Abstract

This study aim was to evaluate quality of anaesthesia by using gravimetric infusion anaesthesia withketamine and propofol in dogs. The quality of anaesthesia, duration of actions, and the physiological responsseof anaesthesia were evaluated in twenty domestic dogs. Anaesthesia was induced intramuscularly withatropine (0.03 mg/kg)-xylazine (2 mg/kg) (AX), intravenously ketamine-propofol (KP) (4 mg/kg), andmaintained with continuous intravenous infusion with pre-mixed propofol (P) and normal saline containing2 mg/ml of propofol and 2 mg/ml of ketamine (K). Domestic stray dogs were randomly divided into fivegroups. Groups AXKP-K2P2, AXKP-K4P4, and AXKP-K6P6 were treated with ketamine-propofol the dose0.2 mg/kg/minute, 0.4 and 0.6 mg/kg/minute respectively, while group AXKP-P4 was given propofol 0.4 mg/kg/minute and group AXKP-I was given isoflurane 1-2%. Heart rate (HR), respiratory rate (RR),electrocardiogram (ECG), blood oxygen saturation (SpO2), end tidal CO2 (ET CO2), and capillary refill time(CRT) were measured. No significant difference (P>0.05) found between the groups in anaesthetion times.All groups showed rapid and smooth inductions, prolonged surgical stage, and rapid recovery. Groups AXKPK2P2and AXKP-K4P4 showed minimal physiological effect on the dogs. The HR, RR, ET CO2, SpO2, CRT,and ECG wave were stabl. Combination of AXKP-K6P6 induced SpO2 depression, increased and instabilityof HR, RR and ET CO2. Groups AXKP-P4 showed decreased of HR and respiratory depression. All anaestheticcombinations showed no significant influence (P>0.05) on the electricity of the dog’s heart. The combinationof ketamine-propofol at dose 0.2 and 0.4 mg/kg/minute were found to be better as an application formaintaining anaesthesia by gravimetric continuous intravenous infusion. The method is a suitablealternative for inhalation anaesthesia in dogs.

Abstract

The purpose of this study was to verify the effectiveness of cumulus cells as a feeder layer in supportingthe growth of mouse embryonic stem cells. Embryos at blastocyst stage were exposed in pronase solution,and then incubated in rabbit anti-mouse antibody and guinea pig complement to lyse and separate thetrophoblast cells from the inner cell mass. Inner cell mass subsequently cultured in a petri dish containinga cumulus feeder layer, mouse embryonic fibroblasts, or without a feeder layer, in stem cells medium. Theresulting stem cells colony passaged in trypsin solution, pipetted repeatedly to produce subcolonies orsingle cells, and cultured as before in some new petri dishes. Characterization of stem cells was identifiedby using alkaline phosphatase staining. The results showed the effectiveness of cumulus cells as feederlayer for culture of mouse embryonic stem cells was comparable with mouse embryonic fibroblasts, andboth of them were better than without a feeder layer method. The number of primary colony, cell lines, andcolony growth rate increased 41.30, 8.70, and 54.20%, respectively, while doubling time was shorter 10:21hours compared to the growth without feeder layer method. These results prove that the cumulus feederlayer effectively supports the growth of mouse embryonic stem cells.

Abstract

The aims of this study was to determine the liver damage from alcohol intoxication in Wistar rats.The design used in this study was a randomized true experimental post test only control group design. Thestudy used 15 rats divided into 3 treatment groups each of which consists of 5 rats. The first group wasgiven distill water. The second group was given 5% alcohol, and the third group was given 20% alcohol. Ratswere treated with alcohol daily for six weeks. Biochemical markers were detected the levels of aldehydedehydrogenase (ALDH) in serum and histological changes in liver tissue. ALDH is a biochemical markerof a sensitive and specific ethanol after chronic alcohol administration. Blood sample was collected at 6and 24 hours after the last peroral administration of repeated alcohol treatment, and serum levels ofALDH was tested by enzyme linked immunosorbent assay (ELISA). The results showed that the levels ofALDH in the blood of alcohol treated Wistar rats significantly higher as compared to those of control rats.ALDH levels increased by 83.11% after administration of 5% alcohol and 112.05% after administration of20% taken after 6 hours of alcohol for 6 weeks. On samples taken after 24 hours, ALDH levels by 95.11%after administration of 5% alcohol and 86.79% after administration of 20% alcohol. Oral treatment with20% alcohol chronically was led to changes in the microscopic structure (necrosis) of liver tissue in Wistarrats. Liver tissue damage occured due to repeated use of alcohol is accompanied by increasing serum levelsof ALDH in Wistar rats.

Abstract

The objective of this study was to identify the proliferation and differentiation of mice cerebrum cellsand its protein product. Research has been conducted on in vitro growth of three days old rat cerebrum cellsin Dulbecco’s Modified Eagle Medium (DMEM) containing 10% Amino Acid Non Essential (AANE), 10%Fetal Bovine Serum (FBS), 3mM NaHCO3, 50 mg/ml gentamycin, and supplemented with and without 5mg/ml insulin, 10 mg/ml transferin, 5 mg/ml selenium (ITS). Culture was done in 5% CO2, then incubatedat 37oC until 90% confluent. Identification were done on cell proliferation and population doubling time(PDT), number of neuron and glial cells, length of axon and dendrit, and protein secretion. The number andlength of neuronal cells were calculated by using hemocytometer and eyepiece micrometer, respectivelly.Protein secreted into culture medium, designed as conditioned medium (CM) was analysized using sodiumdodecyl sulfate–polyacrilamide gel electrophoresis (SDS-PAGE) method. Quantitative data were analyzedusing statistical T-test on Minitab program. In vitro culture of rat cerebrum cells showed two types of cellsincluding nerve cells of bipolar and multipolar neurons and glial cells including astrocyte (the fibrous andprotoplasmic), oligodendrocyte, and microglia. Supplementation of ITS into the culture medium increasedthe cell proliferation rate (P<0.05) with lower PDT, and quantitatively, the 30 kDa secreted protein asindicated by the higher intensity of the protein band.

Abstract

The number of patients with chronic liver disease (fibrosis or cirrhosis of the liver) is increasing fromtime to time. However, until now there is no therapy that really effective to overcome that disease. Therapyfor liver fibrosis typically use substance that have antioxidant activity, anti-inflammatory or anti fibrotic.Various efforts always done to find alternative therapies for liver fibrosis. One of them is the use ofpotential plants that suspected having such activity. Various parts of pomegranate have been shown tohave various activities that beneficial for health. This study was conducted to determine the effect ofpomegranate fruit extract standardized 40% ellagic acid on the improvement degree of liver fibrosis causedby cholestasis by measuring of serum alkaline phosphatase levels (ALP) and gamma-glutamyl transferase(GGT), as a specific indicator of liver damage becaused of cholestasis. The research was conducted by using32 male rats, wistar strain, 2.5 month old, weighing between 150-200 grams. Animal models of liver fibrosis obtained by using BDL technique. Subjects were divided into a control group (P0 = without BDLand giving of pomegranate extract) and treatment groups (P1 = BDL with administration of CMC, P2 =BDL with ellagic acid 90% and P3 = BDL with pomegranate fruit extract standardized 40% ellagic acid).CMC, extract (150 mg / kg BW / PO) and ellagic acid (60 mg / kg BW / PO) administered for 21 consecutivedays in the same volume. At the end of 21 days periods, biochemical evaluation was performed to measureserum levels of GGT and ALP. The result indicated that administration of pomegranate fruit extract ( P3)significantly reduced GGT ( 10.5±9.2 mg/dl) and ALP level ( 509.0±4.2 mg/dl) close to normal level of GGTand ALP ( P0, GGT : 2.8 ± 1.4; ALP : 449.0±62.3) (p<0.05). The level of GGT and ALP in P3 group were lowercompared to the group ellagic acid (P2, GGT=48.5±4.8 and ALP = 691.0± 29.7) and group which only begiven CMC (P1, GGT 191.0±35.4 and ALP 890 ± 5.7) ( p<0.05). Extract of pomegranate fruit thatstandardized with 40% ellagic acid is potential as a antifibrotic agent.

Abstract

This study was carried out to determine the use of bird flu polyvalent vaccines containing two or threeor more virus isolates representating of circulating viruses in the region. Three seed isolates of avianinfluenza H5N1 virus were used in this experiment. The isolates were Chicken/Denpasar/Unud-01/2004,Chicken/Klungkung/Unud-12/2006, and Chicken/Jembrana/Unud-17/2006. The seeds were inactivatedusing 0.01% formaldehide than mixed (AI3G) alumunium hidroxide adjuvant and then injectedintramuscularly to Isa Brown layer chicken at 3 weeks of age and repeated at the age of 5 weeks. The doseof each seed virus was 27 HA units. Sera were collected at one and two weeks after the second vaccination.The result showed that the arithmetic meant titer (AMT) of sera that tested with homologous isolate washigher than the test using a heterologous isolates, in the standard haemaglutination inhibition (HI) assay.The mixed AI3G vaccine produced a uniform AMT against the constituent isolates, while vaccines withindividual isolate yielded a lower and more variation in AMT. Further experiments using a commercialhomologous H5N1 and heterologous H5N2 commercial vaccines has resulted AMT that 1-4 log lower thanAI3G vaccine. It is concluded that polyvalent vaccine with field seed isolates is recommended to be appliedin the poultry farm in Indonesia.

Abstract

Cysticercosis zoonotic parasitic disesase caused by Taenia solium larva, is a major public healthproblem. Cysticercosis results from the development of larval tapeworms in humans harboring adult T.solium or from ingesting soil containing eggs shed in the feces of humans, in areas where there are nolatrines. Humans are accidental intermediate hosts and pigs are the normal intermediate hosts. Clinicallythey are most serious when located in the central nervous system or in the eye where they persist formonths to years. Cysticercosis occurs worldwide primarily in developing country where pigs are raised,pork consumed and poor sanitation allows pigs’ access to human faeces. The occurance of the diseases isexpected to increase in relation to the growing demand for pork in those countries. In Indonesia cyticercosisis still a very important health problem, especially in Bali and Papua. The majority of the populations inBali and Papua are Hindus and Protestants/Catholics, respectively, so pork widely consumed in thoseislands. This brief review summarizes the present situation and problems of cysticercosis in animal inBali and Papua. Epidemiological data of the diseases were obtained from several sources.

Abstract

The objective of this study was to observe the effect of functional synbiotics yoghurt (made ofindigenous probiotic + oligofructosaccharides) in rats that were infected with Enteropathogenic Escherichiacoli (EPEC), and to detect its effect on hematological status. A total of 25 male Sprague Dawley rats wereused in this study and divided into five treatment groups: (i) negative-control; (ii) positive-control; (iii)synbiotic yoghurt; (iv) synbiotic yoghurt + EPEC; and (v) conventional prebiotic yoghurt. Yoghurt (109 cfu/mL lactic acid bacteria) was given orally using feeding tube at day one until day 4. At day 8 – day 14animals were infected with EPEC (107 cfu/mL) orally using feeding tube; at day 22 blood samples werecollected for hematology analysis (erythrocytes, hematocrit (Hct), hemoglobulin (Hb), platelets, andleucocytes). Post infection with the EPEC diarrhea was developed in rats both in the control positivegroups and in the synbiotic yoghurt + EPEC group. Animals in the positive-control group had a significantlyhigher thrombocytes and leucocytes counts and Hct compared to that in animals in the other groups(P<0.01). Whereas there were no significant effect on the erythrocytes counts and Hb (P>0.05). The treatmentwith synbiotic yoghurt, in both synbiotic yoghurt only and synbiotic yoghurt + EPEC groups significantlyreduced the platelets and leucocytes counts and Hct of rats

Abstract

Coccidiosis is one of the important diseases in poultry industry. In Indonesia the morbidity of thedisease is between 80 to 90%. A rapid and prompt diagnosis would be one of the essential steps ineradication and control of the disease. The objective of this study is to determine the protein profile ofsporozoite of Eimeria tenella isolated in Yogyakarta using sodium dodecyl sulphate polyacrilamide gelelectrophoresis (SDS-PAGE) protein analysis. Protein analysis was performed in 12% polyacrilamide geland further electrophoresis at 100 volts and over-staining with Coomasie brilliant blue. The resultsshowed that the sporozoite of E. tenella isolated in Yogyakarta contained five proteins with molecularweights of 15, 26, 32, 80, and 91 kDa, respectively.

Abstract

This study aimed to determine the antiplasmodial activity of sernai leaves (Wedelia biflora) extractagainst Plasmodium berghei which were infected to mice experimentally using the 4-days suppressive testmethod. The extraction was performed by maceration in three different solvents: ethanol, n-hexane, andethyl acetate. Inoculation of 1x106 P. berghei was performed in 60 Swiss mice intraperitoneally. Theantiplasmodial activity of the ethanol, n-hexana, and ethyl-acetate leaves extract at 4 different doses(100, 80, 60 and 40 mg/kg BW/day, respectively) were tested against P. berghei in vivo for 4 consecutivedays. Animals were given the leaves extracted orally. The negative-control animals were given dimethylsulfoxide diluents (DMSO) and animals in the positive-control were treated with P.berghei + DMSO.Parasitemia status were observed from day 1-4 by thin blood smear from tail and stained with Giemsa.On day 5, levels of urea, creatinine, SGPT and SGOT were measured. The results showed that extract ofsernai leaves using ethanol, n-hexane, and ethyl acetate had the antiplasmodial capacity on P.berghei inmice with effective doses (ED50) of 5.379 mg/kg BW, 25.306 mg/kg BW, and 27.442 mg/kg BW, respectively.The sernai leaves extract had the capability to maintain the liver function but not the kidney function.
