Abstract

The macadamia plant (Macadamia integrifolia) is a high-quality nut-producing commodity with significant economic value; however, conventional propagation still faces limitations in producing superior seedlings in large quantities that are uniform and sustainable. This internship activity aimed to study the procedures for in vitro macadamia tissue culture, from the preparation of Murashige and Skoog (MS) medium, sterilization of explants, tools, and materials, incubation, to subculture. The activity was carried out at the UPTD Balai Sertifikasi dan Perbenihan Tanaman Hutan (BSPTH) Dinas Kehutanan Provinsi Sumatera Barat from January 5 to February 15, 2026. The explants used were young shoots of macadamia plants grown on MS medium supplemented with the plant growth regulator BAP. A total of 81 explants were observed during the culture process. The observations showed that 34 explants (41.98%) grew successfully without contamination, whereas 47 explants (58.02%) experienced fungal and bacterial contamination as well as browning. The high level of contamination was presumed to be related to the presence of endophytic contaminants, the suboptimal sterilization protocol, and the condition of explants originating from open field conditions. Browning can be managed through the addition of antioxidants, such as PVP, activated charcoal, and ascorbic acid, to the medium. This activity confirms that tissue culture has the potential to become an alternative method for macadamia propagation, but its success is highly dependent on the optimization of explant sterilization and the control of tissue browning. The implications of this activity point to the need for implementing staged sterilization and conditioning mother plants in a greenhouse to improve the success of macadamia micropropagation in subsequent studies.

Abstract

Agarwood (Aquilaria malaccensis) is one of the non-timber forest products with high economic value because it produces aromatic resin used in the perfume, cosmetics, traditional medicine, and aromatherapy industries. This internship activity aimed to study and describe the early growth and maintenance of agarwood seedlings at the Permanent Nursery of the UPTD Balai Sertifikasi dan Perbenihan Tanaman Hutan (BSPTH) Dinas Kehutanan Sumatera Barat as part of efforts to support the cultivation and conservation of agarwood plants. The activity was carried out from January to February 2026 using a descriptive method through direct observation of the seedling growth and maintenance process. The observations showed that the agarwood seeds used had good quality with high viability. Optimal seedling growth was supported by regular watering, the use of shade, loose planting media, and periodic weed and pest control. The conclusion of this activity affirms that the maintenance of agarwood seedlings in nurseries plays an important role in producing quality seedlings, supporting successful cultivation, and promoting the conservation and sustainable management of forest resources. The implications of this activity indicate the need to apply regular and adaptive nursery maintenance techniques so that the early growth of agarwood seedlings can proceed optimally.

Abstract

Bamboo plants (Bambusa sp.) have high economic and ecological value; however, conventional propagation still faces limitations in producing uniform, high-quality, and disease-free seedlings. This activity aimed to study the stages of bamboo tissue culture, including the preparation of MS medium, explant sterilization, inoculation, incubation, subculture, and acclimatization. The internship activity was carried out from January to February 2026 at the UPTD BSPTH Dinas Kehutanan Provinsi Sumatera Barat. Data were obtained through observations of the bamboo tissue culture process at each stage and were then analyzed descriptively based on the success of explant growth and the constraints that emerged during the culture process. The results of the activity showed that some explants grew successfully, whereas others experienced browning and contamination. The success of bamboo tissue culture was influenced by the accuracy of sterilization, medium composition, the use of plant growth regulators, and incubation conditions. The conclusion of this activity affirms that tissue culture techniques have the potential to become an effective alternative for mass propagation and conservation of bamboo germplasm. Its implications indicate that the optimization of sterilization procedures, medium formulation, and culture condition regulation needs to be carried out continuously to improve the success of in vitro bamboo propagation.
