Abstract

Agricultural activity can’t be separated from synthetic herbicide. Some synthetic herbicide use metsulfuron methyl as active ingredient that works to change chemical materials in weed’s tissues so able to exterminate the tissue or disordering physiological system that needed for survivability of weeds. Because of that effect a question arises whether this compound also can affect non target organisms like periphyton algae. Periphyton is a group of microscopic organisms that grows attached to surface of objects or water plants, not penetrate the substrate, staying or moving on the substrate surfaces. One of main compilers of periphyton community is algae. Periphyton algae in water has significant role to determining primary productivity and as bioindicator of water quality. Some research that has been done shows if some active ingredient in particular concentration significantly affects toward primary productivity of periphyton algae and able to inhibit growth of certain algae species. This research was conducted to find out effects of metsulfuron methyl herbicide on periphyton algae community structure. Parameters measured are concentration of chlorophyll a, density, richness, diversity, evenness, and domination indices also periphyton algae species identification. Research conducted in laboratory scale use samples taken from Rawa Pening Lakes, Semarang District, Central Java. Observation time is 21 days with herbicide exposure at 0, 0.05, 0.1, 0.2, and 0.4 mg/L. Results show that periphyton algae community structure more impacted by exposure period of herbicide actve ingredient metsulphuron methyl than concentration that used in research. Abstract

Lamtoro (Leucaena leucocephala Lam. De Wit) merupakan tanaman yang digunakan sebagai obat dan pakan ternak. Lamtoro mengandung zat aktif dari golongan senyawa streroid, alkaloid, flavonoid, polifenol dan tannin. Zat aktif yang terkandung pada lamtoro bersifat fitoestrogen karena mengandung golongan senyawa steroid, alkaloid dan flavonoid. Tujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak daun lamtoro terhadap kadar hormon estrogen dan ketebalan endometrium tikus (Rattus norvegicus) betina yang telah diovariektomi. Penelitian ini menggunakan Rancangan Acak Lengkap dengan empat perlakuan yaitu K- (kontrol negatif diberi minyak jagung), K+ (kontrol positif diberi estrogen sintetik 2 mg), P1dan P2 diberi diberi ekstrak daun lamtoro sebanyak 200 mg/kg BB dan 250 mg/kg BB). Masing masing perlakuan terdiri dari 6 ulangan sehingga digunakan 24 ekor tikus betina. Ekstrak daun lamtoro diberikan secara oral sebanyak 2 ml/ekor/hari selama 15 hari. Data yang diperoleh dianalisis dengan One Way ANOVA dan dilanjutkan dengan uji Duncan Multiple Range Test (DMRT). Hasil penelitian menunjukkan pemberian ekstrak daun Lamtoro (Leucaena leucocephala Lam. De Wit) dosis 250 mg/kg BB dapat meningkatkan kadar hormon estrogen serum darah dan ketebalan endometrium tikus betina yang diovariektomi. Abstract

Phalaenopsis amabilis is an orchid native of Indonesia that included in the appendix II category and one of the flowers included in the national and international flower trade, so that needs to be used in vitro propagation. The type of media used in the subculture determines the results of plantlet growth. Media used include New Phalaenopsis (NP), Vacint and Went (VW), and Murashige and Skoog (MS). Pepton as supplement added to the media. This study aims to analyze the different media types and peptone concentrations during the subculture. The method used is a plantlet subculture to a media bottle. This study uses a completely randomized design with 4x3 factorial patterns and 3 replications. Data were analyzed with ANOVA at 95% significance. Plantlet P. amabilis was subcultured in MS, VW and NP media with peptone (0; 1; 2; 3 g/L). Growth was observed for 2 months. The parameters observed were the number, length, width, appearance of leaves; number, length, appearance of the root. ANOVA test showed that the two treatments did not have interactions with all parameters, except leaf length. The results showed that the treatment of media type and peptone concentration significantly affected all parameters. NP media and peptone concentration of 3 g/L are the best media and concentrations for the number of leaves (5.67; 5.89), leaf appearance (19.83; 20.22 days ); leaf width (0.42; 0.47 cm), leaf length (1.47 cm); root length (2.83; 2.86 cm), number of roots (2.33; 2.56), and root appearance (28; 27.22 days). The combination of NP media with pepton concentration of 3 g/L is the best combination for the growth of P. amabilis plantlets at the subculture stage.

Abstract

This study aimed to isolate and test the antibacterial activity of endophytic fungi from elephant ginger rhizome (Zingiber officinale var. Roscoe) against MRSA. This research is an exploratory research which is continued with experimental. The research sample was obtained from three locations. The first sample was from Pancasari Village, Singaraja, Bali. The second sample was from Peguyanganan Village, Denpasar, Bali. The third sample is from Tebuana Village, Tampaksiring, Bali. This research was conducted at the Microbiology Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, Udayana University. Eight isolates of endophytic fungi were successfully isolated with each inhibitory zone against MRSA, Fusarium sp. 1 (21 mm), Fusarium sp. 2 (19 mm), Fusarium sp. 3 (20 mm), Fusarium sp. 4 (21 mm), Aspergillus sp. 1 (32 mm), Aspergillus sp. 2 (25 mm), Fungi S3F1 (13 mm) and Fusarium sp. 5 (25 mm). This shows that endophytic fungi can be used as an alternative producer of antibacterial compounds. Keywords: Antibacterial, Endophytic fungi, Methicillin Resistant Staphylococcus aureus.

Abstract

The study was carried out to isolate and identify active compounds in jeringau leaf extract (Acorus calamus) and test their activity to inhibit the growth of Athelia rolfsii fungus that causes stem rot in soybean plants. The study began with extraction of Jeringau leaves, extract inhibition test on Athelia rolfsii fungus, partition, fractionation with column chromatography and thin layer chromatography (TLC), fraction inhibition test and identification of active compounds by Gas Chromatography-Mass Spectroscopy (GC-MS) analysis. Coarse jeringau leaf extract can inhibit A. rolfsii fungus with inhibition zone diameter of 25 mm. The results of the analysis of active fractions of jeringau extract using GC-MS (GCMS-QP2010 Ultra SHIMADZU) obtained 24 active compounds namely: Ethanol, 2-(2-ethoxyethoxy)-CAS; 6-Okthenal,3,7-dimethyl; beta-Citronellol; Trans-Geraniol; 6-Octen-1-ol 3,7-dimethil-acetate; Benzene 1-methoxy-4-(2-propenil); 2,6-Otahen-1-ol 3,7-dimethil-acetate; Caryophyllen; 3-Allyl-6- methoxyphenol; Dodecanoic acid methil ester; Methil tetradecanoate; Cis-Asarone; Alpha hexylcinnamic aldehyde; Hexadecanoic acid methyl esteter; 7-Acethyl-6-ethyl-1-1-4-4-tetramethylhetralin; benzyl benzoate; (2(Chloro-phenyl)-(2-Methoxy-6-MI; 9-Octadecenoic acid methyl ester; 9.12-Octadecadienoic acid (Z.Z)-methyl ester; Oxinaneoctanoic acid 3-octyl-methyl ester cis:; Octadecenoic acid 9.10-didydroxyl-methyl ester; Oxinaneoctanoic acid 3-octyl-methyl trans; 9-Octadecenoic acid (Z)-methyl ester (CAS); 1.2-Benzenedicarboxylic acid, bis (2-ethylhexy). Abstract

Bigeye tuna (Thunnus obesus) is a main commodity for international markets of tuna fisheries in Indonesia, resulting in an increasing tuna capturing across Indian Ocean; Therefore, improvements in management and catchment are needed for tuna sustainability, such as a better understanding in its population structures. 30 fins tissues of bigeye tuna in total, were collected from two parts of Indian Ocean, (western Sumatra and southern Nusa Tenggara) during 20 December 2015 until Mei 2016. This study was aimed to gain some information on population structures of bigeye tuna of Indian Ocean based on microsatellite analysis. DNA analysis were conducted in a laboratory of Institute of Marine Research and Development, Gondol, Bali, Indonesia and the 1st Base -Singapore. The benefit of this study is to provide data and information on population structures of bigeye tuna of Indian Ocean. The results of Analysis of Molecular Variance (AMOVA) indicated that populations of bigeye tunas captured from the two fishing areas are included in one population.

Abstract

Cananga odorata and Cymbopogon citratus are two plant species producing inhibitory compounds in their essential oils. In our current research, the potential of their essential oils to inhibit the growth of Staphylococcus aureus and Candida albicans were investigated. MIC (Minimum Inhibitory Concentration) values of mixture of those essential oils with a ratio of 1:1 on those microbes were also assayed. Bioassays were conducted on Nutrient Agar (NA) or Potato Dextrose Agar (PDA) medium added with various ratios of the two plant species essential oils. Medium added with sterile distilled water served as control. The data obtained were analyzed quantitatively with SPSS software for Windows version 22. When significant different at p<0.05 was indicated, the analysis was continued with Duncan's multiple range test. The result showed that all treatment of essential oils applied effectively eliminated the growth of S.aureus. C.albicans appeared to be relatively resistant to 100% of C.odorata essential oil. Elimination of C.albicans was observed when the essential oil of C.odorata was combined with that of C.citratus. When compared to control, all ratios of essential oils treatments were statistically significant (p<0.05). The MIC values of essential oil mixture a ratio of 1:1 on S.aureus and C.albicans were 0,1% and 2,6% respectively. Abstract

Sodium nitrite (NaNO2) is a food preservative that is often added to processed meat products. Excessive consumption of NaNO2 can be detrimental to health. One effort to minimize the negative effects caused by NaNO2 is by consuming antioxidants. This study aimed to determine the effectiveness of Eucheuma cotonii seaweed extract on improving blood profile, liver and kidney function in male white rats (Rattus norvegicus) induced by NaNO2. This research used a Completely Randomized Design (CRD) consisting of two control groups and three treatment groups. Each group consists of six replications. Negative control (K-) receive aquades and CMC NA 0,5%. Positive control (K +) was only received 22.5 mg/kgBW NaNO2 for 35 days then on the day 36 until the day 45 only given 0,5% CMC Na. The treatment groups P1, P2, and P3 respectively on day 1 to day 15, only received 22.5 mg/kgBW NaNO2 in the morning, and then day 16 to day 35 received 22.5 mg/kgBW NaNO2 in the morning and received 150 mg/kgBW (P1), 300 mg/kgBW (P2), 450 mg/kgBW (P3) Eucheuma cottoni seaweed extract in the evening. Day 30 to day 45, the treatment groups only received Eucheuma cottoni seaweed extracts at each dose. Blood and liver were collected on day 46. Data analysis of serum alanine transaminase (ALT) and aspartate aminotransferase (AST) enzyme levels and liver histology were performed with Anova statistical test and continued with Duncan post hoc test. The results showed Eucheuma cottonii extract dose of 150 mg / kgBB significantly reduced the level of liver damage seen from the levels of ALT and AST and liver histology (percentage inflammatory cell infiltration, congestion, fatty degeneration and necrosis) in white rats induced by sodium nitrite (NaNO2). Abstract

Pollination is the most important live processes for plant especially for seedling plant. Some previous research results show that insects play an important role in helping the pollination process both in wild plants and in productive plants. The eggplant (Solanum melongena L.) is one of the important agricultural commodities. However, the eggplant production in the local farming system today often experiences obstacles when its production is not in sufficient quantities. The potential of Apis cerana L. as a pollinator is a chance solve the productions problems. The purpose of this study was to determine the role of honey bees (Apis cerana L.) in increasing yield of eggplant (Solanum melongena L.).This study obtained the eggplant productivity data. All data was collected and analyzed with SPSS 17 statistical program. Before analysis, all data normality were tested and it was analyzed by One-way ANOVA and when significant differences were found, analysis was continued with the Tuckey Test to find out the difference within treatment with a significance value ???? <0.05 .The results of this study note that bees have a certain amount of time to carry out their activities and the addition of bees colony to the experimental field produced two times more of yield compared with the other groups that were not treated with he colony. Abstract

Diabetes mellitus (DM) may lead to disruption of normal sexual function in males including abnormalities of orgasmic/ejaculatory function and desire/libido. Simplisia of dried Jengkol fruit peel has been used by people to treat diabetes. The aim of this study was to examine the effectiveness of ethanol extract of jengkol fruit peel (A. pauciflorum) (EEJFP) in improving the sexual behavior of male rats (R. norvegicus) diabetic models. Induction of diabetes was carried out intravenously by using streptozotocin dose 60 mg/ kg BW in the test animals except for the negative control group.The treatments are negative control (Carboxy Methyl Cellulose (CMC) 0.05%), positive control (CMC 0.05%), P1 (EEJFP 385 mg/kg BW), P2 (EEJFP 770 mg/kg BW) and comparison (glibenclamide dose 10 mg/kg BW) for 54 days. The parameters observed are the number of introduction and climbing of male rats to female rats. Data obtained from each parameter were analyzed by analysis of variance (ANAVA) test with a 95% confidence level and Duncan's test. The results of this study indicate that the number of introductions and climbing in the P1 and P2 test animal groups are significantly different from the positive control treatment and close to the value in the negative control treatment group. It can be concluded that the ethanol extract of jengkol fruit peel (A. pauciflorum) affects the sexual behavior of male rats (R. norvegicus) diabetes models by increasing introduction and climbing.
