Abstract

This research was conducted to determine the quality of beef in slaughterhouses and Traditional markets in Denpasar, Badung, and Klungkung, in terms of Escherichia coli and isolation E. coli O157:H7 which are suspected of contaminating beef. Experimental research was conducted in a laboratory with a heating test of E. coli O157: H7 to determine the heat resistance of E. coli O157: H7. Beef taken at Nyanggelan, Panjer market shows the highest value of E. coli which is 222.3 colonies/g, and the lowest value of E. coli is obtained in beef taken at Kaliunda abattoir which is 2.3 colonies/g. The identification results on 39 beef samples showed that all beef samples contained E. coli, 24 samples positive of E. coli O157, and 9 other samples showed positive E. coli O157:H7. The heating treatment at 60? still grows E. coli O157:H7 colonies in the Petri dish, but there has been a decrease in the number of initial colonies before the heating process. All locations were 100% contaminated with E. coli, 61.5% positive E. coli O157, and 25.6% positive E. coli O157:H7. Heating 65? for 15 seconds and 70? for 5 seconds can kill E. coli O157: H7 in beef. Abstract

Moringa is a plant whose leaf are commonly consumed as vegetables and now moringa leaves are also used as tea and additives in cakes. The results of the study on moringa are still controversial, on the one hand saying moringa can improve the quality of spermatozoa and on the other hand say it can inhibit fertility.The aim of this study was to analyze the effect of moringa leaf steeping on the concentration of spermatozoa and epithelial thickness of seminiferous tubules. A male rat with as many as 24 with weighing 200 g were divided into four treatments namely control (P0), 18 mg / kg BB (P1), 36 mg / kg BB (P2) and 72 mg / kg BB (P3) for 30 days. The results showed that moringa leaves steeping had a significant effect by decreasing the concentration of spermatozoa (P?0.05), moringa leaf steeping has no effect on the epithelial thickness of the seminiferous tubules but causes the germinal epithelium to become loose because many epithelium is loose.

Abstract

Flies are insects that live close to the human environment. Flies around the settlement were many and carry a variety of diseases that can danger the public health. Research purpose is to determine the relative abundance, frequency and dominance of flies in several traditional markets at Martapura District. This research was conducted in five traditional markets, namely Sejumput market, Batuah market, Sekumpul market, Permata market, and Kuliner market in April to May 2020. Flies were collected using a sweep net by swinging it around the landfills. The collected flies were then identified using identification keys. Results showed there were four species of flies in research location, that are Musca domestica, M. cinducens, Chrysomya megacephala, and C. rufifacies. The relative abundance, species frequency and species dominance signified that the capturing of M. domestica flies is the highest. Furthermore, Batuah market location have the highest amount of relative abundance, species frequency, and species dominance of flies compared to other locations. This research is useful as preliminary information in determining the potential of flies as a vector of disease transmission in an environment. Abstract

Physiological changes caused by environmental stress and associated health status can be detected by haematological levels. This study aims to determine the health status of groupers (Epinephelus sp.) in the East Coast cage Pangandaran. The method used is observation method which includes the observation of location, sampling of fish, and interview with keramba owner and direct observation in the laboratory which includes identification of fish species, visual observation of fish, blood sampling, calculation of erythrocyte count, leucocytes, and measurement of hemoglobin . The data obtained were analyzed descriptively qualitatively presented in the form of tables and drawings. Observations showed that beautiful groupers (Epinephelus sp.) were the most cultivated fish and one of the most popular fish. This grouper has an erythrocyte count of 2.31 x 106 ± 1.79 cells / mm3, a leukocyte count of 226,325 x 103 ± 0.23 cells / mm3, and hemoglobin levels of 9.8 ± 2.8 gram%. Based on these data, it can be concluded that there is the beautiful grouper fish as unhealthy which cultivated in East Coast cages Pangandaran because the number of leukocytes that exceed the normal range and low hemoglobin levels. Abstract

The main objective of this research was to isolate and identify microbes residing in the rhizosphere zone of strawberry (Fragaria x ananassa Dutch.), cultivated at Pancasari village, Bedugul Bali. This was an explorative and descriptive research conducted from April to July 2018. Soil samples were collected from the rhizosphere of strawberry plants and microbes residing in them were isolated by applying dilution and platting method, purified, and identified at the Food Microbology Laboratory, Faculty of Agricultural Technology, Udayana University. Important characteristics of the isolated microbes were compared to those specified in references, such as Bergey’s Manual of Determine Bacteriology 9th Edition (Holt et al., 1994) for bacterial isolates and Fungal Identification keys (Barnett and Hunter, 1972; dan Pitt and Hocking, 2009) for fungal isolates. Microbes successfully isolated and identified in this research included fungi and bacteria belong to Aspergillus flavus, Aspergillus niger, Fusarium oxysporum, and Colletotrichum sp., Pseudomonas sp., and Bacillus sp. Abstract

Aedes aegypti L. mosquito is the vector of dengue fever disease. The effort to control the vector is by applying anti-mosquito lotion to the skin. Basil essential oil was formulated into anttimosquito lotions which is practice and easy to be carried anywhere. The use of atimosquito lotion with natural active compound of basil essential oil is one of safe solutions to repelt mosquitos. This study aimed to calculate yield of essential oil from basil leaves, to identify the active compounds, to measure protection ability of the antimosquito lotions, to to anlyse physical properties of the lotions and to analyse perception of panelists toward the lotions. The essential oil was extracted using steam distillation. The active compounds were analysed using GS-MS. Protection ability of the lotion toard Ae. aegypti L. was tested under laboratory condition using hand in cage method. Physical properties of the lotion were measured for organoleptic, pH, viscosity and homogeneity. Results showed that the yield of essential oil from basil leaves was 0,26% ± 0,05 w/w. GCMS test results showed there were 25 active compounds of the oil. The highest protection ability against A aegypti L. was lotion with 5% basil essential oil. Organoleptic test results show a thick texture, yellowish white color, aroma of olive oil and basil and a soft impression on the skin. Results of pH, viscosity and homogeneity tests show that anti-mosquito lotions containing essential oil from basil leaves met the requirements in the Indonesian National Standard. The hedonic test shows that panelists like lotion with 5% basil essential oil. The effect of lotion gives softness, comfort and the absence of sensitivity symptoms after being applied to the skin. Abstract

Lepidotrigona terminata is one of the stingless bees of the Apidae family. Morphological characteristics of L. terminata species are very rare due to their genetic characteristics. The elongation factor 1? gene has been studied in the growth and development of both morphology and molecular systematics. This study aims to describe the morphological character of L. terminata from Temanggung Regency, Central Java with a purposive sampling method and analyze the conserved gene elongation factor 1? area to explain genetic variation based on the proportion of DNA bases analyzed in silico and computational methods using a database in GenBank with number access DQ813123.1, DQ813125.1 and DQ813124.1. The results of the analysis explained that the morphological character of L. terminata is very difficult to distinguish from several other Lepidotrigona groups and it is suspected that the genetic trait of the elongation factor 1? gene has an influence on the morphological character of L. terminata. The proportion of DNA bases of the target gene has a length of about 682 bp to 780 bp with the highest proportion of bases found in Adenine (A) around 216 bp (31.67%) to 251 bp (32.18%), the least number of bases found in Guanin (G) around 121 bp (17.74%) to 140 bp (17.95%) and has twelve conserved gene regions. The data obtained from the research provide basic information to study the morphological characteristics and conserved regions of the elongation factor 1? gene in L. terminata. keywords: L. terminata, morphology, elongation factor 1?, in silico, computational methods

Abstract

Plant breeding is an effort to develop plants by manipulating the genetic system of plants to get new superior cultivars. Plant breeding often produces incompatible seeds, so a rescue process is needed to reduce the risk of embryo death. The addition of gibberellic acid could break the seed dormancy for seed that hard to germinate. The purpose of this study was to find out the best concentration of gibberellic acid for germination of citrus globular embryos from protoplast fusion. Research was conducted at the Biology and Cell Biology Research Group In Vitro Culture Laboratory, Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development (BB BIOGEN), Agricultural Research and Development Agency, Ministry of Agriculture, Bogor. The study used Completely Randomized Design (CRD) with concentration 0, 1, 2 and 3 ppm of gibberellic acid as treatment, each consists of 7 replicates. This study used globular embryos measuring ± 3 mm of inbred FS 29 and FS 89 from protoplast fusion between Siam Simadu and Mandarin Satsuma citrus. Observations were made on the percentage of germinated embryos, time of germination, the number of the complete and incomplete shoot (abnormal, only part of the roots or buds that developed). Data were analyzed using Crosstab analysis test with IBM SPSS Statistics 22 software. Result showed that addition of gibberellic acid induce faster germination time and increase seedling percentage. The best germination medium for citrus FS 29 was VMW with 2 ppm and 3 ppm gibberellic acid with the seedling percentage around 85,7% in 7 days, while the best germination medium for citrus FS 89 was VMW with 1 ppm gibberellic acid with 100% seedling embryos in 8,5 days. VMW medium added with 1 ppm, 2 ppm and 3 ppm gibberellic acid for citrus FS29 and VMW medium added with 2 ppm and 3 ppm for citrus FS89 were good to get the complete sprouts with a succes rate of 100%.
