Abstract

In this research had been done the development of analysis qualitative and quantitative methods of LDL-C in human serum using agarose electrophoresis. The methods that had been developments are separation lipoprotein in human serum using agarose electrophoresis with TAE pH 8,6 buffer system and quantification methods for LDL-C using spectrophotometer UV-Vis. The aim of this research is to find out a simple method that can separate lipoproteins of human serum and quantificating LDL-C. The results of this research shows agarose electrophoresis with TAE pH 8,6 buffer system could  separate LDL-C in human serum as good as electrophoresis with Na-Barbital buffer system. However, for its quantification only can be done with spectrophotometer UV-Vis.

Abstract

Advance derivatization study of several amines with dansyl chloride has been carried out. The aim of this study was to determine influence of molecul structures compound toward dansylation. Derivatization was done by reacting  4000 ng drugs which contain primary or secondary amine group with 100 ng dansyl chloride at a temperature 450 C and pH 9 for a hour. Interference was separated using stationary phase silica G60 and mobile phase  etyl acetat : methanol : ammonia (85:10:5), detection on spectrophotodensytometer ?eks 312 nm mode fluorescence with filter K400. Compounds that have a primary or secondary amine groups can be derivatized using  dansyl chloride though that most affects  of  the reaction is  rigidity of molecul structure. Differences fluorescence intensity between each derivatives influenced by the reactivity amine groups and steric hindrance of the compound.

Abstract

The research was conducted to determine the influence of mobile phase polarity of acetone-dichloromethana : methanol-acetic acid on the percent distribution of (+)-catechin from gambier by vacuum liquid chromatography and to determine the polarity mobile phase required to obtain the largest amount of (+)-catechin percent distribution. Gambier extract was fractionated using vacuum liquid chromatography with several variations of polarity of acetone-dichloromethane : methanol-acetic acid mobile phase expressed by the dielectric constant value (?) of 13.74 to 17.51. Fractions were analyzed by TLC-spectrophotodensitometer to calculate the concentration and the percent distribution of (+)-catechin. To know the effect of variations in mobile phase polarity on the percent distribution of (+)-catechin performed statistical analysis using one-way ANOVA test (? = 0.05). The results show that the variation of polarity of acetone-dichloromethane : methanol-acetic acid mobile phase (? = 13.74 to 17.51) had a significant influence on percent distribution of (+)-catechin from gambier by vacuum liquid chromatography method (p <0.05). Fraction with a mobile phase of acetone-dichloromethane (4:6) 50% : methanol-acetic acid (3:4) 50%, which had a value of dielectric constant of 15.62 yield the largest amount of (+)-catechin percent distribution value (45.40%).

Abstract

The purpose of this research was to compare the DPPH scavenging activity of black tea (Camellia sinensis (L.) OK) and gambier (Uncaria gambir (Hunter) Roxb) product extract and to determine their TLC-densitometer profile. This research was conducted in two stages, first stage was to examine the DPPH scavenging activity of black tea and gambier product extract. This assay used some concentrations of black tea product extract, gambier product extract, and standard vitamin C (10, 20, 40, 80, 160, 200, and 320 µg/mL). The second stage was to determine the extracts profile by TLC-densitometer. The result of this research showed that the DPPH scavenging activity of gambier product extract was greater (IC50 = 88,57±0,64 µg/mL) than black tea product extract (IC50 = 311,54±2,79 µg/mL). The result of profile determination of black tea product extract at a mobile phase of MeOH:toluene (50:50) showed that no spot was supposed a volatile oil group; at mobile phase of ethyl acetate:formic acid:acetic acid:H2O (100:11:11:26) showed two spots that was supposed a flavonoid group, spot with Rf 0,41 (?max 365 nm) and spot with Rf 0,97 (?max 280 nm); at mobile phase of CHCl3:MeOH (90:10) showed that spot with Rf 0,87 (?max 275 nm) was supposed an alkaloid group; at mobile phase of acetic acid:diethyl ether:n-hexane:ethyl acetate (20:20:20:40) showed that spot with Rf 0,62 (?max 290 nm) was supposed a tannin group. While the result of profile determination of gambier product extract at mobile phase of ethyl acetate:formic acid:acetic acid:H2O (100:11:11:26) showed two spots that was supposed a flavonoid group, spot with Rf 0,81 (?max 280 nm) and spot with Rf 0,92 (?max 280 nm); at mobile phase of acetic acid:diethyl ether:n-hexane:ethyl acetate (20:20:20:40) showed that spot with Rf 0,42 (?max 285 nm) was supposed a tannin group.

Abstract

Corn starch is widely used as excipient in the manufacture of tablets. Native corn starch that have a bad flowability were modificated into pregelatinized corn starch to increase it flowability. The aim of this research was to know the effect of the use of pregelatinized corn starch as disintegrant on the physical properties of Vitamin E tablets for dogs with direct compression method. Pregelatinized corn starch is made by heating the suspenssion of starch at 550 C for 10 minutes and then dried. Pregelatinized corn starch is used as a disintegrants of vitamin E tablets for dog which made by direct compression with three variation of pregelatinized corn starch concentration, that is 5%; 7.5%; and 10%. Quality of resulting tablet are known through the hardness test, friability test, and disintegrating time test. The result from the research will be analysed by One-way ANOVA method. The result showed that the use of pregelatinized corn starch as disintegrant at concentration 5%; 7.5%; and 10% with direct compression method do not give an effect to physical properties of the tablets and  do not meet the requirements of a good tablet.

Abstract

Fluorescence detection could be used for enhance  the  sensitivity of morphine  detection  by TLC-Spectrophotodensytometry. Morfin is a compound  that fluoresces intrinsically incapable. ence, morphine  was derivatized using dansyl chloride. Morphine was derivatized by two methods, derivatization in solution and overspotting method. The derivatization reaction was kept at pH 9 and 450  C for 20 minute. The ratio between dansyl chloride and analyte concentration was 6:1. The derivatization result was separated from the interference using TLC. The TLC method was using Al-TLC silica G60 plate as a stationary phase and etyl acetate : methanol : amonia (85:10:5) as the mobile phase. The dansyl derivative was detected at ?eks 365 nm and K540 filter for fluoresence. Derivatization  of  morphine-dansyl chloride capable  to  enhancing  the  sensitivity of  morphine detection using TLC-Spectrophotodensytometry. Derivatization in solution and overspotting method took the same time in procedure  but  they  gave  different sensitivity. Derivatization in solution method gave more quantity  than derivatization of overspotting.

Abstract

A research which observing drug interaction in chronic kidney diseasse hypertension compliance patients in RSUP Sanglah Denpasar has been carried out. This research was carried to find out the number of drug interactions in  chronic kidney diseasse hypertension compliance patients therapy. This research was run descriptively and retrospectively. The studies were done by observing  the adverse effect that may raised from the drug interaction which was used. Furthermore, the research also covered about the management to overcome the problem that raised from each interactions. There are about 20 patients which has been confirmed passed the inclusions and exclusions criteria. The observations which were held literaturely based on Lexi Comp was resulting 16 drug interactions. There was no drug interaction with A and X risk factor found. While, drug interactions of B and C risk factor occurred 37,5% with 6 spesific drug interactions for each. Four drug interactions occurred in D risk factor category, it is about 25%. The studies which were held literaturely  based on Drug.com showed 26 spesific interactios with 7 minor interactions, 18 moderate interactions and 1 major interaction, which in percentage about 26,92%, 69,23% and 3,85% respectively.

Abstract

Cassava starch is the common type of starch with enormous application in pharmaceutical industries. However, cassava starch has poor compressibility and flowing hard. Therefore, it is need to be modified. One of starch modification methods is physical method like gelatinization because easier than the other method. A good pregelatin starch will be formed by the addition of water and use the proper temperature. This research aimed todetermine the effect ofratio starch:waterandheatingtemperature of cassavastarchtowardphysical characteristic ofpregelatinized cassavastarchintendedasa tabletexcipient. This research was conducted through several stage. First, the manufacture of pregalatinized cassava starch by using ratio starch:water that are 1:0,5 ; 1:0,75, 1:1 and heating temperature are 50°C ; 55°C ; 60°C. Furthermore, cassava starch and pregelatinized cassava starch preliminary tested. Starch which meets the standard then tested their physical properties including moisture testing, particle size distribution test, flow properties test dan compressibility test. Starch with physical properties that meet the requirements then compressed with direct compression method and tested. Testing includes weight uniformity, hardness, friability and disintegrant time. Data were analyzed with one-way ANOVA and LSD with 95% confidence level. The result indicated, the addition of water and heating temperature gives a significance influence (P<0,05) toward the flow characteristic and compressibility of pregelatinized cassava starch. Increasing the amount of water and heating temperature causes decreasing of compressibility. Decreasing of compressibility make flow properties is better. Based on physical properties of test criteria tablet, then selected ratio 1:1 (50°C) as selected formula for tablet excipient direct compression method.

Abstract

Pediatric prescribing need to get special attention  because drug utilization for pediatric is related with different rate of the development of the organ, body system, as well as enzyme that responsible to drug metabolism and drug excretion are not  complete yet. Medication error perhaps will be found in pediatric prescribing so a study about prescription that potentially made medication error happened must be carried out, therefore the medication error occurrence can be minimized or preventable. This study was run by using descriptive evaluative method by observing the prescription of some children in hospital at Gianyar regency during February to April 2009. The sample was conducted by accidental sampling. The total of prescriptions that used to be sample was 96. The data’s including: the age of patient, the variant of the drugs, dosages, and the type of drugs, so medication error could be observed or not. The Result of study showed that the total of male patients were 60,42% and female patients were 39,58%, while the age range of patients showed were between 6-11 months. There were 16 categories of drugs that used most frequent were cough and cold medicine about 20%, vitamin and mineral about 18,93%, and analgesic and antipyretic13,93%. The prevalence of error occured was 277 error that consist of 78,70% in pharmaceutical adjustment, 16,61% in clinical concideration, and 4,69 % in administration.

Abstract

Corn starch is the one type of starch that potential to be developed as an excipient in manufacture of pharmaceutical dosage forms. However, corn starch has the bad flow and compactibility properties. According to that matters, chemicaly modified corn starch using ? amilase enzym which is produced by Lactobacillus acidophilus highly neccesesary. It is known that ?-amylase enzyme is capable of changed the microscopic structure of starch then produced better flow rate and compactibility properties. This study was conducted to see the effect of variations in the concentration of bacteria Lactobacillus acidophilus producing ?-amilase enzime on the granule physical properties of fermented corn starch. This research was done by fermentating the corn starch using the Lactobacillus acidophilus bacteria with a variations time of fermentation which is 0, 24, 48, 72, and 84. The effect of variations time of fermentation which used Lactobacillus acidophilus on physical properties of fermented corn starch is performed by flow rate and compressibility test. The data were analyzed using one-way ANOVA with 95% confidence level. Results showed that the concentration of the Lactobacillus acidophilus bacteria significantly influence the granule physical properties of fermented corn starch. Fermented corn starch for 72 hours had been produced the best physical properties in flow rate and compactibility properties.
