Abstract

One of the problems related to the establishment of mangosteen plantation is to obtain seedlings throughout the year, which can be solved by micropropagation.   The propagation of Mangosteen was done through somatic embryogenesis. The objective of this study was to determine the effect of Thidiazuron concentration in ½ MS medium to achieve somatic embryo stages of mangosteen seed. The study consisted of two experiments. Experiment 1: The embryogenic callus c.a.1 g derived from MS solid medium containing of Thidiazuron (0,1; 0,5; dan 1 mg/l) and 2,4-Dichlorophenoxy acetic acid (4,6,8 dan 10 mg/l) was subcultured into ½ MS0 liquid medium. 2). The embryogenic callus from the previous medium were subcultured into ½ MS liquid medium containing Thidiazuron (0, 1, 2, 4 dan 8 mg/L) and Casein hydrolysate 500 mg/L. The results showed that cell suspension were developed after mangosteen embryogenic callus subcultured into ½ MS0 liquid medium.  Somatic embryo stages such as globular, heart and torpedo were formed after the calli derived from the previous ½ MS solid medium containing 10 mg/L 2,4-D and 1 mg/L Thidiazuron were subcultured into ½ MS0 liquid medium in 8 weeks.  Cell suspension in ½ MS liquid medium with or without the addition of  Thidiazuron and Casein hydrolysate has induced the formation of embryo somatic stages such as globular, heart and torpedo after 6 months of incubation.

Abstract

Yield of shallot bulbs in Banten still low because famers do not use organic fertilizers. This study aims to study the effectiveness of organic fertilizers combined with inorganic fertilizer on the growth and yield of shallot. This study was conducted in farmers’ fields in Sawahluhur village, district Kasemen, Serang. Studies used a randomized complete block design consisting of 5 treatments and 4 replications. The varieties used are Bima. The results showed that the use of organic fertilizers in combination with inorganic fertilizers provide a good influence on the growth and yield of shallots. The use of organic artificial fertilizer 5.000 kg//ha + Urea 100 kg/ha+ SP-36 200 kg/ha + 300 kg/ha NPK Phonska resulted in higher yield of tubers and profits than without organic fertilizer on shallots farming.

Abstract

Rhizobacteri Merapi isolates have been detected, and MA, MB, and MD isolates could withstand osmotic stress up to >2.75 M NaCl. MD isolate was stronger in dissolving phospate than MA and MB isolates, but MA and MB isolates had much stronger nitrification capability and could perform ammonification than MD isolate. This research studied about Rhizobacteri Merapi isolates as identification, as well as their characterization. The results show that there is a difference in colonial characteristic of four Rhizobacteri Merapi isolates where MB and MC isolates (white circular-entire shape), except for MA isolate (curled-white with undulate edge) and MC isolate (ramose-yellow with filamentous edge) and also that MD isolate have the largest diameter (1,5 mm). Cell characteristics, however, were the same, gram-negative and were rod shaped, except for MD isolate (coccus). All showed aerob and fermentative characteristics, although MD isolate was able to very strongly hydrolyze starchs. The growth type is fast growing which reach log phase for after 48 hours and then the number of colony decrease.

Abstract

Bondowoso tobacco is a type of  sliced tobacco that is developed in eleven district i.e. Bondowoso, Tenggarang, Tegal Ampel, Pakem, Curahdami, Maesan, Pujer, Wonosari, Binakal, Taman Krocok and Wringin. In order to fulfill the improvement of new variety program, yield evaluations is a step that should be done. The aims of the experiment were to examine the yield of local varieties, the superior traits and then choose the best variety. The experiment was conducted on March to September 2009 in tobacco producing areas. The experiment tested 6 local  varieties i.e. Sam1, Se2, Mar3, Sam4, Sam5 and Sam6. Each of them was plant with three replicates. The experiment design was  randomized complete block design (RCBD). Each plot consisted of 200 plants. The result showed that three varieties of which relatively had the same superiority on characters: yield potensial, quality index, plant index, nicotine content and disease tolerance. The selected varieties were Sam1, Sam5 dan Sam6, with yield potensial each acession i.e: 1.682 kg/ha, 1.399 kg/ha and 1.282 kg/ ha.

Abstract

In Sumedang, local variety of groundnut DM Situraja has a great potential and showed increasing production rates. This support the agribusiness development of groundnut in Sumedang District and surrounding areas. The objective of this experiment was to determine production potential of local variety groundnut Situraja especially in Sumedang. This research was arranged in a randomized complete block design whereas each treatment was replicated 4 times. The data were analyzed using SPSS version 16 and treatment means were separated using LSD (Least Significant Difference) with 5% error level. The results indicated that morphology of Situraja variety had an upright growth habit, rod-shaped and green purplefish, 2-seeded, and seed weight 40-55 g for 100 seeds, maturing age of 90 days, and the production potential 3,02 t/ha. The content of oil was 45%, with a relatively high protein content of 28% and crunchy sweet taste. High productivity of DM Situraja local variety was resulted by interaction between variety and spesific enviroment.

Abstract

Problems crop cultivation on rain fed land after rice is still too wet for crops, while for rice crop will experience drought on the generative phase. The technology was required to decrease the moisture content of the soil so that crops can be planted after rice. The study aims to examine the application of the use of the underground drainage system to lower the water logging. Local raw materials were used in order to easily adopted by farmers, because common uses of the pipe is still too expensive. The drainage material was made of a collection of coconut husk and wood twigs. As a test medium performed using texture medium sandy clay loam soil and sand. The test results showed that the ability of the drainage flow on coconut fiber, lower than that of wood sticks consecutive 0.37 and 0.48 liters / sec. And the maximum flow capability was shown in the sandy soil of 0.75 and 1.93 liters / sec. Condition of the land with drainage modulus 10mm / day and the structure of materials was used in the field of wood sticks with inter-channel spacing is 10 m, then there is a 10 in 1 ha pipeline, so the ability to discharge to 69 m3 / h. Therefore, it took time for water discharge at 100/69 = 1.45 This means that the potential of using systematically drainage disposal is fit for use primarily in the light texture such as sandy clay loam.

Abstract

A laboratory experiment was conducted to determine the organic matter in reducing Nitrate leaching of coastal sandy soil of Kulon Progo, Yogyakarta, done from February up to August 2014. This experiment was arranged in a factorial completely randomized design. The first factor were ratio of cow dung and rice straw (1:1; 1:2 and 1:3), and the second factor were N fertilizer (urea and ZA). The result showed (a) Urea as source of N-fertilizer was more suitable to apply in coastal sandy soil and (b) Organic matter with ratio of cow dung and rice straw (1:1) was able to decrease nitrate leaching of coastal sandy soil of Kulon Progo.

Abstract

Saponin is one of the bioactive compounds present in plants. Saponin has the benefit of human health. Indonesian has been used binahong (Anredera cordifolia (Ten) Steenis) to medicine. However, empirical evidence regarding the content of saponin in binahong not been revealed. This study aims to determine the qualitative and quantitative content of saponins in plants binahong. The results showed that base on Tin Layer Chromatography (TLC) analysis, saponin content in the leaves, stems and tubers binahong both young and old were positive. Quantification results the young leaves, old leaves, young stems, old stems, young tubers and old tubers: 1.37ìg/mg, 2.36ìg/mg, 1.89ìg/mg, 1.2ìg/mg, 1.55ìg/mg and 1.35ìg/mg respectively.
