Abstract

Soft rot bacteria infection in carrot tuber (D. carota L.) causes severe economic losses. Soft rot disease can be caused by various bacteria belonging to Enterobacteriaceae. This study aimed to isolate and identify bacteria as causal agent of soft rot disease in local carrot variety in Bali. Samples were collected at Badung Tradisional Market, Denpasar, Bali. Isolation was carried out by serial dilution method (Platting Method). Eight bacteria (BL1, BL2, BL3, BL4, BL5, BL6, BL7 and BL8) were isolated from soft rot tuber. BL6 isolate showed positive result in Postulat Koch test that caused soft rot on carrot tuber. The result of identification by Microgen™ GnA+B-ID System and identification book Bergeys’s Manual of Determinative Bacteriology Ninth Edition (Holt et al., 1994), BL6 was identified as Citrobacter.

Abstract

A one Ha (100 x 100 m) of Permanent Sampling Plot with 25 subplots (20 x 20 m with 2 x 2 m nested plot) and 5 level rows of altitudinal difference has been established to determine groundcover species stucture and composition in Pohen Mountain, Batukahu Nature Reserve, Bali. Enumeration of all groundcover species revealed that there were 69 species and 47 families with selaginaceae was the most abundant family in the 1 Ha Permanent Sampling Plot. Shannon Index revealed that groundcover species composition in the first, second and third rows were similar, but different with groundcover species composition in the fourth and fifth rows. Cluster Analysis concluded that groundcover species composition were varied. Generally, most of the groundcover species revealed clumped distribution and only a few species were solitaire.

Abstract

In vitro propagation study was carried out at Plant Tissue Culture Laboratory, Faculty of Agriculture, Udayana University. It was conducted using Completely Randomized Design with two factors.  The first factor was plant growth regulator (PGR), consist of nine concentration mixture of IBA (0, 0.5, 1 mg/L) and BAP (0, 1, 2 mg/L). The second factor was grape varieties, consisted of Prabu Bestari and Jestro Ag 86.  There were five replicates for each treatment combination. Variable observed included presence and absence of callus, percentage of callus growth, percentage of shoot growth, number of shoot per explants, emergence of shoots, percentage of root growth and number of roots per explants.  Data were statistically analyzed using Analysis of Variance (ANOVA).  When treatment showed a significant different (P<0.05) or highly significant (P<0.01) mean separation was conducted following Tukey Test. Data were analyzed using MINITAB Statistical Program. Results showed that PGR and varieties was highly significant for percent of callus growth.  PGR also showed significant effect on shoot growth percentage, while there was significant difference found on root growth percentage.  It can be concluded that combination of 1 mg/L IBA and 2 mg/L BAP was able to produce the highest percentage of callus growth. Addition of 2 mg/L BAP without IBA was able to produce the highest percentage of shoot growth. Percentage of root growth of Jestro Ag 86 was higher than Prabu Bestari.

Abstract

The main objectives of this research were to isolate and identify antagonists of Xanthomonas campestris from rhizosphere zone of broccoli plants. Soil samples were collected from broccoli farm located at Kembang Merta village, Tabanan, Bali. Isolation and identification of the antagonists were conducted at the Laboratory of Microbiology, Udayana University. Two fungal (Trichoderma harzianum and  Trichoderma viride) and two bacterial (Bacillus sp. and Pseudomonas sp.) antagonists potentially to be developed as biocontrol agents of Xanthomonas campestris were successfully identified in this research.

Abstract

Dendrobium heterocarpum Lindl. is one of the orchid species which is often seeked. It is because this species has a mystique appearance and special fragrance. The research aimed to observe the growth and developing responses of protocorm like bodies (PLBs) of D. heterocarpum after 12 weeks in three different medium, i.e. Murashige and Skoog (MS), Kursor C and Western 3 (W3). The D. heterocarpum seeds were planted in MS, Kursor C and W3 medium. Qualitatively the orchid growth were observe including growth score and colour score. The result showed that growing time of MS was 4 weeks, a week faster than the growing time on Kursor C and W3 medium. The growing phase on MS medium and Kursor C medium reached phase 1 whereas on W3 medium reached phase 2. PLB’s colour after 12 weeks on MS medium reached phase E, strong yellowish green phase (142A). Meanwhile, on Kursor C and W3 was on phase D, moderate yellowish green (143D).

Abstract

Soft rot on strawberry fruit was found in strawberry (F. x ananassa) plantation in Candi Kuning, Bedugul, Bali. Soft rot on strawberry fruit can be caused by microorganism i.e. bacteria. Objectives of the research were to isolate pathogen causing soft rot on strawberry fruit with plating method and to identify bacteria causing soft rot by using Kit MicrogenTM GNA+B-ID System and Bergey’s Manual of Determinative Bacteriology reference (Holt et al., 1994). Results showed there were five isolates of bacteria (IB-1, IB-2, IB-3, IB-4, and IB-5). Positive result of Postulat Koch showed that bacteria causing soft rot on strawberry is IB-1. Identification that was done by using Kit MicrogenTM GNA+B-ID System and Bergey’s Manual of Determinative Bacteriology reference (Holt et al., 1994), showed that the isolate IB-1 is Weeksella

Abstract

The purpose of this research was to find out the existence of Vibrio cholerae, bacteria that may cause cholera disease, in some fishery products in Denpasar traditional market, Bali. This research used samples taken from three different fisheries products: tuna fishes (Euthynnus affinis), shrimps (Penaeus indicus), and shellfish (Anodonta sp.). They were taken from three traditional markets in Denpasar City: Ketapian, Kumbasari, and Pidada Markets. All samples were cultured on Alkaline Peptone Water (APW) media, continued by Thiosulfate Citrate Bile Salt Sucrose (TCBS), and then Biochemical Test and Serology Test undertaken. The results of this study showed that two (7.4%) samples taken from Ketapian Market were proved to be positive containing pathogenic bacteria of V. cholerae; they were the shrimps with UA2 code and the shellfish with KA2 code. Meanwhile, there were no V. cholerae contaminations proven to exist in two other kind of products in other two traditional markets.
