Abstract

Background: Indonesia is the second-highest country with hepatitis B cases in the South East Asian region. Clustered regularly interspaced short palindromic repeats (CRISPR)-CRISPR associated protein 12 (Cas12) could be developed as a diagnostic tool to detect hepatitis B infection. This study was aimed to develop a diagnostic method for hepatitis B virus by designing CRISPR target-based single-guide RNA (sgRNA).Materials and method: The preCore/Core-gene sequences of hepatitis B virus were collected from the National Center for Biotechnology Information (NCBI) website. The selected sequence was submitted to Cas Designer and CRISPOR tools to design sgRNA. The resulting sgRNA was cloned in silico into an expression vector using Benchling software.Results: The 23-nucleotide sequence 5'- GTAGTCAGTTATGTCAATGTTAA-3’ had 30% GC content, 68.3 out-of-frame and 76 predicted efficiencies. This sequence had no mismatch based on analysis.Conclusion: This preliminary study will help design a CRISPR-based diagnostic kit for the detection of hepatitis B virus in Indonesia. However, further in vitro and in vivo studies are required to demonstrate its potential and efficiency.Keywords: CRISPR-Cas12b, diagnostic, HBV, sgRNA

Abstract

Obesity is a condition where an individual's body weight is abnormal (BMI >30), leading to excessive fat accumulation. This condition can induce type 2 diabetes mellitus, which results in abnormal insulin production, causing blood sugar levels to spike. This study aims to determine the strength of the correlation between BMI and blood glucose level using a cross-sectional method. We used bivariate data collection, and Pearson's method to determine the strength of the correlation between these two variables. In this study, data from 102 individuals were collected from people in Reformed Millenium Center Indonesia (RMCI) Jakarta. The correlation between BMI and blood glucose levels with r value 0.82 and significance 2-tailed p-value 0.411. The analysis showed a weak positive relationship, but the correlation is not statistically significant. A suggestion for future research is to use body fat percentage as a reference instead of BMI.

Abstract

Infeksi virus hepatitis B (VHB) kronis ditandai dengan keberadaan covalently closed circular DNA (cccDNA) VHB. Metode diagnostik VHB umumnya berbasis molekuler dan serologi, tetapi metode ini memiliki kekurangan terutama di dalam mendeteksi jumlah cccDNA VHB yang rendah, mendeteksi VHB di awal tahapan infeksi, dan membedakan cccDNA dari relaxed circular DNA (rcDNA). Artikel ini bertujuan untuk membandingkan sistem diagnostik konvensional (molekular dan serologi) dengan sistem diagnostik berbasis CRISPR-Cas yang dapat menjadi alternatif uji diagnostik VHB. Metode penelitian menggunakan studi literatur mengenai diagnostik VHB secara konvensional dan metode CRISPR-Cas untuk VHB yang memenuhi kriteria inklusi dan eksklusi. Prinsip metode CRISPR dalam sistem penyuntingan genom melibatkan CRISPR-associated nuclease (Cas) dalam memodifikasi nukleotida asing yang bekerja dalam tiga tahapan yaitu adaptasi, ekspresi, dan intervensi. Beberapa penelitian penggunaan CRISPR-Cas dalam diagnostik VHB menunjukkan bahwa CRISPR-Cas dapat mendeteksi cccDNA VHB dengan sensitivitas dan spesifisitas yang tinggi. Simpulan adalah metode CRISPR-Cas dapat dikembangkan sebagai alternatif uji diagnostik infeksi VHB.
