Abstract

Background: Analgesics are widely used to relieve pain without causing loss of consciousness. However, prolonged use may lead to serious adverse effects, leading to the need for safer natural alternatives. White turmeric (Curcuma zedoaria) contains curcumin, while red ginger (Zingiber officinale Rosc. var. rubrum) contains gingerol, both of which possess analgesic properties. Objective: This study evaluated the analgesic activity of combined ethanol extracts of white turmeric and red ginger using writhing and hot plate tests in mice. Methods: The experimental design consisted of seven groups: negative control (CMC-Na), positive controls (mefenamic acid and tramadol), single extracts of white turmeric (500 mg/kg) and red ginger (600 mg/kg), and three extract combinations (25:75, 50:50, and 75:25). In the writhing test, pain was induced using 0.6% acetic acid, while the hot plate test measured responses to thermal stimulation. Data were analyzed using one-way ANOVA followed by the LSD post hoc test. Results: The combination containing 75% white turmeric extract (315 mg/kg) and 25% red ginger extract (150 mg/kg) showed the strongest analgesic activity among all treatment groups. Conclusion: The combination of white turmeric and red ginger ethanol extracts demonstrated effective analgesic activity, with the 75:25 combination producing the most potent effect in mice.
