian ini sebagai sumber inokulum penting, baik untuk tanaman karet yang sehat di sebelahnya maupun pada saat melakukan penanaman ulang. Oleh karena itu, pengurangan sumber inokulum ini menjadi kunci penting untuk mencegah penyebaran lebih lanjut. Solarisasi tanah merupakan salah satu metode disinfeksi tanah sebelum tanam dalam pengendalian beberapa hama penyakit tertentu dan dapat mengurangi penggunaan senyawa sintetik di dalam tanah. Penelitian ini bertujuan menentukan solarisasi tanah dan penambahan bahan organik terhadap kelangsungan hidup R. microporus. Solarisasi tanah secara nyata menekan kelangsungan hidup R. microporus pada kedalaman lokasi patogen, yaitu 5 dan 15 cm dari permukaan tanah. Sementara itu perlakuan bahan organik tidak berpengaruh nyata dalam menekan perkembangan R. microporus. Semakin lama waktu solarisasi, kemampuan penekanan terhadap daya hidup inokulum semakin tinggi. Hasil penelitian kami menunjukkan bahwa solarisasi tanah memicu aktivitas bakteri kelompok fluoresens yang diduga dapat menekan inokulum patogen penyebab busuk akar tersebut.

Abstract

Insect Vector and Seedborne Transmission of Papaya ringspot virus Ringspot disease of papaya caused by Papaya ringspot virus (PRSV) is widely spread in Indonesia. Dissemination of this disease is known to occur through infected seedlings and aphid vectors. This study was conducted to determine transmission efficiency of PRSV through two aphid species, i.e. Aphis gossypii and Myzus persicae and to confirm that PRSV cannot be transmitted through seeds. Aphid transmission of PRSV isolate from Medan was carried out in papaya var. California with an acquisition feeding period and an inoculation feeding period of 10 minutes each. A minimum of 5 A. gossypii and 10 M. persicae were required for successful PRSV transmission. Transmission of PRSV by A. gossypii resulted higher disease incidence and more severe disease symptoms than transmission by M. persicae. Disease symptoms was not observed in all papaya seedlings grown from seeds extracted from fruits showing ringspot symptoms. Detection of PRSV by reverse transcription polymerase chain reaction method showed no amplification of specific DNA fragment of PRSV. The results of this study confirmed the potential of aphids as PRSV vectors and proved that PRSV was not seed borne.

Abstract

Liquid smoke to Control Bulkholderia glumae and Growth Promoter of Rice Seeds Bacterial grain rot disease caused by Bulkholderia glumae is an important disease that can be transmitted through rice seeds. Various bacterial grain rot disease control techniques have been developed to prevent crop loss, one of which is liquid smoke. Liquid smoke has been widely studied as an agent for controlling various plant diseases. The aim of the study was to determine the potential of liquid smoke to control bacterial grain rot disease caused by B. glumae and to promote the growth of rice seedlings. Research was conducted in some stages, included: potential test of liquid smoke against B. glumae, phytotoxicity test of liquid smoke and its effect on rice seed growth; and test the effectiveness of liquid smoke in suppressing B. glumae on infected seeds. The results showed that liquid smoke with a concentration of 2% could suppress the development of B. glumae in vitro and did not cause phytotoxicity in rice seeds with a vigor index of 90% and germination of 90% compared to controls with a vigor index value and germination value of 76% and 83%. Liquid smoke with a concentration of 2% can also reduce the severity of disease in the nursery phase with a relative inhibition level of 43.8%. The value of the length of the plumule of rice seedlings in the 2% liquid smoke treatment was 5.87 cm. The plumule value was greater than the control which was only 5.22 cm. In contrast to the plumula, the value of radicular length did not increase significantly compared to the control.
