The high prevalence of skin diseases in Indonesia necessitates the exploration of natural therapeutic alternatives, particularly utilizing sweet orange peel (Citrus sinensis L. Osbeck) waste. This study aimed to evaluate the antibacterial efficacy of sweet orange peel extract against skin infection pathogens, specifically Staphylococcus aureus and Staphylococcus epidermidis. Extraction was performed using the Ultrasonic Assisted Extraction (UAE) method. Antibacterial assays included the determination of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) using the solid dilution method, followed by the identification of active compounds via Thin Layer Chromatography (TLC)-Bioautography. Phytochemical screening revealed the presence of alkaloids, flavonoids, saponins, tannins, and triterpenoids in the extract. The extract demonstrated antibacterial activity with an MIC value of 0.4% and an MBC value of 25.6%. TLC-Bioautography analysis detected inhibition zones corresponding to three Rf values (0.90, 0.32, 0.16) against S. aureus and four Rf values (0.90, 0.67, 0.32, 0.16) against S. epidermidis. The bioactive compounds responsible for this inhibition were identified as alkaloids and flavonoids. In conclusion, the ethanol extract of sweet orange peel exhibits effective antibacterial activity against skin pathogens, mediated by its alkaloid and flavonoid content
