Abstract

ABSTRACTConstipation is a common gastrointestinal disorder frequently associated with reduced intestinal motility. Intestinal motility is regulated by the enteric nervous system, in which serotonin plays a key role as a neurotransmitter. Physical activity has been reported to influence the serotonergic system; however, its potential role as a non-pharmacological intervention for constipation remains limitedly explored. This study aimed to analyze the modulation of serum serotonin levels and intestinal motility through swimming exercise in male constipated mice (Mus musculus). This experimental study employed a post-test-only control group design. A total of 30 male mice aged 2–3 months with body weights of 20–30 g were divided into three groups: normal control (P0), constipated control without intervention (P1), and treatment group (P2). Constipation was induced using loperamide HCl at a dose of 0.04 mg/mouse/day for seven days. The P2 group subsequently underwent swimming exercise three times per week for four weeks. Serum serotonin levels were analyzed using the Kruskal–Wallis test, while intestinal motility was analyzed using one-way ANOVA. The treatment group demonstrated significantly higher mean serum serotonin levels compared to the P0 and P1 groups (p=0.000). However, no significant differences in intestinal motility were observed among the study groups (p=0.395). Swimming exercise effectively modulates serum serotonin levels in male mice with constipation, although it does not significantly improve intestinal motility. Keywords: Swimming Exercise, Serum Serotonin, Intestinal Motility, Constipation.  ABSTRAKKonstipasi merupakan gangguan gastrointestinal yang sering dijumpai dan berkaitan erat dengan penurunan motilitas usus. Regulasi motilitas usus dipengaruhi oleh sistem saraf enterik, salah satunya melalui peran serotonin sebagai neurotransmiter utama. Aktivitas fisik diketahui mampu memengaruhi sistem serotonergik, namun mekanisme ini masih jarang dikaji sebagai pendekatan nonfarmakologis pada konstipasi.Penelitian ini bertujuan untuk menganalisis modulasi kadar serotonin serum dan motilitas usus melalui aktivitas fisik renang pada mencit (Mus musculus) jantan model konstipasi. Penelitian ini merupakan studi eksperimental dengan rancangan post-test only control group design. Sebanyak 30 ekor mencit jantan usia 2–3 bulan dengan berat badan 20–30 gram dibagi menjadi tiga kelompok, yaitu kontrol normal (P0), kontrol konstipasi tanpa perlakuan (P1), dan kelompok perlakuan (P2). Konstipasi diinduksi menggunakan loperamide HCl 0,04 mg/ekor/hari selama 7 hari. Kelompok P2 selanjutnya diberikan aktivitas fisik renang dengan frekuensi tiga kali per minggu selama empat minggu. Kadar serotonin serum dianalisis menggunakan uji Kruskal–Wallis, sedangkan motilitas usus dianalisis menggunakan uji ANOVA. Kadar serotonin serum pada kelompok perlakuan (P2) menunjukkan rerata yang lebih tinggi dibandingkan kelompok P0 dan P1 (p=0,000). Namun, analisis motilitas usus tidak menunjukkan perbedaan yang bermakna antara kelompok kontrol dan kelompok perlakuan (p=0,395).Aktivitas fisik renang mampu memodulasi kadar serotonin serum pada mencit jantan model konstipasi, namun belum menunjukkan pengaruh yang signifikan terhadap motilitas usus. Kata Kunci: Aktivitas Fisik Renang, Serotonin Serum, Motilitas Usus, Konstipasi.
