Abstract

Abstract Purification is a process of refining to remove interfering components such as chlorophyll. This study was conducted to assess the effectiveness of 96% ethanol extract, 70% ethanol extract, hexane-soluble extract, and hexane-insoluble extract from red betel leaves against Streptococcus mutans by examining their differences. The red betel leaves were macerated with 96% ethanol and 70% ethanol. After the extraction process with 96% ethanol, the extract was then purified using hexane solvent. This purification process resulted in hexane-soluble extract and hexane-insoluble extract. The antibacterial effectiveness was tested using the well diffusion method on each extract at a concentration of 15%. Based on laboratory tests, the inhibition zone diameters for the 96% ethanol extract, 70% ethanol extract, and hexane-insoluble extract showed differences in diameter of 5.27 mm, 10.4 mm, and 7.25 mm, falling under the categories of moderate, strong, and moderate inhibition, respectively. However, the purified hexane-soluble extract did not show any inhibition zone. The presence of alkaloid, flavonoid, saponin, tannin, and steroid compounds is suspected to trigger this antibacterial activity. ANOVA analysis showed significant differences among these extracts. Based on the results obtained, it was concluded that there are differences in effectiveness between the 96% ethanol extract, 70% ethanol extract, and hexane-insoluble extract as antibacterials against Streptococcus mutans.
