Abstract

This study aimed to evaluate the effect of green tea extract (Camellia sinensis) supplementation in Tris–egg yolk extender on the post-cryopreservation quality of cauda epididymal spermatozoa of Simmental crossbred cattle. Spermatozoa were obtained from the cauda epididymis of adult Simmental crossbred bulls collected at the Padang City Slaughterhouse. The study employed a Randomized Complete Block Design (RCBD) with four treatments and five replications, namely P0 (Tris–egg yolk without green tea extract), P1 (0.1 mg/mL green tea extract), P2 (0.2 mg/mL green tea extract), and P3 (0.3 mg/mL green tea extract). Post-thaw sperm quality parameters observed included sperm viability, motility, and morphological abnormalities after the freezing and thawing process (post-thawing). The results demonstrated that green tea extract supplementation significantly improved sperm viability and motility (P = 0.032; P < 0.05) while reducing morphological abnormalities. Treatment P2 showed the highest values, with viability of 56.4 ± 3.2%, motility of 53.4 ± 3.2%, and abnormalities of 10.8 ± 1.8%. The antioxidant activity of polyphenols, particularly epigallocatechin gallate (EGCG), is believed to protect sperm plasma membranes and mitochondria from oxidative stress-induced damage during cryopreservation. A concentration of 0.2 mg/mL appeared to be the most effective concentration under the conditions of this study.
