Abstract

The aim of this research was to study the effect of a-pinene on number of cell that express iNOS (inducible Nitric Oxide Synthase) on lipopolysaccharide (LPS)-induced neuron-glial culture. Neuron-glial culture were divided into eight groups: (1) normal; (2), (3) and (4) incubated with a-pinene (0,2; 0,4 and 0,8 μg/mL respectively, for 24 hours); (5) incubated with LPS 1 μg/mL; (6), (7) and (8) incubated with a-pinene (0,2; 0,4 and 0,8 μg/mL respectively, for 24 hours) and then induced with LPS (1 μg/mL, on 1 last hour of treatment period). The expression of iNOS in neuron-glial culture was detected by immunocytochemistry using anti-iNOS rabbit polyclonal IgG and goat anti-rabbit IgG Biotin Conjugated. The result shown that iNOS were expressed on normal group. Lipopolysaccharide 1 μg/mL induction causing iNOS expression increased approximately three times higher than normal (from 9,7% became 28,7%). Inducible NOS expression on non LPS-induced neuron-glial culture did not affected by a-pinene. Whereas a-pinene 0,2 and 0,4 μg/mL could suppress iNOS expression on LPSinduced condition. a-pinene 0,2; 0,4 and 0,8 μg/mL could help neuronglial cells survived on LPS-induced condition, but normal neuron-glial culture’s density was decreased when incubated with a-pinene 0,8 μg/mL.  Keywords: alpha-pinene, cell culture, glial, iNOS, LPS, neuron

Abstract

Penelitian bertujuan mengetahui pengaruh sari seduh teh hitam terhadap pengekspresian PPAR ã sel adiposa jaringan lemak visera Rattus norvegicus strain Wistar. Duabelas ekor tikus (Rattus norvegicus strain Wistar) jantan umur 6-8 minggu, berat 200 gram, diberi 4 macam perlakuan, yaitu A (diet tinggi lemak + SSTH 0 g/hari), B (diet tinggi lemak + SSTH 0,015 g/hari), C (diet tinggi lemak + SSTH 0,030 g/hari) dan D (diet tinggi lemak + SSTH 0,045 g/hari) selama 90 hari. Setelah masa perlakuan, tikus dibedah dan diambil lemak viseranya. Jaringan tersebut kemudian dibuat preparat dengan metode parafin. Jumlah sel yang mengekspresikan PPAR ã dianalisis dengan menggunakan pewarnaan immunohistokimia dan untuk mengkonfirmasi bentuk sel adiposa digunakan pewarnaan Hematoxylene&Eosin. Antibodi primer yang digunakan adalah anti PPAR gamma poliklonal antibody rabbit IgG dan antibodi sekunder biotin-goat-anti rabbit IgG. Hasil penelitian menunjukkan jumlah sel adiposa yang mengekspresikan PPAR ã mengalami penurunan seiring dengan penambahan dosis SSTH. Hal tersebut mengindikasikan SSTH dapat menurunkan pengekspresian PPAR ã pada sel adiposa jaringan lemak visera. Kata kunci: jaringan lemak visera, PPAR ã, diet tinggi lemak, sari seduh teh hitam

Abstract

The seed of Nigella sativa has long time ago employed as medicinal remedy for numerous disorders. N.sativa has active compounds in its seed likes nigellisine, nigellidine, nigellimine-N-oxide, thymoquinone, dithymoquinone, thymohydroquinon, nigellone, thymol, arvacrol, oxy-coumarin, 6-methoxycoumarin, 7-hydroxy-coumarin, alpha-hedrin, steryl-glucoside, flavonoids, tannins, essential amino acid, ascorbic acid, and minerals. The seed has various curative activities. As body immunity stimulator by stimulating the formations of bone marrow and many kind of antibody cells, antihistamine, anti-hypertension, anti-inflammation, antimicrobial activities by protecting bodies from viruses and decreasing the risk of infections, anti-diabetic (metabolic syndrome), and anti cancer. These papers have to discuss deeper about the beneficial aspect of N. sativa seed against metabolic syndrome and cancer. Based on several research known that N. sativa has potential to decrease total cholesterol, low density lipoprotein and blood glucose level. This seed also has potential to maintain the homeostatic of blood vessels as the backbone of the circulatory system in the body. At the cancer case, this seed substances has potential to decreasing the development of cancerous cells, kills the cells by enhances its apoptotic programs and also lessening the metastasis of the cancer cells. Keywords: cancer, metabolic syndrome, Nigella sativa
