Abstract

Pregnancy is an integral part of the circulatory system between pups and mother. Growth anddevelopment of pups is determined by intake from their mother. The purpose of this study was to measurethe growth of pup’s bones given ethanol extract purwoceng (Pampinella alpine) for 1 to 13 days of ratgestation. Eight pregnant rats were divided into two groups: control group and purwoceng group that wasgiven ethanol extract purwoceng at a dose of 25 mg/300 g bw. The born pups were observed for 70 days tomeasure the growth and development of head bone, back bone, front legs bone, and hind legs. The resultsshowed that the growth and development of all bones parameters of female pups were higher on purwocenggroup than control the bone while male pups did not difference between purwoceng and control group exceptback bone and hind leg bone. The conclusion of this study is that administration of purwoceng roots ethanolextract on pregnant rats during organogenesis phase can extends the bone length of their pups.

Abstract

The research objectives were to evaluate and compare of production performances and blood metabolicprofiles of garut and jonggol rams fed with mung bean sprout waste and omega-3. This research used 24rams aged of 1-2 years, consisted of 12 garut rams weighed of 36,43±1,45 kg (CV=13,87%) and 12 jonggolrams weighed of 23,09±0,57kg (CV=8,64%). Research used Completely Randomized factorial Design(CRD 2x3) with 2 main treatments x 3 factorials and four replications. The first factors as the maintreatments were different breeds of garut and jonggol. The second factors as the factorial treatments werethree different feed percentages which consisted of P0 (40% grass+60% concentrate I), P1 (40% mung beansprouts waste+60% concentrate II), and P2 (40% mung bean sprouts waste+60% concentrate II+omega-3).Data were analyzed by analysis of variance (ANOVA) and proceeded by Duncan Range Test. The resultsshowed that P1 had the highest performance on feed efficiency and average daily gain (ADG) parameters.Blood metabolic test showed that blood total protein, cholesterol, and glucose of all rams were normal. Inconclusion feeding rams with mung bean sprout waste as grass substitution have a positive effect on theperformance of the rams production performance.

Abstract

Bali cattle is an indigenous spesies in Bali, which pay great attention due to its uniqueness. Numerousarticles have been published on Bali cattle especially related to its disease, nutritional requirement forgrowth and domestication. Nevertherless, it was no any report has been published on the lactic acidbacteria (LAB) assosiated with the cattles raw milk and its potential used as probiotic. This work isaimed to identify LAB isolated from bali cattle raw milk and its resistance to secondary bile acid (sodiumdeoxy cholic), a prequisite in development of probiotic for human. The results revealed that based upon thehomology studies of the variable region I, II, and III sequences of the 16S rDNA showed that 44 out of 62isolates were closely related to Pediococcus acidilactici; 11 out of 62 isolats were closely related to Enterococusgallinarum, five out of 62 isolates were closely related to Lactococcus garvieae, while only one isolate was closely related to Lactobacillus plantarum and Weisella confusa. Some isolates showed resistant to 0.2-0.6mM deoxy cholic acid, which might be also resist in human gastrointestinal tract conditions. Based onthose finding, it can be concluded that the LAB associated with raw bali cattle milk were closey related toP. acidilactici, E. gallinarum, Lac. garvieae, Lb. plantarum and W. confusa, which different from thosecommonly LAB found in others cattle raw milk. Somes isolates were potential to be developed as probioticfrom human helath.

Abstract

This research was aimed to investigate the potential use of leaf extract of Carica papaya and Ipomoeaaquatica lto improve egg hatchability and larval viability of catfish. Dried leaves of Carica papaya andIpomoea aquatica were macerated and extracted in water and ethanol. Eggs and larvae were tested in theaquarium size of 10 L with a a diameter of 28 cm. The extracts in concentration 600, 800 and 1.000 ppmwere tested on the egg hatchability of catfish with immersion method, and challed with Aeromonashydrophyla, Pseudomonas sp., and Saprolegnia spp. The extracts in concentration 800 and 1.000 ppm weretested on the larval viability with immersion method, and challed with pathogens. Water or ethanolextract of Carica papaya and Ipomoea aquatica can improve egg hatchability 67±8% until 90±6% andlarval viability of catfish 77±0,5 until 90±9%. Eight hundred ppm ethanol extract of Carica papaya has thebest egg hatchability and 1000 ppm can improve larval viability of catfish.

Abstract

Lymphocytes play important role in host defence system against pathogenic agents both in mammalianand avian species. Monoclonal antibodies (mAbs) have been widely used to identify lymphocytessubpopulation in a host based on their surface cluster differentiation (CD) markers. Currently, mAbsagainst lymphocytes surface markers of village chickens have been produced by fusion of myeloma withlymphocytes derived from spleen of mice immune to chicken lymphocytes. In two fusion experiments, 623clones of hybridomas were produced and four (BG4, CB1, DB2 and BB2) of which secreted mAbs againstchickens lymphocyte surface molecules. Two mAbs (BG4 and DB2) recognized protein of 32 kDa, one mAb(CB1) recognized protein of 64 kDa, and one mAb was unable to recognize any protein of chicken lymphocytesurface molecule. Three mAbs recognized lymphocyte subpopulation in spleen and peripheral blood ofvillage chickens. In peripheral blood, mAbs BG4, CB1 and DB2 recognized lymphocytes subpopulationwith the percentages of 11.2%, 21.4% and 7.4% respectively. In spleen those three mAbs recognizedlymphocytes subpopulations at the percentages of 38.2%, 51.54% and 31.5% respectively. Based on thoseresult, it is very likely that mAbs BG4 and DB2 recognized CD4 molecule and mAb CB1 recognized CD8molecule of village chickens lymphocytes.

Abstract

Until recently, two clades of of avian influenza viruses (AIVs) designated as 2.3.2 and 2.2.3 havebeen circulating in Indonesia. Mutations of AIV genes have cretaed many more variants of the virus. It istherefore important to evaluate the appropriate methods used for the detection and diagnosis of AI virusin the field. Reverse Transcriptase-Polymerase Chain Reaction (RT-PCR) have been used as a standardmethod for detection of AIV in many laboratories in Indonesia. The success of RT-PCR for detection ofAIV virus is dependent on the nucleotide sequences of primer that match with the circulating of AIVs. Theaims of this study was to develop RT-PCR by designing primers for H5 subtype specific to the circulatingAIVs in the field. The primers were designed using Primer Design software, and optimization andvalidation of the primer were conducted using AIVs that have been characterized in the previous study.The primers were then used RT-PCR using AIV isolates from field samples and their sensitivity andspecificity were then determined. The results showed that the H5 primers designed in this study, H5-IDand H5-NLP, was able to detect the AIVs in field samples better than the H5-specific primers have beenused previously. In conclusion, H5 primers designed based on recent viruses in the field showed betterresults in the detection of AI virus as compared to the previous primers. As AIV-H5N1 subtype in the fieldwill continue to change and evolve, the use of primers designed in this study is recommended for diagnosisof H5 AIV.

Abstract

The availability of superior seeds garut sheep relatively few and the unavailability of seed supply ofsuperior male garut sheep that is continuously high productive is also a problem. . The success ofcryopreservation is influenced by the diluent being used. This study aimed was to compare the Tris EggYolk containing omega-3 (TEYO) and Tris used commercial egg yolk plus omega-3 fish oil salmon (TEYOS)to the success of cryopreservation of garut sheep semen. Five garut ram aged 1,5 – 2,0 years were used inthis study. Semen was collected using an artificial vagina, then it was evaluated and divided into twotubes, each of them was diluted with TEYO and TEYOS (50x106 sel/straw), before they then packed intostraws, equilibrated (5°C), and frozen with liquid nitrogen vapour (-130oC) for10 minutes and storedthem in the container (-196oC) for further evaluation. The results showed that post thawing values of thesperm motility, viability and intact plasma membrane were 53,75±2,46; 60,75±2,17and 72,58±2,12%respectively, diluted with TEYOS were higher (P<0,05) than those diluted with TEYOS with the spermmotility, viability and intact plasma membrane only at 41,50±1,25; 50,50±1,04 and 57,58±1,03%respectively. This result demonstrated that the recovery rate of spermatozoa in TEYOS diluent washigher (P<0,05) than TEYO. Its concluded that fish salmon oil omega-3 supplemented in TEYOS better incryopreservation of garut ram semen compared to the TEYO diluent.

Abstract

Eschericia coli contamination was found on egg powder imported to Indonesia during microbialexamination. The study was conducted to describe the presence of E. coli and its resistance to antibioticsin egg powder imported to Indonesia through The port of Tanjung Priok. Samples was determined usingcross sectional study and the sample size was calculated based on assumption of confidence level of 95%with margin of error of 10% and predicted prevalence of 50%. Total of 100 egg powder samples wascollected in August 2014 from two exporting countries, Ukraine (whole egg powder, n=30) and India (wholeegg powder, n=40 and egg yolk powder, n=30). Examination was performed by packaging and label inspectionof the product followed by samples collection and testing for bacterial contamination using rapid test andconventional isolation and identification methods. Detected E. coli was isolated and then tested forantibiotic resistance. Examination results showed that 4 samples were positive to E. coli. E. coli isolatesshowed resistance against 6 types of antibiotics and 75% of the isolates had resistance against minimumof 3 types of antibiotics. These conditions should be taken into consideration since antibiotic resistance inE. coli would cause negative impacts on human, animal and environmental health.

Abstract

This study aim was to determine the best dimethyl sulfoxide (DMSO) concentration for native chicken’s(Gallus gallus domesticus) frozen semen. Semen was collected from three native chickens and then evaluatedmacroscopically and microscopically. Spermatozoa that used in this study had more than 75% spermmotility and more than 25 X 108 mL in sperm concentration. Semen was divided into three equal tubesand each of them diluted with Ringer lactate egg yolk (RLEY) extender consisted of 5, 7 and 9% of DMSO.Diluted semen was added into 0.25 mL of mini straw, then equilibrated at 5 °C for two hours. The semenwas then frozen placed in liquid nitrogen vapors for10 min. Finally, the semen was put into liquid nitrogenwith a temperature of – 196oC. Post thawing quality semen was assessed after 24 hours storage bythawing the straw in warm water (37 °C) for 30 seconds. Data were analyzed using one-way analysis ofvariance. The results demonstrated that the sperm motility and viability diluted with RLEY extenderwith 7% DMSO was better (p<0.05) than those in 5 or 9% DMSO. Semen quality decreased at every stepof freezing, especially between equilibration and thawing. Its concluded that 7% was the best DMSOconcentration for freezing of native chicken semen in Ringer lactate egg yolk extender compared to 5% and9%.

Abstract

The observation on morphological body conformation of Hyla (CC), Hycole (FF), HycolexNZW (FN),New Zealand White (NN), Rex (RR), and Satin (SS) were carried out to determine estimation of mahalanobisgenetic distance. This research was held in Ciawi Bogor Livestock Research Institute (Balai PenelitianTernak Ciawi, Bogor) which 95 head of male rabbits were used. Fourteen different body parts measuredwere head (length, width, and height), ear (length and width), chest (depth, girth, and width), scapulalength, ulna length, femur length, tibia length, body length, and hips wide. CC, FF, FN, and NN rabbits hadbigger morphological size than RR and SS rabbits. Mahalanobis genetic distance showed that geneticdistance of FF and SS rabbits was the biggest (4.36). Result of canonical analysis showed that the mostdiscriminate variables were obtained by head width, ear length, ulna length, tibia length, body length, andhips wide for first canonical. The conclusion of this study that Rex and Satin rabbits were have differentgenetic distance with Hyla, Hycole, HycolexNZW and NZW rabbits.
