Abstract

The purpose of this study was to examine the effect of medium combination of IVM and IVC on the in vitro development of bovine embryos. The study involved 4 groups in a 2 (IVM medium) x 2 (IVC medium) factorial in a randomized block design. Each group was replicated for 5 times. The treatments were as follows: TCM-199/CR1aa (T1); TCM-199/SOF (T2); B- 199/CR1aa (T3) and B-199/SOF (T4). Oocytes were aspirated from ovaries collected at local abattoirs using aspiration medium of PBS supplemented with 3% FCS and 0.1% Penicillin and Streptomycin. The oocytes were matured in medium of TCM-199 or B-199 supplemented with 10% FCS, hormones: 10μg/ml FSH+ 10μg/ml hCG+ 1μg/ml Estradiol. Maturation was maintained at 37oC for 22 hours in 5% CO2 incubator with high humidity. A method of BRACKETT & Oliphant (BO) was used to fertilize the matured oocytes. The fertilization was incubated for 7 hours in the 5% CO2 incubator. Two culture media of CR1aa or SOF/AA/BSA were used to develop the fertilized oocytes undergo to morula and blastocyst embryos. The findings showed that the proportion of oocytes cleaved and formation of blastocysts were affected significantly by a combination of IVM and IVC media (P<0.05). A combination of B-199/SOF (T4) resulted in a higher blastocyst rate (32%) than others (T3= 29%; T2=T1= 23%). This study suggests that either SOF/AA/BSA or CR1aa has similar competence in development of bovine embryos in vitro.   Key words: Bovine embryo, IVF, maturation medium, culture medium

Abstract

Research was done to study the profile of morphological characteristics of Simmental cross cattle in West Sumatera on several age categories and estimating the genetic distance compared to Simmental pure breed based on morphological parameters. Several morphological parameters of the 82 of Simmental cross cattle in West Sumatera and the 23 of Simmental pure breed has been measured. To analyze morphological data and the genetic distance a discriminant analysis was done. Based on statistical tests (t-test), body length, body height, hip height, hip width, chest circumference, chest width, chest depth, head length, and head height had significant difference (P<0.05) between 24-60 months Simmental pure breed and Simmental cross cattle bulls. Based on the similarity phenotypic analysis, it is known that all subpopulations of cattle in this research had a relatively high value (73.68-78.57%). Chest depth was the highest different morphometric factor (0.910). Morphological distribution map indicated that the Simmental cross cattle (subpopulations Agam and Lima Puluh Kota) evenly distributed across all quadrants (I-IV) while the Simmental pure breed (subpopulations BIB Lembang and BIBD Tuah Sakato) distributed only in the I quadrants and II quadrants. The highest genetic distance was identified between cattle in Agam and BIBD Tuah Sakato while the lowest genetic distance was identified between cattle in Agam and Lima Puluh Kota. Based on phenogram tree structure, the observed Simmental cattle could be classified into 2 clusters, subpopulations of BIBD Tuah Sakato and BIB Lembang that represent Simmental pure breed configured their own cluster as well as a subpopulation of Agam and Lima Puluh Kota that represent Simmental cross cattle configured the others. Abstract

ABSTRAK Penelitian ini bertujuan untuk meningkatkan motivasi dan prestasi belajar matematika siswa kelas v semester II dengan penerapan model pembelajaran Talking Stick. Metode pengumpulan data yang digunakan adalah metode angket dan metode tes. Hasil penelitian menunjukkan bahwa penerapan model pembelajaran Talking Stick dalam proses pembelajaran dapat meningkatkan motivasi dan prestasi belajar matematika. Keadaan ini ditunjukkan dengan adanya peningkatan motivasi pada siklus I sebesar 86,27% dimana rata-rata motivasi belajar siswa termasuk dalam kategori sangat baik. Sedangkan prestasi belajar matematika siswa dapat dilihat dari tes UTS-II dengan rerata 69,31 dan persentase ketuntasan klasikal sebesar 77,27%, setelah pelaksanaan tindakan pada siklus I ada peningkatan rerata sebesar 86,31 dengan persentase ketuntasan klasiskal sebesar 86,36%. Demikian dapat disimpulkan bahwa dengan adanya penerapan model pembelajaran Talking Stick dengan media LKS dapat meningkatkan motivasi belajar dan prestasi belajar matematika siswa dikelas V SD Negeri Girirejo Kecamatan Ngombol Kabupaten Purworejo tahun pelajaran 2012/2013.   Kata kunci: motivasi,prestasi,model pembelajaran talking stick, LKS
