Abstract

Adhesi merupakan tahap inisiasi dari proses kolonisasi bakteri. Kemampuan adhesi ini diperantarai oleh keberadaan hemaglutinin pada permukaan bakteri. Tujuan penelitian ini adalah melihat kemampuan hemaglutinin pada Escherichia coli dalam proses adhesi. Uji adhesi pada sel epitel pipi dilakukan dengan menggunakan metode Velentine-Weiggand. Hasil penelitian menunjukkan bahwa Escherichia coli yang memiliki hemaglutinin menunjukkan kemampuan adhesi yang jauh lebih baik yakni rata-rata 67,6 bakteri/sel dibandingkan dengan   yang tidak memiliki hemaglutinin rata-rata 7,2 bakteri/sel. Kemampuan adhesi ini berkaitan dengan keberadaan hemaglutinin di permukaan bakteri.

Abstract

This study was conducted with aim to tend out morphology of oesophagus and stomach of the cave swiflets (Collocalia linchi) at macroscopic and microscopic levels.  The data revealed that esophagus was 2.93 cm in length and possessed no crop.  The stomach was small and the isthmus was not clear. Mucosa of the esophagus was lined by stratified squoamus epithelium. Esophageal gland was mucous type. The glands were well developed and distributed along the esophagus. The external muscle layer consisted of inner circular and outer longitudinal layers.  The stomach could be distinguished into proventriculus and ventriculus with no clear isthmus between them. The mucosa of proventriculus was lined by single columnar epithelium. The ventricular gland area was divided into cardiac, fundic  and pyloric gland areas. The surface of ventriculus was lined with cuticula. In general the esophagus and stomach of the cave swiftlets were simple with no crop in the esophagus and isthmus in the stomach. These might be related with the kind of food and feeding behavior of this species.

Abstract

Penelitian ini dilakukan untuk mengetahui pengaruh lama maturasi dan lama inkubasi fertilisasi terhadap angka fertilitas oosit sapi peranakan Ongole (PO) secara in vitro. Oosit diaspirasi dari folikel ovarium yang berdiameter 2-6 mm. Oosit dirandom untuk penelitian menggunakan rancangan blok acak sempurna (RCBD), dengan lama maturasi 20, 22 dan 24 jam sebagai blok dan lama inkubasi fertilisasi 4, 5 dan 6 jam sebagai perlakuan. Hasil penelitian menunjukkan angka fertilitas oosit sapi PO dengan lama maturasi 20 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 48,86%+3,88, 48,08%+2,22 dan 51,96%+ 2,27. Angka fertilitas oosit sapi PO dengan lama maturasi 22 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masing-masing adalah 50,14%+4,18, 51,14%+4,35 dan 52,27%+2,26. Angka fertilitas oosit sapi PO dengan lama maturasi 24 jam dan lama inkubasi fertilisasi 4, 5 dan 6 jam masingmasing adalah 51,21%+4,49, 53,49%+2,43 dan 50,96%+4,18. Tidak ada perbedaan yang nyata (P0,05) angka fertilitas antara lama maturasi 20, 22 dan 24 jam dan tidak ada perbedaan yang nyata (P0,05)  angka fertilitas antara lama inkubasi fertilisasi 4, 5 dan 6 jam.

Abstract

The propagation of E. mivati local strain was used single cell inoculation.  The objective of this study was to determine the pathogenicity of Eimeria mivati inoculation in broiler chicken by measuring the growth rate, oocysts production and levels of changes in small intestine.  The chickens were devided into two age groups of infection, three and teen days old.  Each group was received 1x104, 1x105   and 1x106  oocysts/chick.  The chichen were raised in wire cages and sampled accordingly.  Data collected was analyzed using analysis of variance. A large number of oocysts production, intestinal villi lesions and depression of growth rate were seen after infection.  Most of the peak production of oocysts were accured after the 9th day of infection.  However, after the peak production of oocysts,  oocysts production decreased extremely.  The choice of parasitic development were in the surface epithelial cells of the villi and the region dominantly effected was the third anterior of small intestine. The result revealed no significant differences regarding photogenicity findings (P0.05) between chicken aged three and ten days after infected with E. mivati.

Abstract

Penelitian ini bertujuan menentukan konsentrasi dan berat molekul protein  ekskretori/sekretori larva (L3) Ascaridia galli (A. galli). Larva L3 diperoleh dari usus halus 100  ayam tujuh hari setelah pemberian dosis 6000 L2 melalui esofagus ayam. Sebanyak 5–10  L3 dikultur secara in vitro  dalam setiap ml medium Rosswell Park Memorial Institute (RPMI 1640), pH 6,8, tanpa merah fenol dalam inkubator pada temperatur 37 0C dan 5% CO2 selama 3 hari. Ke dalam medium ditambahkan 100 unit ml-1 penisilin G, 100 µg ml-1 streptomisin, 5 µg ml-1 gentamisin dan 0,25 µg ml-1 kanamisin. Ekskretori/sekretori dipreparasi dari produk metabolisme L3 yang dilepaskan ke dalam medium kultur. Untuk mendapatkan protein ekskretori/sekretori, medium kultur dipekatkan dengan vivaspin 30.000 MWCO, dan kuantitas protein dihitung dengan metode Bradford. Berat molekul protein ekskretori/sekretori divisualisasikan dengan sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS PAGE). Hasil penelitian  menunjukkan bahwa konsentrasi protein ekskretori/sekretori adalah 0,595 mg/ml dengan berat molekul 28 kDa.

Abstract

Increasing in ambient temperature inside the cage could lead to heat stress in broilers. This research was conducted to find out effects of heat stress on body weight gain, heterophile-lymphocite ratio and body temperature in chicken broiler. Twenty broilers aged 20 days (strain Cobb) were randomly divided into 2 groups. The first group was treated with no heat stress, the second one was caged in 33±1 0C temperature for 4 hours per day for 14 days. The results indicated that heat stress reduced body weight gain, increased body temperature, and changed behavior, but no effect on feed conversion ratio (FCR) and heterophile-lymphocyte ratio. It suggested that the heat stress caused detrimental effects on broiler chicken.
