Background: Type 2 diabetes mellitus is a chronic metabolic disorder characterized by impaired insulin function, elevated blood glucose levels, and abnormal lipid profiles, including HDL. Purpose: To determine the effect of Musa paradisiaca L. (Kepok banana) peel extract on random blood glucose (RBG) and HDL levels in a type 2 diabetes mellitus mouse model. Method: A true experimental study with a pretest-posttest control group design. A total of 20 male mice were divided into four groups: normal control, negative control, positive control, and treatment group, each consisting of five mice. The negative control, positive control, and treatment groups were induced with alloxan. The positive control group was given metformin, while the treatment group was given kepok banana peel extract for 14 days. The data were analyzed using normality and homogeneity tests, one-way ANOVA, and the Games-Howell post hoc test. Results: The treatment group showed the greatest reduction in GDS, with a ΔGDS of 112.4 mg/dL, followed by the positive control at 99.6 mg/dL, the negative control at 4.8 mg/dL, and the normal control at 1.4 mg/dL. The ANOVA test showed a significant difference in ΔGDS between groups (p=0.031), but the Games-Howell test did not show significant differences between pairs of groups: KN vs K- (p=0.973), KN vs K+ (p=0.222), KN vs P (p=0.170), K- vs K+ (p=0.266), K- vs P (p=0.200), and K+ vs P (p=0.996). HDL levels in the treatment groups were 88.39 ± 22.98 mg/dL, with no significant differences between groups (p=0.788). Conclusion: Kepok banana peel extract tends to lower GDS, but has no significant effect on HDL.
