Abstract

The objective of  this research was  to identify the effect of various dosages of sweet basil essential oil in estrus cycle of Sprague-Dawley rats. Twenty four healthy female of Sprague-Dawley rats, with the age of 2 month, 120-140 gr of body weight were used in this experiment. After estrus synchronization using Whitten effect, the rats were divided randomly into 4 groups, such as groups I, II, III and IV, treated with dosages of  ½ x, x, and 2x, while the control rats were given extract solvent of 5% polyethylene glycol. Vaginal smears and Giemsa staining were  used  to determine each estrus cycle phase. Each estrus cycle could be distinguished according to type of cells appeared in preparation of vaginal smears under microscope with 40x. The change of duration of estrus cycle everyday (proestrus, estrus, metestrus, and diestrus) was  recorded. The result indicated phase of proestrus had the longest change in time (2.43±0.54 days in x dosage), while for control: 1.23±0.47, 0.5x dosage: 2.02±0.41  and 2x dosage: 1.98±0.28.  In estrus phase, the longest phase was 2.10±0.46 at 2x dosage, while other phase durations for control, 0.5 dosage and x dosage were 1.65±0.35, 1.87±0.37 and 1.90±0.27, respectively. The result also indicated that duration of proestrus was significantly different compared to others (P<0,05). Based on this result, it could be conclouded sweet basil essential oil can affect duration of estrus cycle in Sprague-Dawley rats, especially proestrus (P<0,05).Keywords: sweet basil essential oil, estrus cycle, vaginal smear, Sprague-Dawley rats, proestrus

Abstract

Cryopreservation technique can be used to preserve animal cell, plant, or other genetic materials  including  frozen semen. Herpestes javanicus is one of wild animals with a large number but no data about the sperm quality and technique how to preserve it. This research is proposed to know sperm quality of H. javanicus and the technique to preserve it. In this research, the three different methods were used to collect sperms. Those methods were  electroejaculator, flushing  and maceration/cutting, respectively. The sperm was then analyzed for quality, such as:  volume, pH, viscosity, colour, smell, motility, concentrasion, viability, morphology and morphometry. Then the sperms were  preserved in tris citrate glucose egg yolk extender by different concentrations. The result showed that the best method for collecting sperms was  by maceration/cuting method, and the best extender for sperms was low concentration of tris cytrate glucose egg yolk. Keywords: cryopreservation, Herpentes javanicus, sperm quality, preserve method, extender

Abstract

One of the important reproductive disorders in dairy cows was repeat breeding. Economic losses was quite high, due to the low reproductive efficiency and productivity.  Breeder is one of the risk faktors on the incidence of repeat breeding. The aims of this research were to determine the incidence  of repeat breeding cases in dairy cattle at Special Regency of Yogyakarta (DIY) and many factors from the farmers and dairy cattle aspects that caused the cases. In this study, 401 traditional farmers at DIY were used. The method of data collection was done by interviewing the farmers and direct observation to the dairy cattle. The data consist of farmer education level, length period of farming experience, stall condition, length distance from inseminator, estrous detection ability,knowledge of estrous, estrous observation by farmer, stall cleanliness, cattle cleanlines, and squeezing time.Data were collected on a descriptive analysis of each variable studied. Analysis used logistic regression models to analyze the faktors that influence the incidence of repeat breeding. The sensitivity and specificity of the models were analyzed by Lemeshow goodness of fit test.The result of this study showed that the prevalence of repeat breeding in dairy cattle from farmer aspect was 29.4%. The factor from farmer aspect which affected repeat breeding cases was estrus detection once per day(coefficient regression 4.09563, OR=60,1),  detection of estrus twice daily (+ 2.92553, OR=18.6) , disordersof pregnancy (+2.22630, OR= 9.3), dirty environment (+0.91889, OR=2,5), worst drainage (+2.58072, OR=13,2), and education  (+1.31295, OR=3,7). It was concluded that the prevalence of repeat breeding at DIY as big as 29.4%. Risk factors affecting the repeat breeding were estrus detection once per day, detection of estrus twice a day, dirty environment, worst drainage, and education. Keywords : prevalency, repeat breeding, dairy cattle, logistic regression, odds ratio

Abstract

Intestinal motility and contractility in cats can be affected by many factors that originate from inside and outside the body. Anesthesia is one of the factors that could influence it.This research was conducted to figure the effect of tiletamine-zolazepam on the intestinal motility and contractility in the cats through contrast radiography study. Three male cats weighing between 2,5-3,5kg were used in this present study. Radiography procedure was performed twice on each cat: without anesthesia and one week later with anesthesia (0,02 mg/BW atropine sulphate and 10 mg/BW Zoletil®).The cats were given a positive contrast, barium sulphate (BaSO4) 30% w/v (12 ml/kg BWper oral) followed by radiography study. Radiography was performed at 5, 30, 60, 120, and 180 minutes after administration of BaSO4. The cats were positioned on laterolateral view. Data were analyzed descriptively using zone division and quantitatively by measuring the difference between the intestines diameter during contraction and relaxation. The intestinal diameter was measured and statistically tested using ANOVA. Result showed that in minute 180, BaSO4 reached zone four in the unanesthetized cat while in the anesthetized cat, it was still empty. The diameter differences of the unanesthetized cat was higher compared to the anesthetized meaning that the intestinal contraction in the unanesthetized cat was higher. This means that tiletamine-zolazepam anesthetic reducedof intestinal motility andcontractility. Keywords: cats, barium sulphate, gastrointestinal motility, contrast radiography, tiletamine-zolazepam

Abstract

A Golden retriever dog, male, 9 years old was a patient of Surgery Clinic, Department of Surgery and Radiology, Faculty of Veterinary Medicine, Gadjah Mada University. The patient was dated on September 27, 2012. Clinically observation showed skinny, normal appetite, and not showing any physiologic change except that there were superficially bumps on the inguinal skin 4 cm in diameter size. The tumor biopsy was performed using lidocaine 2% as local anaesthetic circullary application. Histopathologically analysis of the tumor tissue was identified as hemangiosarcoma.
