Abstract

Sangkareho (Callicarpa longifolia Lam.), a plant in the Lamiaceae family, is utilized as traditional medicine in Kalimantan. Its leaves are rich in flavonoids, known for their pharmacological activities like antimicrobial, anti-inflammatory, and astringent properties. This study aimed to conduct phytochemical screening and to determine the total flavonoid content of sangkareho leaf extracts obtained through sequential extraction using n-hexane, ethyl acetate, and ethanol 96%, analyzed by UV- Vis spectrophotometry. Phytochemical screening showed that the sangkareho leaf extract contains flavonoids, tannins, saponins, steroids/triterpenoids, and alkaloids. Total flavonoid content was quantified using quercetin as the standard and expressed as quercetin equivalents (QE) per gram of extract.The Result was shown that, the average total flavonoid content in n-hexane, ethyl acetate, and 96% ethanol extracts were 43.93 QE/g extract, 65.80 QE/g extract, and 92.21 QE/g extract, respectively. The data processing by One Way Anova with a p value <0.05, indicating a significant influence of multistage extraction methods on determining the total flavonoid content in Sangkareho leaf extracts.
