Biodiversitas 20: 1312-1317.  Endophytic actinobacteria from ginger (Zingiber officinale Rosc.) is a bacterium that is capable of producing secondary metabolites that are the same as their hosts. This study aims to look at the potential of endophytic actinobacteria from ginger as a pancreatic lipase inhibitor and its toxicity. Endophytic actinobacteria were isolated, purified, then tested for pancreatic lipase inhibitors and their toxicity using the BSLT method (Brine Shrimp Lethality Test) and phytochemical tested on ethanol extract of selected isolates. Seven endophytic actinobacterial isolates were isolated from the ginger rhizome. The isolates had different morphological diversity based on colony and microscopic observations and 5 isolates had pancreatic lipase inhibitor activity. The highest inhibitors were found in AJ4 isolates (89.9%), compared with pancreatic lipase inhibitors crude extracts of ginger (68.9%) and orlistat (88.1%) as positive controls. The LC50 value of AJ4 isolates was 653,381 ppm and the value of LT50 was 17,569 hours. AJ4 isolates contain terpenoids, phenols, tannins, flavonoids, alkaloids, and saponins. This research data is considered as new information about the potential of endophytic actinobacteria from ginger as pancreatic lipase inhibitors and their toxicity.

Abstract. Amfar F, Fitri L, Suhartono. 2021. Molecular identification of a new isolate of actinobacteria ATIS61 and characterization of the protease activities. Biodiversitas 22: 1564-1569. Protease is an enzyme that catalyzes the hydrolysis of peptide bonds in protein. Actinobacteria are one of bacterial groups that is able to produce protease. Actinobacteria are Gram-positive bacteria and mostly aerobic. This study aimed to identify protease-producing actinobacteria ATIS61 isolate using 16S rRNA gene and to characterize the protease activity. This study was experimental research, consisted of amplification and sequencing of the 16S rRNA gene, and protease activity test. The 16S rRNA gene analysis showed that ATIS61 isolate was closely related to Nocardia sp. strain 335427 with a 99.88% similarity and the result of phylogenetic tree construction was related to Nocardia farcinica strain ARS8 with Bootstrap 94%. Protease activity test showed the highest activity was on the eighth day of incubation at 0.115 U/mL. Protease activity based on temperature showed the highest activity at 40°C of 0.156 U/mL and the stability of protease towards temperature was stable at 40°C and 50°C. Protease activity showed that the highest protease activity was at pH 8 of 0.096 U/mL and the highest protease stability was also at pH 8. The addition of HgCl2 showed that it could inhibit protease activity with a value of 0.059 U/mL.

Biodiversitas 22: 5169-5180. Cellulolytic actinobacteria are cellulase-producing bacteria capable of degrading cellulose. This study aimed to isolate, characterize, evaluate the cellulolytic ability, and to determine physiological characterization of soil cellulolytic actinobacteria isolated from the Ujung Pancu area, Aceh Besar. Isolation of actinobacteria from soil samples was performed using serial dilution method on Yeast Malt Agar (YMA) medium. Morphological characterization was carried out by growing isolates on YMA, Oatmeal Agar (OA), and Yeast Starch Agar (YSA) media. Cellulolytic ability was determined by calculating the cellulolytic index (IS) on 1% carboxymethyl cellulose (CMC) medium after adding 0.1% congo red solution. Physiological characterization of cellulolytic actinobacteria tested in this study was salinity, pH, and carbon source in liquid Yeast Malt (liquid YM), and the growth was measured at a wavelength of 581nm. The results showed that a total of nine isolates of actinobacteria were isolated, which belonged to the genus Streptomyces. Cellulolytic test results showed that eight isolates had the ability to degrade cellulose. Isolates AUP-04, AUP-03, and AUP-01 had the highest cellulolytic index value. Physiological characterization results revealed that three isolates had different tolerances for salinity levels, pH, and types of carbon sources. AUP-03 isolate grew well at 10% salinity with an OD value of 0.88, isolate AUP-01 grew at 5% salinity with an OD value of 0.49, whereas isolate AUP-04 grew well on media that did not contain salinity. All three isolates grew well at pH 6 with OD values of 0.93, 1.12, and 1.27. AUP-03 and AUP-01 isolates grew well on media containing dextrose as carbon source with OD values of 0.154 and 0.17, respectively, while isolate AUP-04 grew well on glucose-containing media with an OD value of 0.22.

Daily behavioral research on Sumatran Orangutan (Pongo abelii) mothers and offsprings at Suaq Balimbing Research Station, Gunung Leuser National Park, Aceh Province, Indonesia. Biodiversitas 23: 5108-5121. Discovering the daily behavior of female orangutan mothers and their offspring can provide information for use as a recent reference in orangutan conservation action plans. This research aims to determine the daily behavioral patterns of mother and offspring Sumatran orangutans (Pongo abelii) at the Suaq Balimbing research station. This study was conducted between June 2019 and March 2020 and used focal animal sampling as the method. The method used consists of searching and daily behavioral logging. The results of the research showed that six orangutan mothers and offsprings (three mother-offspring pairs) showed different behavioral patterns. The distinction was felt in the distribution of time spent on activities. Eating, moving, resting, nesting, and social activity are all part of an orangutan's daily behavior. The proportion of time allocation used in the daily activities of female mother and Sumatran orangutan pairs (Pongo abelii) that is successfully observed in the Suaq Balimbing research station are eating (68.79%), resting (11.59%), moving (11.02%), nesting (1.11%), others (5.04%) and unknown (2.45%). Orangutans' diet plants in Suaq Balimbing consist of 30 species from 25 families, including the liana diet, plants consumed with the seeds, and plants consumed by the bark. According to the findings of this study, the behavior of a mother Sumatran orangutan, while it is with its offspring can be described.
