Abstract

Diabetes is marked by elevated levels of blood glucose, and progressive changes of the structure of pancreatic islet histopathology. The objective of this research was to analyse the glucose level and histophatological feature in pancreatic islet in alloxan compound induced rats. A total of ten male Spraque Dawley rats of 2 months old were used in this study. The rats were divided into two groups: (1) negative control group (K-), and (2) positif induced alloxan group (diabetic group =DM). The rats were induced by a single dose intraperitonial injection of alloxan compound 120 mg/kg of body weight. The treatment was conducted for 28 days. Blood glucose levels of rats were analysed at 0, 4, 7, 14, 21, and 28 days following treatment. At the end of the experiment, rats were sacrificed by cervical dislocation. Pancreas was collected for analysis of histopathological study by Immunohistochemical technique, and ultrastructural study using transmission electron microscope (TEM). The result showed that Langerhans islet of diabetic rat (rat of DM group) showed a marked reduction of size, number of Langerhans islet of diabetic rat decrease, and characterized by hyperglycemic condition. By using TEM, beta cells of DM group showed the rupture of mitochondrial membrane, the lost of cisternal structure of inner membrane of mitocondria, reduction of insulin secretory granules, linkage between cells acinar with free Langerhans islet, and the caryopicnotic of nucleus. Abstract

This research was conducted to investigate the antibacterial activity of powder of temulawak, ginger and garlic to S.typhimurium using modified agar well method. Preliminary study showed that garlic powder had the best antibacterial activity, therefore it was further tested in a feeding trial to evaluate the effect on growth performance and immune response in broiler chicken challenged with S. typhimurium (ST). Experimental treatments were arranged in Completely Randomized Design with four treatment and three replications (Negative control, positive control, garlic powder and tetracycline). Seventy-two Day-Old-Chickens with body weight 46,7 g strain “Hubbard Wonokoyo” were used in a 28 days experiment. Broiler chickens were fed garlic powder and tetracycline diets for 10 days and then challenged orally with ST 4,1 x 1011 cfu. Body weight, feed intake and salmonella colony in faeces were monitored. Blood serum was collected at 18-d after infection. Results indicated that there was no significant effect of garlic powder on body weight and feed intake, but feed intake tended to decrease. However, feed conversion ratio of ration with garlic powder was better than rations with tetracycline and other treatments. Salmonella population in faeces also decreased with addition of garlic powder in diets. Total protein serum was influenced by disease challenged. Serum immunoglobulin (gamma globulin) was not influenced by disease challenged, but addition garlic powder in diet tended to increase gamma-globulin concentration. It can be concluded that garlic powder has antibacterial activity to S.typhimurium. Furthermore, this result indicated that some beneficial effect of dietary garlic powder at 2,5% supplementation on growth performance and no effect on immune response in the presence of ST-challenge. Abstract

The antimicrobial activity of garlic extract against Gram-positive and Gram-negative bacterial isolates was well studied. However, reports on antibacterial activity of garlic extract against some pathogenic bacteria in animals in Indonesia, are still limited. Therefore, the aim of this study was to evaluate the antibacterial activity of water and ethanol extracts of garlic against Salmonella typhimurium in chickens, and Streptococcus agalactie, Escherichia coli, and Staphylococcus aureus causing mastitis in dairy cows in Indonesia. A filtrate of fresh garlic was used to determine the antibacterial activity against S. typhimurium at concentrations of 5%, 10%, 20%, 30%, and 40% w/v, whereas, the antibacterial activity of water and ethanol extracts was determined against S. agalactie, E. coli, and S. aureus at concentrations of 5%, 10%, 15%, 20%, and 25% w/v. Results showed that antibacterial activity of 30% garlic filtrate was equivalent to 10% tetracycline. Meanwhile, antibacterial activity of garlic aqueous extract on mastitis bacteria was better than that of the garlic ethanol extract. Aqueous extract of garlic at 20% had the same antibacterial activity as 0.01% ampicillin on mastitis bacteria. Filtrates of fresh garlic can be used to inhibit growth of S. typhimurium and mastitis bacteria.
