Abstract

Trypanosoma evansi, the agent of Surra, is a widely distributed blood protozoan infecting various species of livestock, pets and wild animals. So far, the biological character differences of T. evansi isolate from outbreak areas after re-passaging have not been reported. The aims of this study was to investigate characteristics of T. evansi collected from an outbreak area in 2012 in Sumba Island after re-passaging in mice based on pathogenicity. The study was divided into two steps of biological assays. Step 1 was addressed to observe the pathogenicity level of isolate compared to both high and low virulence controls. A total of 27 mice was divided into 9 groups (3 mice /group) e.g. negative control (KN); Positive control of high virulence (KBang87); Positive control of low virulence (KPml287); PI (Smb370); PII (Smb371); PIII (Smb372); PIV (Smb373); PV (Smb374) and PVI (Smb375). Step 2 was to look at the concistentcy of parasitemia level of isolates selected from step 1. A total of 21 mice was divided into 6 groups reinfected by Smb 372 (P VII, PVIII, PIX); Smb 375 (PX, PXI, PXII) and negative control. The results demonstrated that the parasitemia pattern of mice groups infected by Smb372 (PIII) and Smb375 (P VI) was similar to KBang87 (high virulence) and KPml287 (low virulence) group, respectively (Step 1). The step 2 results revealed that repassaging of the Smb372 isolate (PIII) in three different groups of mice (PVII, PVIII, PIX) showed similar parasitemia pattern with the origin isolates. However, the Smb375 had various parasitemia patterns among PX, PXI and PXII groups. They were moderate virulence (mixed high and low virulences) indicating that Smb375 was a mixed infection of T. evansi varians. Animals infected with either low or moderate virulence isolates need serious attention because they might act as potential reservoirs to infect other animals.

Abstract

Obesity and dyslipidemia are health problems that can increase the risk of cardiovascular disease. This study aims to analyze the potency of antiobesitas and antidislipidemia of jamu galian singset (JGS). The research method used complete randomized design with 8 treatment groups and 4 replications as follows: P1 (Standard Feed + Aquadest), P2 (Standard Feed + 200 mg Orlistat / kg BW), P3 (Standard Feed + 176,4mg JGS / 200g BW) P4 (Standard Feed + 352,8mg JGS / 200 g BW), P5 (High Fat Feed + Aquadest), P6 (High Fat Feed + 200 mg Orlistat / kg BW), P7 (High Fat Feed + 176,4 mg JGS / 200 g BW) and P8 (High Fat Feed + 352,8 mg JGS / 200 g BW). The parameters observed were feed intake, body weight gain, body weight gain ratio, fat intake, fat absorption, fat digestibility, feces fat, abdominal fat, muscle fat, body weight, body fat percentage and lipid profile, include Cholesterol total (TC), triglycerides (TG), HDL and LDL. The results showed that JGS significantly (P <0,05) decreased the body weight gain of the rat, the ratio of body weight gain per feed intake and the weight of abdominal fat. Giving JGS increases the absorption of fat, but not found high fat in the feces. Jamu galian singset has no effect (P> 0.05) on rat lipid profile. Baced on the results of this study it can be concluded that JGS can be used as antiobesity and more effectively decrease body weight gain on high fat diet than standard feeding diet. Jamu galian singset are suspected to have a different working mechanism with orlistat as antiobesitas.

Abstract

Antibiotic resistance in avian bacterial pathogens is a common problem in the Indonesia poultry industry. The present study aimed to provide information on the present status of antibiotic resistance in Escherichia coli (E. coli) from a broiler farm environment. The research method uses a cross sectional study with a stratified random sampling pattern. E. coli was collected from litter using boot swab method at the broiler house farms in Subang District. E. coli isolates (n = 74) were obtained from 74 samples of boot swabs. The sample was isolated and identified morphologically and biochemically using the IMVIC test to obtain E. coli isolates. The test was followed by a sensitivity test for nine antibiotics, namely tetracycline, sulfamethoxazole, trimethoprim, ampicillin, nalidixic acid, ciprofloxacin, enrofloxacin, gentamicin and chloramphenicol by using the agar dilution method. The interpretation of the results of this sensitivity test refers to the Clinical and Laboratory Standards Institute CLSI VET01S (2015) and CLSI M100 (2018). Measurement of the level of knowledge is done using a structured questionnaire that has been tested for validity and reliability. E. coli was isolated from 74 samples of boot swab. 93.2% of E. coli samples were found to be MDR with the highest prevalence in four types of antibiotics at 40.5%. The most common resistances observed were against tetracycline 97.3%, sulfamethoxazole 87.8%, trimethoprim 74.3%, ampicillin 68.9%, nalidixic acid 64.8%, ciprofloxacin 45.9%, enrofloxacin 40.5%, gentamicin 28.4% and chloramphenicol 10.8%. The findings of this study revealed the high level of antibiotic resistance in broiler environment is worrisome and have negative implications for human and animal health.

Abstract

The use of herbs as contraception has become the focus of current contraceptive development.. In the previous study, the extract ethanol of cogon grass roots (Imperata cylindrica) has the effect of decreasing sperm production and changing metabolite profiles in male mice. However, the effect of this Indonesian well-known herb is still indefinite in female. The purpose of this study was to investigate the effect of ethanol extract of cogon grass roots to the fertility of female mice. The study was conducted at Animal Laboratory of Faculty of Medicine and Central laboratory Universitas Padjadjaran from November to December 2017. Mice were given ethanol extract of cogon grass roots 90 and 115 mg/kg body weight per oral. After three weeks, vaginal cytology was observed and metabolite profiles were measured. The results showed a shortening of the estrus phase in 90 and 115 mg/kg treatment group. However, the level of glucose and cholesterol in serum were not different among groups. This study indicated the extract ethanol of cogon grass roots has potential effect as contraceptive agent and the effect is independent from metabolite profiles. Abstract

Red yeast rice is a fermented product of rice and mold Monascus purpureus. Red yeast rice contains secondary metabolites such as lovastatin, phenol and flavonoids that have potential as antioxidants. This study was aimed to examine the effectiveness of red yeast rice antioxidants on performance of japanese quail administered with dexamethasone through the parameters of feed digestibility, feed consumption, growth in body weight, stress index, malondialdehyde (MDA) andsuperoxide dismutase (SOD). This study used a factorial completely randomized design. The animals used in this study were 60 quails (Coturnix coturnix japonica), with a body weight of 140195 grams. Group K0 was a control, group D were quails given only dexamethasone at a dose of 1.25 mg/g BW for 7 days after one-week adaptation. Group D+VC were quails given dexamethasone at a dose of 1.25 mg/g BW then added with vitamin C at a dose of 20 mg. Group D+K1, D+K2, and D+K3 were quails given dexamethasone then orally given red yeast rice of 18 mg, 24 mg, and 30 mg for 14 days. The feed consumption, feed digestibility and body weight shows no significant difference among group (P>0.05). The heterophyl/lympocyte ratio was elevated when dexametasone administered and tended to decrease when administered with red yeast rice. Result also showed that red yeast rice has the potential as an antioxidant in quail administered with dexamethasone as seen from the tendency of decreased level of MDA and increased level of SOD enzyme. The results also showed that quail administered with dexamethasone decreased level of MDA and elevated levels of SOD enzyme in the liver after administration of red yeast rice.

Abstract

Fasciolosis is one of the endemic parasitic diseases in Indonesia. Fasciola gigantica is the main species found to infect livestock, especially beef cattle. The purpose of this study was to analyze the morphometry and identify the ultrastructure morphology of the F. gigantica worm isolated from beef cattle in Berau abattoir. The F. gigantica worms were isolated from two cattle breeds, namely Donggala cattle and Bali cattle. Worm was stained by using carmine methode to analyze the morphometric of the worm and scanning electron microscopy to identify ultrastructure morphology. Morphometric of the worm was analysed with the one sample t-test and multidimensional scaling statistical methods. The results of the morphometric analysis showed that F. gigantica from both breeds had significant differences (P<0.05) between the parameters and based on multidimensional scaling analysis had similarities with F. intermedia from Iran. Based on scanning electron microscopy examination showed that F. gigantica from the Donggala cattle breed was infected with type 1 of F. gigantica while F. gigantica from the Bali cattle breed was infected with type 2 of F. gigantica based on ventral sucker, oral sucker, and spine. It can be concluded that F. gigantica found in beef cattle in Berau was type 1 F. gigantica (Donggala cattlebreed) and type 2 F. gigantica (Bali cattle breed) and similar with F. intermedia from Iran.

Abstract

Kintamani dog growth pattern was observed by measuring the length, circumference and body weight. This study aims to obtain a pattern of growth and determine at what age the growth of kintamani dogs reaches the inflection point (begins to grow slowly) and reaches adult size (begins to stop growing). Data on growth patterns and inflection points of kintamani dogs are useful as a reference for determining the right time for the kintamani dog to be first mated, knowing the existence of abnormalities in growth and nutritional status. The research samples were 90 female kintamani dogs and 90 males aged 0 to 800 days. Measurement of body length and circumference uses a meter, while body weight is measured by digital scales. The growth curve used by the estimator is a sigmoid curve with two parameters: the size of body length, chest circumference and body weight at birth and as an adult. The conclusion of this study is that the pattern of growth in body weight, body length, chest circumference, height of the back limbs and height of the front legs of male and female kintamani dogs has a sigmoid growth pattern. The body size of a female dog has an inflection point or begins to grow slowly and reaches an adult size at a younger age than a male. Female dogs also reach the maximum size earlier than male dogs. The speed of reaching the inflection point on the growth of kintamani dogs were: Chest circumference, front limb height, body length, height of the back leg and body weight. While the speed of achieving adult size were : height of the back leg, body length, height of the front limb, chest circumference and weight.
