Abstract

Human behavior is a part of the ecological system, therefore overcoming the ecological crisis and living resources need to explore human traditions, way of life and human behavior toward natural resources. Understanding and translation of human harmonious relationship with all elements along with other living beings is a form of human ecological intelligence. The objective of this study was to describe the traditions and coastal communities behavior that contains the value of ecological intelligence in coastal biodiversity conservation. The study was based on observation, focus group discussion and identification of Bajau coastal community tradition in their lives and livelihoods on fishing, as well as the behavior toward biological resources. Bajau coastal communities settled in three villages in Pohuwato regency, Gorontalo. There are two themes that contains the value of ecological intelligence of  Bajau communities in the conservation of biodiversity, namely; mamia kadialo in tradition of fishing, and fishing behavior. The prohibition in the tradition of mamia kadialo contains the value of conservation of biodiversity. Usage of simple equipment on fishing activities provide a positive ecological consequences for the conservation of coastal biodiversity.

Abstract

The aim of this study was to optimize the digestion process of SacII restriction enzyme in Mycobacterium tuberculosis H37Rv isolate. SacII enzyme that have CCGC^GG restriction site could be used to detect the mutation at -24 promoter region of inhA M. tuberculosis. The mutation in this position is one of the causes of isoniazid resistance. The methods of this study were conducted in several step, respectively: DNA isolation of M. tuberculosis H37Rv, amplification of inhA promoter region of M. tuberculosis H37Rv using PCR method, and digest optimization using SacII. In this study, optimization of SacII digestion was conducted for formulaion and incubation time. The formulation was optimized by GM-ONE Buffer and GM-Buffer IV, meanwhile the incubation time was optimized in 1 hour, 2 hour, and 3 hour. The result of this study showed that SacII work optimally in GM-Buffer IV compared to GM-ONE Buffer and incubate for 3 hours. Under the optimal digest condition, SacII enzyme could be used to detect the mutation at -24 promoter region of inhA in the case of MDR-TB using PCR-RFLP method.

Abstract

Alstonia scholaris (L.) R. Br. is an indigenous plant species in Indonesia which has been use for multiple purposes. This plant tolerant to poor nutrient and alkaline soil, therefore it is potential to explore as a revegetation plant. The study was conducted to identify effect of IBA concentrations on rooting ability of A. scholaris shoot cutting. Plant materials (shoot cutting) were collected from trubusan in Mandeh Area, Tarusan, West Sumatra. This study used Completely Randomized Design (CRD). The treatment was IBA concentrations (0 ppm, 50 ppm, 100 ppm, 150 ppm and 200 ppm). The result showed that IBA concentrations was significant different (P ? 0,05) for length, fresh weight and dry weight of root. The application of 150 ppm IBA was the best concentration to enhance the rooting ability of A. scholaris shoot cuttings.

Abstract

The purpose of this study was to look at the effect of sugar dose, black tea extract dose, interaction of sugar and black tea exstract dose on the fermentation and production of nata de coco. Completely Randomized Design (CRD) in Factorial was used in this experiment with sixteen treatments and two replications. The first factor was the doses of sugar and the second factor was doses of black tea extract. The results of this study showed that the doses of sugar and addition of black tea extract was able to increase the production of nata de coco. The best dose of sugar in 50 g/ l, the best dose of black tea extract in 8 g/ l. The interaction of sugar dose and black tea extract dose contained in the treatment a1b2 the dose of sugar 50 g/l and black tea extract 8 g/l with an average weight of 23,43 g nata de coco. The benefits of this research is to increase production and quality of nata for the nata de coco’s industry.

Abstract

A behavioral study of Great Argus Pheasants (Argusianus argus Linnaeus, 1766) at their mating ring has been done from October 2015 to January 2016 in The Kalaweit Supayang Conservasion Forest, Solok, West Sumatra. The purpose of this research is to know about activities of Great Argus Pheasant and how their activities done. Field observation in the mating ring was conducted using ten video camera traps for three months at their mating ring. Those cameras were activated for 60 seconds per session by 10 seconds intersession intervals. Behavioral data were tabulated from video records using continuous recording method. There were eight activities of Great Argus Pheasant at their mating ring which divided into individual and pairmate activities. The individual activities of male included clearing, walking, vigilance, feeding, grooming, resting, and calling. While female activities included resting, grooming, and walking. Activity with pairmate was dancing where male performed dancing only in the present of female. Male spent 26 times longer than females appeared at the mating ring. The peak activities at mating ring was in the morning between 7:00 and 8:00 am.

Abstract

This research aimed to inventory the insects in pitcher plant (Nepenthes mirabilis and Nepenthes ampularia) found in the Biological Education and Research Forest of Andalas University, Padang. had been conducted manually from April to May 2015. Fourteen species which are belong to three orders namely Diptera, Hymenoptera and Isoptera were collected, which the most genera being sampled was Hymenoptera (10 species), followes by Diptera (3 species) and Isoptera (1 species).The highest number species collected (7 species) was belong to the family of Formicidae, then followed by Culicidae (3 species), and 1 species each for Vespidae, Ichneumonidae, and Termitidae. Two species which have the high number of individual collected were Tripteroides (Rachionotomyia) aranoides (473 individuals) and Topomyia sp. (216 individuals).

Abstract

Black orchid (Coelogyne pandurata) and Pearl orchid (Coelogyne asperata) are endemic orchid from Kalimantan forest, shape and color suitable as ornamental flower. Many people collected the orchids from the wild for commercial purpose but not much effort on their propagation. To prevent population decreased, propagation and production of new varities via interspesific hybridisation need to be done. An important step required in plant breeding is to obtain cross compatiblity. Cross compatibility of some orchids from Indonesia are not known, including compatibility of Coelogyne pandurata x Coelogyne asperata. Information on compatibility and fertility is very important to produce good quality seed as propagation material. In this research pollination was performed on orchid plants at 2 until 5 day after flowering and pollination was done in the morning. There are three replicates for each pollination.  Pollination was done with C. pandurata (black orchid) as male parent and C. asperata (pearl orchid) as female parent. The results showed that successful pollination was obtained from pollination at 2 days after flowering. Seed capsule was harvested at 3 months after pollination but not yet perfectly ripe. Seed germination employed in vitro method on Vacin & Went (VW) and VW with modification media. Seed from C. pandurata x C. asperata abled to germinate on VW media with addition of 0,1 mg/l auxin and 0.1 mg/l cytokinin and germinated 9 weeks after planting.

Abstract

Production of waste in urban areas reached 0.5 kg/person /day and 80% consisted of organic waste, one of many types of waste mostly generated is household waste. Generally, this household organic waste containing structural compoundsuch us long chains of cellulose.Therefore, the use of microorganism that can produce cellulase is very important to reduce the weight of garbage. Trichoderma harzianumis known as the most potential mold compared to other molds in converting cellulose. Sawdust and bran contain cellulose and hemicellulose that can be used as the main component in the mediafor its growth. The objectives of this study was: To find an effective ratio between sawdust and bran as growth mediafor Trichoderma harzianum, To know the cellulase activity of Trichoderma harzianum. This study has been conducted from April 2015 to Juli 2015 in the Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Andalas. The results of this study concluded that an effective ratio between sawdust and bran as a growth media for Trichoderma harzianumwhich degraded organic waste was 50:50. The highest activity of cellulase in degrading organic waste is 100%.

Abstract

The aim of this research was to investigate the impact of terasi, a traditional Indonesian condiment which is made from salted fermented seafood, on the sensory acceptance of beef consommé. Fifty four panelists compared the acceptance of beef consommé containing rebon terasi to the consommé containing fish terasi. The results showed that upon adjusting the salt content, the consommé containing rebon terasi was more preferred than the consommé containing fish terasi. By combining more strong spices or ingredients the acceptance of consommé might be improved while masking the pungent odor of terasi. In addition, six different types of rebon terasi varying in proximate composition, salt and water activity (aw) were added to beef consommé and were evaluated by 10 trained panelists for their specific intensity of taste/flavor (saltiness, sourness, sweetness, bitterness, umami, fishy and rebon). Together with those samples, other consommés were also compared; however, the salt was also added to adjust with the highest salt content among six types of terasi. The results showed that variations of terasi influenced the sensory characteristics of consommé. The salt content of rebon terasi influenced the sourness, bitterness and saltiness in consommé, whilst the salt content should also reach the certain level to fit their acceptance. The distinct flavor from each terasi did have a profound effect on the sensory acceptability. The knowledge of the flavor characteristics of consommé and the role of terasi when mixed in consommé will help to improve the sensory acceptability and marketability of terasi.

Abstract

Seaweed farming activity is now facing some problems caused by pest fish herbivore and ice-ice disease. To solve those and improve our seaweed quality, seaweed production management requires some ecological technique improvements. The purpose of this study was to determine the functions of seaweed H. durvillaei for improving the production of the seaweed E. spinosum in the coastal area of Geger Beach, Peminge Village, South Kuta subdistrict, Badung regency, Bali. The measured data includes parameter of weight, length and water quality. Data was measured every 10 days for up to 40 days. The best growth result was shown by the model of E. spinosum that were fenced by H. durvillaei of 456 g for E. spinosum. H. durvillaei showed good impact on increasing the production of E. spinosum up to 68.7% compared to the control. Water quality conditions on seaweed culture was good with temperature ranged from 27.7 to 30.1°C, salinity from 30 to 32.3 ppt, DO from 3.9 to 5.5 ppm, pH from 7.9 to 8.4, current 0.1 to 0.2 m/s, and clearness of 100%.
