Abstract

Genetic analysis from three species of Indonesia Cervidae (sambar deer, Cervus unicolor; rusa deer, Cervus timorensis; and Bawean deer, Axis kuhlii) was conducted to analyze their relationship. Tissues and blood from twelve sambar deer, one rusa deer and three Bawean deer were collected and analyzed for 12SrRNA using Primer forward (L1091) and reverse (H1478). The results indicated the amplication of mtDNA were 389 base nucleotide. There were 22 polimorphic sites, which were dominated by transition and gave 9 haplotypes that were 5 in sambar deer, 1 in rusa deer and 3 in Bawean deer.

Abstract

This study was aimed to isolate and identify a gene that was responsive to drought stress in sugar cane variety M442-51, a variety that is widely used in pastures or dry land. Under drought stress, M442-51 showed an enhance expression of several proteins. One of the protein, namely dip22, (drought inducible protein, with a molecular weight of 22KDa) was isolated and analyzed. Amino acid sequence of this protein showed some homology with several stressed related proteins (OsAsr 1 from Oryza sativa leaves, ZM Bss1 of Zea mays leaves, LeAsr 1, LeAsr 2 and LeAsr 3 of Lycopersicum lycopersicon leaves). The 343 bp of dip22-cDNA fragment was obtained using RT PCR from mRNA of sugar cane leaves that has been exposed to drought stress. The results indicated that the gene dip22 plays an important role as a regulatory protein that are usually nested within cytoplasmic matrix or in the nucleus.

Abstract

Ultra struktur sperma ikan baung (Mystus nemurus) telah diamati dengan menggunakan transmisi dan imbas elektron mikroskop (TEM dan SEM). Hasil penelitian menunjukkan bahwa spermatozoa normal ikan baung terbagi atas tiga bagian yaitu; kepala, leher yang pendek, dan bagian ekor. Kepala sperma ikan baung berbentuk bulat yang diseliputi oleh membran sitoplasma, pada bagian leher terdapat mitokondria, sedangkan ekornya bersifat tunggal dan irisan melintangnya menunjukkan bahwa centriol mikrotubulusnya terbentuk atas “9+2” axoneme.

Abstract

Auxin has an important role to control both of the growth and tropism of gynophore and the development of fruit and embryo of peanut (Arachis hypogaea (L). Merr.). The experiment was carried out to examine the contents of auxin during peanut development, i.e. at the time of anthesis (day 0), at day 4, 7, 10, 15, 18, 23, and 31 after anthesis, using High Performance Liquid Chromatography (HPLC) with fluorescence detector method.Between the day of anthesis to the day 31 after anthesis, the auxin contents changed according to fruit development stage. The free auxin contents in the developed fruit (entering the soil) were higher than undeveloped fruit (not entering the soil), while the bound auxin content in the developed fruit were lower than undeveloped fruit. The lowest free auxin contents was found at the time of anthesis, then increased drastically when the gynophore grew fast and in the beginning of embryo development stage (day 7). Between the day 7 to the day 15 after anthesis, the free auxin contents were decreased. In the development fruit, the free auxin contents increased when the fruit begin to grow (day 15-18), then decreased until the seed reached its full size (day 31). In the undeveloped fruit, the free auxin contents decreased at day 7 to day 31. The bound auxin contents in the developed fruit decreased until the day 18, and increased gradually until day 31. In the undeveloped fruit, the bound auxin contents decreased at day 15 and afterward.

Abstract

The aims of this research was to obtain diversity of hydrocarbonoclastic yeast isolated from Tanjung perak harbor Surabaya. Exploration of yeast was conducted by isolation and identification of isolated yeast. Identification of yeast based on characteristics of colonies, cell shape, and biochemical tests. From this research, it was obtained 9 hydrocarbonoclastic yeasts. They were 8 generas: Rhodotorula, Candida, Geotrichum, Torulopsis, Trichosporon, Cryptococcus, Debaryomyces, and Saccharomyces.

Abstract

Our previous experiment showed that prenatal exposure of rats to X-radiation in the late gestation period causes heterotopic Purkinje cells in the internal granular layer (IGL) of the cerebellum. The present study demonstrated precisely the involvement of Reelin in the aligment of Purkinje cells, which are different in severity between anterior and posterior lobes. Pregnant rats were exposed to 2.5 Gy X-radiation on gestation day 21 and the cerebellum of progeny was examined histologically and by immunohistochemistry of IP3 Receptor to identify Purkinje cells. By P8, while Purkinje cells with well developed dendrites aligned underneath the EGL in the control cerebellum, Purkinje cells with shorter and abnormally oriented dendrites failed to align and remained in the heterotopic location in the IGL of the anterior lobes but a small number of heterotopic Purkinje cells formed in the posterior lobes of the irradiated rats. Our previous published Reelin is present in the premigratory zone of the EGL and IGL in the control cerebellum. It decreased in the irradiated cerebellum on P1 to P8, especially the anterior lobes, however in posterior lobes on P8 Reelin level compared to the control (Darmanto, 2003). It is thus indicated that anterior lobes are more sensitive to X-radiation than posterior lobes and the different levels of Reelin between anterior and posterior lobes caused by X-irradiation may contribute the different susceptibility in induction of derangedPurkinje cells.

Abstract

The dried rice stalks and banana stem are waste product after their harvesting but have potential as organic compost. Organic compost fertilizer treatment has a purpose to give nutrient for growth media until the root had enough and grow well. In this study, a peanuts root of two cultivars, kelinci and gajah, were grew in soil growth media which enrich an organic compost fertilizer, i.e. dried rice stalks, banana stem and mix of both composting with Agri Simba as starter, would be compared with control to look the kinds of growth media, cultivars and their interaction affect on rooting. The method used in this study was on experiment with on experimental design called completely randomized design. Each unit of the treatment tested four times. Many parameters would be studied in this case: length of the primary root, sum of live root apical, sum of nodules. They analyzed by two ways ANOVA test. Anatomical structure was analyzed by descriptive method. Harvesting of the cell have done 8th and 20th day after planting. The result of this study showed that organic composts have variation content of the N, P, and K on three kind of compost, certainly. The growth media treatment affected significantly (p < 0.05) in peanut rooting at less on root length and on sum of live root apical in early growth phase and sum of nodules in secondary growth. The cultivars treatment affected significantly (p < 0,05) on peanut rooting at less on sum of live root apical but not (p > 0.05) on root length and sum of nodules. Their interaction treatment not affected (p > 0.05) on rooting parameter observed. The treatments may affect on anatomical structure.

Abstract

This study was designed to identify the effects of nicotine on the skeletal and internal organ abnormalities of mice prenatally. Seventy two female mice strain Balb/C were used. Pregnant mice on gestation days (GD) 8, 10, and 12 was injected intraperitoneally with 3, 6, and 12 mg/kg BW nicotine respectively. Control nice was injected by aguabidestylata. On GD 18, and all of pregnant mice were sacrificed, and a half of living fetuses were immersed in Bouin’s solution to observe their internal organs, while another half were immersed in 95% ethanol to observed sekeletal malformation using alizarin red S staining. Results were analyzed using SPSS program version 11 for personal computer.The results showed that nicotine caused Skeletal malformation, especially in the sternum when administered on GD 8 and 10, in doses of 3 and 6 mg/kg BW, delayed of ossification was observed in the supraoccipital and sacrocaudal bones on GD 10 and 12 and doses of 12 mg/kg BW, while in the extremitas the malformation was observed in the proximal and medial phalanx both in the fore and hind limb especially on GD 10 and 12, and doses of 6 and 12 mg/kg BW. Nicotine was effects on the brain development especially hydrocephalus was observed on GD 8 and doses 10 and 12 mg/kg BW. Another internal malformation such as ectopic renalis was observed on GD 8 and doses of 3 and 6 mg/kg BW. These finding suggested that nicotine has caused skeletal malformation, brain malformatin and ectopic renalis, when administered in mice prenatally.

Abstract

Peperomia pellucida L. Kunth herbs (Herba Suruhan) is one of the alternative for inflamation therapy. In this study, albino rats were devided into seven groups. The first three groups was administered orally the extract of Suruhan herbs 1500 mg/Kg body weight, 2000 mg/Kg body weight and 2500 mg/Kg body weight. The next three groups were given Indomethacine solutions with the doses of 13.5 mg/Kg body weight, 15.8 mg/Kg body weight and 15 mg/Kg body weight, and the last groups (control) were given 1% of PGA suspension alone. Fifty microliter of Carrageen solution of 1.5% was injected subcutaneously to all rats in those group.The volume of the foot paw of albino male rat injected with carrageen, was measured and recorded for 4 hours. The number of leucocyte and the number of differential count were observed every two hour, namely at the fourth, sixth and eight hours following carrageen injection. There were significant differences in the antiinflamation effect using plethysmometer and the number of leucocyte counts between control and all the treatment group. For the differential count, not all the results showed significant differences.

Abstract

Curcuma zedoaria, familia Zingiberaceae, has been used for along time as an alternative healing. active compounds within it are coneol, comphene, borneol, camphor, zedoarin, resin, gum, and curcumin. Curcumin is an antioxidan that can slow cell damages caused by toxicant. The purpose of this research was to find out the role of Curcuma zedoaria’s rhizome for minimizing damage of microanatomical structure of liver caused by alcoholic toxicant. Samples in the research were 24 mice (Mus musculus) Swiss Webster. They were grouped randomly into 8 groups. Each group was treated as follow. Group A (control group) was given fresh water. Group B was given whisky. Group C was given 500 mg Curcuma zedoaria/kg body weight. Group D was given 750 mg Curcuma zedoaria/kg body weight. Group E was given Curcuma zedoaria/kg body weight. Group F was given whisky and, 2 hours later, 500 mg Curcumazedoaria/kg body weight. Group G was given whisky and, 2 hours later, 750 mg Curcuma zedoaria/kg body weight. The last group was given whisky and, 2 hours later, 1000 mg Curcuma zedoaria/kg body weight. The treatment was carried out during 8 days. A microanatomy slide of liver was made by using parafin method and Hematoxylin-Eosin dying. Data was analyzed by using descriptive method, comparing between the control group and the experiment groups.The result of the research showed that the powder of the Curcuma zedoaria’s rhizome has been useful in minimizing damage of hepar microanatomy structure of the mice affected by alcohol. The effective dosage was 1000 mg/kg body weight.
