Abstract

Isolation, Identificaton, and Charaterization of Blast Fungus Pyricularia oryzae Derived from Rejuvenation. Puji Lestari, Wawan, Tri P. Priyatno, Wening Enggarini, Reflinur, and Yadi Suryadi. Blast disease caused by Pyricularia oryzae (Po) is one of the important diseases on rice crop in Indonesia. This study was aimed at obtaining blast fungus pathogen rejuvenated from preserved seed origin, agar medium and rice leaf tissue. In addition, we identified blast isolates for long-term storage collection, and characterized isolates towards their specific locus of genes virulence. The results revealed that 22.7% isolates stored in agar media produced a typical mycelium of Po. The specificity of total genomic DNA banding pattern of isolates which were analyzed using specific primers encoding virulence genes of Cut1, Erg2, and Pwl2, showed six haplotypes consist of B-001 (1 isolate), C-011 (1 isolate), D-111 (8 isolate), F-110 (1 isolate), G-100 (3 isolate), and H-101 (2 isolate). None of haplotypes A-000 and E-010 were found among the isolates studied. In regard to its pathogenicity, the majority of Po fungal isolates had Cut1, Pwl2, and Erg2 genes. Among the total isolates of the Po fungus, the greatest proportion of genes were Pwl2 (87.5%) followed by Cut1 (75%) and Erg2 (62.4%) genes. AbstrakPenyakit blas yang disebabkan oleh cendawan Pyricularia oryzae (Po) merupakan salah satu penyakit penting pada pertanaman padi di Indonesia. Penelitian ini bertujuan untuk mendapatkan cendawan patogen blas yang berasal dari hasil rejuvenasi simpanan benih, media agar, dan jaringan daun padi. Selain itu, identifikasi koleksi isolat blas yang ditujukan untuk penyimpanan jangka panjang dan karakterisasi kespesifikan tiap isolat terhadap lokus spesifik gen virulensi. Hasil pengujian menunjukkan 22,7% isolat yang tersimpan dalam media agar masih menunjukkan tipikal miselium Po. Kespesifikan pola pita DNA genomik total isolat yang dianalisis berdasarkan primer penyandi gen spesifik virulensi Cut1, Erg2, dan Pwl2, diperoleh sebanyak enam haplotipe meliputi B-001 (1 isolat), C-011 (1 isolat), D-111 (8 isolat), F-110 (1 isolat), G-100 (3 isolat), dan H-101 (2 isolat). Tidak ditemukan haplotipe A-000 dan E-010 pada isolat Po yang dianalisis. Dalam hubungannnya dengan patogenisitas, mayoritas cendawan Po mempunyai gen Cut1, Pwl2, dan Erg2. Di antara total isolat cendawan Po, gen yang paling besar ditemukan proporsinya adalah gen Pwl2 (87,5%) diikuti Cut1 (75%) dan Erg2 (62,4%).

Abstract

Utilization of Forest Products by the Society at the Time Have Been Amended Due to Changes in Tastes and Consumption Levels of Society and Increase Social Dynamics of Society. Slamet E. Sumanto and Mariana Takandjandji. This study aims to determine patterns of utilization of forest products by communities and the pattern changes and the influence on forest management. Research conducted in Timor Tengah Selatan regency of East Nusa Tenggara. Methods of data collection using direct interview techniques, field documentation and library literature. The results showed that the general public has long recognized and use forest products for housing construction, energy and wood fences, craft materials and traditional musical instruments, a dye, and the use of other (honey bees, forest products followup likes as tamarin and walnut, as well as handicrafts). Local knowledge and utilization of local plant species should be a consideration in forest management policy in the future. AbstrakPemanfaatan hasil hutan oleh masyarakat pada saat ini telah mengalami perubahan karena perubahan selera dan tingkat konsumsi masyarakat serta peningkatan dinamika sosial masyarakat. Penelitian ini bertujuan untuk mengetahui pola pemanfaatan hasil hutan oleh masyarakat dan pola perubahannya serta pengaruhnya terhadap pengelolaan hutan. Penelitian dilakukan di Kabupaten Timor Tengah Selatan, Provinsi Nusa Tenggara Timur. Pengumpulan data menggunakan teknik wawancara langsung, dokumentasi lapang serta beberapa literatur. Hasil penelitian menunjukan bahwa secara umum masyarakat telah lama mengenal dan memanfaatkan hasil hutan untuk keperluan pembangunan rumah, kayu energi dan pagar, bahan kerajinan dan alat musik tradisional, bahan pewarna kain, serta pemanfaatan lainnya (lebah madu, hasil hutan ikutan asam dan kemiri, kerajinan tangan). Pengetahuan lokal dan pemanfaatan tumbuhan lokal seyogianya dapat menjadi pertimbangan kebijakan pengelolaan hutan di masa depan.

Abstract

Plant Diversity in the Mount Celering Nature Reserve. Titi Kalima and Nur M. Heriyanto. Study of the plant species diversity in the Nature Reserve of Mount Celering (CAGC) Jepara, Central Java, was carried out from April to May 2008, with aimed to get information about plant diversity after natural disasters and encroachment. The method used was the checkered path with length of 500 m and width of 20 m, made of three lines cut slopes. The research was found 32 species, 29 genera, and 21 families. Tree level was dominated by Hibiscus macrophyllus Roxb. (IVI = 26.75%), Artocarpus elasticus Blume (IVI = 26.53%), and Alseodaphne umbelliflora Blume. (IVI = 22.75%). Saplings by Syzygium acuminatissimum (Blume) A.DC. (IVI = 23.86%), Dipterocarpus hasseltii Blume (IVI = 18.71%), and Artocarpus elasticus Blume (IVI = 18.52%). Six species of endangered trees were Alstonia scholaris (L.) R.Br., Alstonia angustifolia Wall, D. hasseltii Blume, Parkia javanica (Lamk.) Merr., Stelechocarpus burahol Hk.f.et Th., Sterculia cordata Blume. AbstrakPenelitian keragaman jenis tumbuhan di Cagar Alam Gunung Celering (CAGC) Jepara, Jawa Tengah, dilakukan bulan April sampai dengan Mei 2008 dengan tujuan untuk mendapatkan informasi tentang keragaman jenis tumbuhan pasca bencana alam dan perambahan. Metode yang digunakan adalah jalur berpetak dengan panjang 500 m dan lebar 20 m, dibuat tiga jalur memotong lereng. Dari hasil penelitian ditemukan 32 jenis, 29 genera, dan 21 famili. Tingkat pohon didominasi Hibiscus macrophyllus Roxb. (INP = 26,75%), Artocarpus elasticus Blume (INP = 26,53%), dan Alseodaphne umbelliflora Blume. (INP = 22,75%). Anakan pohon oleh Syzygium acuminatissimum (Blume) A.DC. (INP = 23,86%), Dipterocarpus hasseltii Blume (INP = 18,71%), dan Artocarpus elasticus Blume (INP = 18,52%). Enam jenis pohon yang termasuk katagori kritis dan terancam punah yaitu Alstonia scholaris (L.) R.Br., Alstonia angustifolia Wall, D. hasseltii Blume, Parkia javanica (Lamk.) Merr., Stelechocarpus burahol Hk.f.et Th., Sterculia cordata Blume.

Abstract

Evaluation of Growth of Six Commercial Red Mango Varieties Grafted by Top Working Technique on Two Interstock Varieties. Rebin, Lukitariati Sadwiyanti, Djoko Sudarso, and Karsinah. The objective of the research was to evaluate the growth and leaf anthocyanin content of six commercial red mango varieties that grafted by top working technique on two interstock varietties. The extensive development of commercial red mango as new superior variety in production center, especially in Java, faces limited land constraint. Meanwhile, the existing mango plants in growers is dominated by Arumanis and Durih varieties that are now old and less productive. In addition there are many local mango plant that are not good quality in their growth. In the condition mentioned, replacing less productive mango variety with commercial red mango through top working technology is a wise solution alternative. Research was conducted at Cukurgondang Experimental Field 50 m asl, Pasuruan, East Java from May until December 2009. The research was arranged in Split Plot Design where the main plot were two interstock varieties i.e.: Arumanis-143 and Durih, meanwhile the sub plot were six commercial red mango scions i.e.: Ken Layung, Marifta-01, Garifta Orange, Garifta Merah; Garifta Kuning and Garifta Gading. Each tratment were repeated three times. The results indicated that there were five varieties of red mango varieties compatible with Arumanis-143 interstock i.e.: Marifta-01, Garifta Orange, Garifta Merah, Garifta Kuning, and Garifta Gading. While Durih interstock was suitable for Ken Layung, Garifta Orange and Garifta Merah. Interstocks were not influence on leaf color of scion (both young and mature leaves). Anthocyanin content of Garifta Orange, Garifta Merah, and Garifta Gading leaves on Arumanis-143 interstock and on Durih interstock were higher than their mother plant respectively. The effect of Arumanis-143 was beter than Durih interstock on both vegetative character and leaf antocianin content of scion. AbstrakTujuan penelitian ini ialah mengevaluasi pertumbuhan dan kadar antosianin daun enam varietas mangga merah komersial yang disambung melalui teknik top working pada dua varietas batang antara. Pengembangan mangga merah komersial sebagai varietas unggul baru secara ekstensifikasi di sentra produksi, terutama di Jawa, mengalami kendala terbatasnya lahan. Sementara itu, pertanaman mangga yang ada di petani didominasi oleh varietas Arumanis dan Durih yang tanamannya sudah tua dan kurang produktif. Di samping itu banyak juga tumbuh mangga lokal yang kurang berkualitas. Dalam kondisi yang demikian, penggantian varietas mangga yang kurang produktif dengan mangga merah komersial melalui teknologi top working merupakan sebuah alternatif solusi yang bijaksana. Penelitian dilaksanakan di KP. Cukurgondang (Pasuruan) 50 m dpl, mulai Mei-Desember 2009. Perlakuan disusun dalam Rancangan Petak Terpisah (Split Plot Design) dengan Petak Utama berupa dua varietas batang antara, yaitu Arumanis-143 dan Durih. Sedangkan Anak Petak berupa enam varietas batang atas mangga merah komersial, yaitu Ken Layung, Marifta-01, Garifta Orange, Garifta Merah, Garifta Kuning, dan Garifta Gading. Setiap perlakuan diulang sebanyak tiga kali. Hasil penelitian menunjukkan bahwa lima varietas batang atas yaitu Marifta-01, Garifta Orange, Garifta Merah, Garifta Kuning, dan Garifta Gading kompatibel pada batang antara Arumanis-143. Sementara itu, batang antara Durih sesuai untuk batang atas Ken Layung, Garifta Orange, dan Garifta Merah. Batang antara tidak berpengaruh terhadap warna daun (baik daun muda maupun daun tua). Kadar antosianin daun pada varietas Garifta Orange, Garifta Merah, dan Garifta Gading yang di top working pada batang antara Arumanis-143 dan Durih lebih tinggi dibanding dengan kadar antosanin daun dari pohon induknya. Pengaruh batang antara Arumanis 143 lebih baik dibanding batang antara Durih terhadap sifat vegetatif maupun kandungan antosianin daun batang atas.

Abstract

Estimation of Genetic Parameter on Days to Harvest and Weight Per Fruit Characters of Hybrization Chilli and Cayyenne Papper (Capsicum annuum L.). Abdullah B. Arif, Linda Oktaviana, Sriani Sujiprihati, and Muhamad Syukur. Selection method is one of the most important factors in determining the success of pepper breeding programs. Selection method will be effective if it is supported by a complete knowledge of genetic character inheritance. The objective of this study was to aim information of inheritance pattern of pepper adaptability of quantitative characters. There are two steps in this experiment i.e developing material genetic and inheritance study of days to harvest and weight per fruit characters in the field. The results showed that days to harvest and weight per fruit characters are not maternal effect. The m[d][h][i][l] genetic model is suitable for days to harvest. The m[d][h][i][j] genetic model is suitable for fruit weight. Broadsense and narrow-sense heritabilities range from low to high. AbstrakMetode seleksi merupakan faktor penting yang mempengaruhi kesuksesan program pemuliaan cabai. Metode seleksi akan lebih efektif jika didukung oleh sebuah pengetahuan yang lengkap tentang pola pewarisan karakter genetik. Penelitian ini bertujuan mendapatkan informasi pola pewarisan karakter kuantitatif. Terdapat dua tahapan pada penelitian ini, yaitu pembentukan materi genetik dan studi pewarisan karakter umur panen dan bobot per buah di lapang. Hasil penelitian menunjukkan tidak terdapat pengaruh tetua betina terhadap karakter umur panen dan bobot per buah. Model genetik m[d][h][i][l] sesuai untuk karakter umur panen. Model genetik m[d][h][i][j] sesuai karakter bobot per buah. Heritabilitas dalam arti luas dan dalam arti sempit berada dalam kisaran rendah sampai tinggi.
