Abstract

Liquid semen preservation extends the usable period of rooster semen for artificial insemination, but sperm quality progressively deteriorates during chilled storage. This study evaluated the effect of storage duration on motility, viability, and plasma membrane integrity of KUB rooster sperm in a lactated Ringer-egg yolk extender at 4°C. Semen was collected from five 48-week-old KUB roosters. Ejaculates with initial motility above 75% and sperm concentration above 2.5 billion cells/mL were diluted and assigned to four storage durations: 0, 5, 24, and 48 h, with four replications. Data were analyzed by one-way ANOVA followed by Tukey's HSD test at a 1% significance level. Storage duration significantly affected all sperm-quality variables (P < 0.01). Motility declined from 84.25% at 0 h to 72.00%, 53.16%, and 33.41% at 5, 24, and 48 h, respectively. Viability decreased from 92.25% to 47.08%, while plasma membrane integrity decreased from 92.25% to 47.83% after 48 h. The 24-h samples retained motility above 50% and comparatively higher viability and membrane integrity than 48-h samples. Thus, the extender supported short-term chilled preservation of KUB rooster semen for up to 24 h based on in vitro sperm quality.
