Abstract

Avian Influenza (AI) virus  is pathogenic agent that can spread from one area to another area through the transportation of infected animals or their products such as feathers. This research was aimed to inactivated AI virus with 775 ppm sodium hypochlorite and 0.1% chloroxylenol to examine the difference of treatment by time (day) for inactivation AI virus on the feathers. AI virus isolate A/chicken/sidrap/ 07160336‐2/2016 used in this research was obtained from Balai Besar Veteriner Maros. The treatment of disinfectants were performed on the first day (the day of disinfectan solutions were prepared), the third day and the seventh day by soaked the feathers in disinfectant solution for 10 minutes. The effectiveness of disinfectans were evaluated by inactivation index. The result show that the average of inactivation index of 775 ppm of sodium hypochlorit was 4.17 for the first day, 5.17 for the third day, and 4.20 for the seventh day, while the average of inactivation index of 0.1% chloroxylenol was 5.50 for the first day, 6.43 for the third day, and 5.77 for the seventh day. Our result indicated that the sodium hypochlorit and chloroxylenol were effective for inactivation of AI virus. The 0.1% of chloroxylenol was more effective for inactivation AI virus than 775 ppm of sodium hypochlorit, whilst the most effective duration for the treatment is the three days.

Abstract

Chronic Respiratory Disease (CRD) is a bacterial infection in chickens caused by Mycoplasma gallisepticum. CRD may result in economic loss in the livestock industry, decreasing of egg production and feed efficiency, increasing of  medication costs, downgrading of carcass and egg qualities. The aim of this study was to develop the Enzyme-Linked Immunosorbent Assay (ELISA) kit for detecting antibodies of M. gallisepticum using freeze-thawing antigens. The cut-off point was determined by S/N method. Chicken antiserum againts M. gallisepticum was collected every week on 1st – 6th week post immunization of whole cell antigen of M. gallisepticum. Optimization of antigen, serum and conjugate were 10 μg/ml protein concentrations, 1:800 serum dilution and 1:10000 conjugate dilution. The validation results on developed ELISA kit (MyGELISA) analyzed  by ROC curve using MedCalc statistical software revealed that developed ELISA kit (MyGELISA) had 100% sensitivity and 86.21% specificity with 95% confidence interval. The results of RSA, MyGELISA and commercial ELISA kit showed antibodies positif againts M. gallisepticum.  In conclusion, MyGELISA using freeze-thawing antigen can be used to detect antibodies of M. gallisepticum.

Abstract

Both of Nectarinia jugularis parents, male and female, usually raise their offsprings together. They feed the chicks, carry away faecal sac and keep their offspring safe from any threats such as predators. On one day the female did not appear, it was not clear why she did not come back home. Since then, the male took care of the chicks by feeding them, disposing the faecal sac and keep the chicks save all day, even though he had never slept in the nest at night as the female did. The male tried to increase the feeding rate and faecal sac removal to compensate the absence of the female. Parental care by single parent male continued until they leave the nest. The male still fed the chicks after they flew off from the nest.

Abstract

The prevalence of antibiotic resistance is an increasing problem due to the use of false and excessive antibiotics. The use of antibiotics in pig farms is increasing and needs to be aware of side effects, it is also influenced by the knowledge, attitudes and practices of farmers. This study is a cross-sectional study using a structured questionnaire interview to analyze the relationship between knowledge and attitudes towards the practice of using antibiotics by pig farmers in Kupang city. A total of 96 respondents pig farmers spread over 6 districts, District of Maulafa (9 villages) District of Alak (12 villages) District of Oebobo (7 villages), District of Kelapa Lima (5 villages), District of Kota Raja (2 villages) and the district of Kota Lama (1 village). The variables observed were individual characteristics, knowledge, attitudes and practices of the use of antibiotics by pig farmers in relation to the occurrence of antibiotic resistance. The relationship between the variables was tested using the Pearson correlation, while the relationship and magnitude of the direct and indirect impact on the observed variables was determined using path analysis. The results showed knowledge has a very significant impact (p <0.01) and indirectly contributes to the level of antibiotic usage practices preventing the occurrence of antibiotic resistance. Attitudes have a significant effect (p <0.05) and directly affect the practice of antibiotics to prevent the use of antibiotic resistance in Kupang. Conclusion of this study shows that knowledge has a major impact on the practice of using antibiotics by pig farmers, so by increasing farmers' knowledge of the facts incidence of antibiotic resistance, the attitude of farmers will increasingly have a positive significant effect on the level of practice use of antibiotics Prevent antibiotic resistance at a pig farm in the city of Kupang.

Abstract

Beef meat has a complete nutritional content, the high prices in the market causing some unscrupulous traders to cheat by adding water to meat to get more weight. This beef meat has a bad quality, so needs the right method to identify the quality of beef meat in the field.  This study was designed to identify the addition of water to beef meat by measuring the water holding capacity (filter paper press method) and electrical conductivity (EC meter). The experimental used one hundred bovine longisimus dorsi and the samples were divided into five groups. One group as a control and four other groups were treatment group. The treatment groups were injected by water with a volume of 1%, 2%, 3%, and 4% of the sample weight and all groups were repeated for twenty times. The results were analyzed by ANOVA and Duncan Multiple Range test. The data showed that the addition of water by injection in beef meat of treatment group 2%, 3%, and 4% had a significant influence on water holding capacity and electrical conductivity (p<0.05). Pearson correlation results showed a strong to very strong correlation with value in control group (r=0.704; p<0.01), treatment group 1% (r=0.628; p<0.01), 2% (r=0.700; p<0.01), 3% (r=0.642; p<0.01) and 4% (r=0.760; p<0.01). Based on the result of the study, it is known that a water increase in beef at least 2% of the weight significantly affected water holding capacity and it can be identified by using electrical conductivity meter.

Abstract

Research of interest marine algae and seagrasses have been renewed, because they are considered to be promising resources of functional ingredients in the development of novel products. Fucoidans are a class of fucose-rich sulfated carbohydrates found in brown marine algae and echinoderms, and more recently identified in seagrasses. Chemical compositions of most fucoidans are complex, mainly being composed of fucose and sulfate. They also contain other monosaccharides like mannose, galactose, glucose, xylose and uronic acids, even acetyl groups and protein and it’s have potential therapeutic properties, including anti-viral activity. Currently, fucoidans are available for use in therapeutic and dietary supplements for inclusion in pet, livestock and aquaculture feed supplements. This review summarizes experimental evidence indicating that Fucoidan can improve the quality of feed ingredients for livestock, pets and aquaculture to improve health.

Abstract

Reticulated python (Malayopython reticulatus) java locality is highly demand as pet animal. Respiratory and pulse rate were important physiological data needed for diagnostic references. The aim of this research was to study the repiratory and pulse rate on Java M. reticulatus. Thirthy healthy adult Java M. reticulatus were used in this study. The respiratory rate recorded by counting inspiratory and expiratory movement of the thorax for one minute and pulse rate recorded by heart palpation for one minute, measured at 6-8 AM and at 4-6 PM for five consecutive days. Temperature and humidity were record as secondary data. The data were analyzed using independent t-Test. The respiratory rate in this study recorded in the morning ranged 5-48 breaths/minute averaging 20 breaths/minute and in the evening ranged 7-49 breaths/minute averaging 24 breaths/minute. The pulse rate in the morning ranged 21-68 beats per minute (bpm) averaging 45.5 bpm and in the evening ranged 40-82 bpm averaging 56 bpm. Totally, M. reticulatus having the respiratory rate 5-49 breaths/minute and pulse rate 21-82 bpm. The morning temperature ranged 25-27oC and the evening 26-27.5oC. The range of morning and evening air humidity is 66-75% and 65-70% respectively. The results showed that the comparison of respiratory rate (R) and pulse rate (P) were in normal range namely 1:2.25 in the morning and 1:2.33 in the evening. There were no significant differences (P>0.05) between morning and evening respiratory rate and no significant effect (P>0.05) of temperature and humidity on respiratory and pulse rate. It could be concluded that healthy adult Java M. reticulatus has a respiratory rate 5-49 breaths/minute dan pulse rate 21-82 bpm.

Abstract

Klebsiella spp. is a common bacteria causing mastitis. Klebsiella pneumoniae and Klebsiella varicola is the most infected Klebsiella spp in the dairy farm. This study used 16 milk samples from 8 lactation goats in Sahabat Ternak farm, Sleman, Yogyakarta. Samples were tested by california mastitis test (CMT) reagent to determine the mastitis status, inoculated on blood agar (BA) then on Mac Conkey Agar (MCA) as a selective media for Gram negative bacteria. The colonies from BA and MCA were stained by Gram staining to determine Gram-positive or Gram-negative bacteria and its cell morphology. The colonies from MCA were identified by biochemical tests such as sugar fermentation tests (glucose, lactose, maltose, and saccharose), arginine, ornithine decarboxylase, indole, citrate, urea hydrolysis (Christensen’s method), lysin and malonate. The result of mastitis test showed 5 goats were positive result and 3 samples were negative. Bacterial staining showed 2 samples were Gram-negative, rods and the others were Gram-positive, coccus. The samples with rods shapes were continued by biochemical tests. The characterization result of biochemical test indicated that the rods shapes bacteria were Klebsiella pneumoniae. These bacteria identification conducted in Laboratorium Preklinis Program Studi Kesehatan Hewan, Sekolah Vokasi Universitas Gadjah Mada and the result confirmation using positive control of  Klebsiella pneumoniae conducted in Balai Laboratorium Kesehatan Yogyakarta. These results showed that milk from Etawah crossbreed goat was infected by Klebsiella pneumoniae.

Abstract

Digestive dysfunction is a common symptom of diabetes. Diabetes can lead to the loss of enteric neurons as well as nerve dysfunction and gastrointestinal complications. There has been no complete report of mienteric nitrergic of small intestines on hyperglycaemia condition, so the purpose of this study was to assess changes of nitrergic neural profile of small intestines on hyperglycemic Wistar strain rats (Rattus norvegicus). Twelve rats were adapted for 3 days, then they were randomly divided into 3 groups namely group I (K-1, as control), group II (K-2), and group III (K-3). Before treatment all rats were fasted for 12 hours, rats K-1 were induced with 1.0 ml of buffer citrate single peritoneal dose; rats K-2 were induced with 40 mg streptozotocin (STZ)/ kg body weight of single peritoneal dose; and rats K-3 were induced with 45 mg STZ /kg body weight of single peritoneal dose. On the 1st and 5th days after STZ induction, blood was collected through the coccigea vein to measure blood glucose levels and weigh the weight. On the day 5th the rats were uthanated with a high dose of ketamine, dissected and taken small intestines (duodenum, jejunum, and ileum) for staining with Nicotinamide Adenine Dinucleotide Phosphate-diaphorase (NADPH-d). Morphometric changes of nitrergic neurons of the small intestine segments were observed. The results showed that on the 5th day increased blood glucose levels of the K-2 and K-3 groups were significantly higher than K-1 (P <0.01). The weight of three groups did not show any significant improvement difference (P> 0.05). The number of neurons in the duodenum, jejunum and ileum of K-2 group increased higher than that of K-1 and K-3 groups. These results indicate that in hyperglycaemic states, the number of nitrergic neurons increases (K-2 group), and there is a significant decrease in the number of neurons (group K-3) compared with K-1 and K-2 groups (P <0.01). The results of this study concluded, that the STZ induction treatment causes hyperglycemia. Hyperglycemia does not affect to the body weight and the number of  nitrergic neurons of small intestines in Wistar strain rats (Rattus norvegicus).

Abstract

The rabies outbreaks of Indonesia in these two decades tends to spread faster to other islands/regions. Trade trafficking and people's habits of carrying dogs between islands are contributing factor that triggers rabies cases in Provinces previously free of rabies. This study aims to qualitative risk assessment entry and spread of rabies in dogs into Sorong City of West Papua Province. Methods of collecting primary data obtained from expert opinions, questionnaires, interviews, and direct observation in the field. Secondary data is taken through search of scientific publications, surveillance results, unpublished data in the form of reports, and documents from authorized agencies. The results of study showed that release assessment was high, the incidence of rabies in dogs was 52%, and 180 cases of rabies in humans (lyssa) in Sulawesi, Maluku and North Maluku. The exposure assessment was high based on the presence of rabies transmite animal (RTA) traffic from endemic areas, 58% dogs, 38% cats, and 4% apes from Java 68% (Surabaya 50%, Jakarta 18%), Sulawesi 10% (Manado and Bitung), Maluku 14% (Ambon) and North Maluku 8% (Ternate). The consequence assessment is high because there is a single impact that is categorized as nationally significant. The estimated risk of getting into rabies is high. The potential pathways used in RTA traffic to Sorong City is by sea at 89.3% and air at 10.7%. The results of the study concluded that the qualitative risk assessment of entry and spread of rabies in dogs to the Sorong City of West Papua Province was high. All risk assessments have low uncertainty.
