Abstract

In Vitro Culture Application in the form of Somaclonal Variation Combined with Mutagen Introduction for Plant Improvement. Fatmawati is a new type of rice potentially to be developed. The development of this new type of rice in various places of West Java, Central Java and Lampung is often hampered by the blast disease causing the empty grain resulted in the harvest failure. Hence, from January to December 2007. The Indonesia Research Institute for rice in cooperation with Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development conducted research aimed at improving the quality of Fatmawati type of rice through somaclonal variation by mutative induction. In this research, the calli were treated with 1-50 gy gamma ray prior to its regeneration. The shoots produced by this regeneration were then acclimatized in the green house until the production stage. All 342 somaclone lines were sub-sequently tested on its endurance against leaf blast disease using three races of blast isolate namely 001, 033, and 173. The research yielded 21 somaclone lines which were absolutely tolerant to blast disease. These new somaclones were then planted in the green house for further morphological and agronomical observation. AbstrakFatmawati merupakan varietas padi tipe baru yang mempunyai potensi hasil tinggi. Pengembangan varietas tersebut di beberapa daerah seperti Jawa Barat, Jawa Tengah, dan Lampung masih mengalami masalah berupa bulir hampa tinggi dan serangan penyakit blas yang menyebabkan kegagalan panen. Penelitian pemuliaan untuk memperbaiki sifat unggul pada varietas Fatmawati telah dilakukan oleh Balai Besar Penelitian Padi bekerjasama dengan Balai Besar Penelitian dan Pengembangan Bioteknologi dan Sumberdaya Genetik Pertanian melalui keragaman somaklonal menggunakan induksi mutasi. Penelitian dilakukan pada bulan Januari-Desember 2007. Bahan tanaman yang digunakan adalah kalus yang diberi perlakuan irradiasi dengan sinar gamma dosis 1-50 Gy kemudian diregenerasikan pada media MS + BA 1 mg/l +IAA 0,8 mg/l. Tunas yang dihasilkan kemudian diaklimatisasi di rumah kaca sampai menghasilkan benih. Sebanyak 342 galur somaklon diuji ketahanannya terhadap penyakit blas daun menggunakan ras isolat 001, 033, dan 173. Hasil penelitian menghasilkan 21 galur somaklon yang sama sekali tidak terserang penyakit blas. Galur somaklon tahan tersebut selanjutnya ditanam di rumah kaca untuk diamati keragaman morfologi dan agronominya.

Abstract

Agronomic Characters and Resistance of Several Dihaploid Maintainer Lines to Bacterial Leaf Blight. Bacterial blight (Xanthomonas oryzae pv. oryzae, Xoo) is one of the most serious diseases of rice in Indonesia. From previous research thirteen lines of dihaploid (DH) maintainer lines-derived anther culture were selected for developing cytoplasmic male sterile lines. In this research those DH maintainers were evaluated for their agronomic characters such as plant height, number of productive tiller, and seed weight per hill as well as their resistance to Bacterial Leaf Blight (BLB) pathotypes III, IV and VIII at Indonesian Center Rice Research (ICRR), Sukamandi during wet season 2008/2009. The results showed that 10 DH maintainer lines i.e. BioMAc18-H36-3-Ma, BioMAc19-H36-3-Mb, BioMAc20- H36-3-Mc, BioMAc21-H36-4-M, BioMAc26-B1-1-Mb, BioMAc29-B2-1-Db, BioMAc31-B2-1-M, BioMAc33-B2-4- Pb, BioMAc34-B4-1-Da and BioMAc35-B4-1-Dc having plant height ranged from 88.79-104.08 cm, productive tiller ranged from 9 to 13 tillers/hill. Among those DH maintainer lines three lines were resistant to BLB pathotype III, i.e. BioMAc26-B1-1-Mb, BioMAc29-B2-1-Db and BioMAc31- B2-1-M lines, and two lines, i.e. BioMAc21-H36-4-M and BioMAc35-B4-1-Dc were highly resistant to BLB pathotype VIII. Only BioMAc35-B4-1-Dc lines highly resistant to BLB pathotype IV. AbstrakHawar daun bakteri yang disebabkan oleh bakteri Xanthomonas oryzae pv. oryzae, (Xoo) adalah salah satu penyakit utama padi di Indonesia. Dari penelitian sebelumnya 13 galur pelestari dihaploid (DH pelestari) yang berasal dari kultur antera telah diseleksi untuk perakitan galur mandul jantan baru. Tujuan penelitian ini adalah untuk mengevaluasi karakter agronomi dan ketahanan galur-galur DH pelestari terhadap patogen HDB. Karakter agronomi yang diamati meliputi tinggi tanaman, jumlah anakan produktif, dan bobot hasil per rumpun, sedangkan ketahanan terhadap HDB diamati berdasarkan skor ketahanan terhadap Xoo patotipe III, IV dan VIII di Balai Penelitian Padi, Sukamandi pada musim hujan 2008/2009. Hasil penelitian menunjukkan 10 galur DH pelestari, yaitu galur BioMAc18-H36-3-Ma, BioMAc19-H36-3-Mb, BioMAc20- H36-3-Mc, BioMAc21-H36-4-M, BioMAc26-B1-1-Mb, BioMAc29-B2-1-Db, BioMAc31-B2-1-M, BioMAc33-B2-4- Pb, BioMAc34-B4-1-Da, dan BioMAc35-B4-1-Dc mempunyai tinggi tanaman berkisar antara 88,79-104,08 cm, anakan produktif berkisar antara 9-13 batang/rumpun. Di antara galur DH pelestari yang tahan terhadap HDB patotipe III, yaitu galur BioMAc26-B1-1-Mb, BioMAc29-B2-1-Db dan BioMAc31- B2-1-M, dua galur yaitu galur BioMAc21-H36-4-M dan BioMAc35-B4-1-Dc sangat tahan terhadap HDB patotipe VIII, sedangkan galur BioMAc35-B4-1-Dc yang sangat tahan terhadap HDB patotipe IV.

Abstract

Development of Very Early Maturing Rice Genotypes through Anther Culture. Iswari S. Dewi, A. Dinar Ambarwati, Aniversari Apriana, Atmitri Sisharmini, Ida H. Somantri, Bambang Suprihatno, and Iman Ridwan. Rice is the most important food crop in Indonesia. Increase in production is needed due to population increase. Rice production in rainfed area is contributed the second after irrigated area. Rainfed condition requiring very early maturity (90-104 days) varieties. Rice anther culture can be applied to accelerate obtainment of doubled haploids (DHs) or pure lines needed in rice breeding. The experiment was aimed to obtain pure lines for developing very early maturing and high yielding rice varieties. Materials used for anther culture were F1s of Fatmawati/Kinamase, Inpari 1/Kinamase, Fatmawati/ Waseaikoku, Inpari 1/Waseaikoku, Fatmawati/IR71146, Inpari 1/IR71146, OM4495/Silugonggo, IR7146/Dodokan, and IR71730/OM1490. Anther culture media were N6 + NAA 2,0 mg/l + kinetin 0,5 mg/l for callus induction, MS+ NAA 0,5 mg/l + kinetin 2,0 mg/l for plantlet regeneration, and MS + 0,5 mg/l IBA for rooting. Putrescine 10-3 M was added to callus induction and regeneration media. The results shown that calli forming green plantlet (CFGP) were ranged from 0.25 to 83.33%. Fatmawati/Kinamase gave the highest CFGP (245 calli), followed by Inpari 1/Kinamase (78 calli) and Fatmawati/ Waseaikoku (68 calli). Total green plantlets obtained were 2.038 plantlets. After plantlet acclimatization and greenhouse grow-out, we obtained 507 DHs. The evaluation of 100 DHs at farmer field (Ciranjang District in Cianjur), based on their 50% heading date of 65 days, resulted in 33 lines cathegorized as very early maturing lines (+100 days). They were 18 lines from Fatmawati/Kinamase, 5 lines from Inpari 1/Kinamase, 8 lines from Fatmawati/Waseaikoku, and 2 lines from Inpari 1/ Waseaikoku. AbstrakPadi (Oryza sativa L.) merupakan komoditi pangan terpenting di Indonesia. Peningkatan produksi diperlukan seiring dengan peningkatan jumlah penduduk. Lahan sawah tadah hujan merupakan lumbung padi kedua setelah sawah irigasi. Kondisi lahan sawah tadah hujan memerlukan varietas-varietas padi berumur sangat genjah (90-104 hari). Teknik kultur antera dapat digunakan untuk mempercepat perolehan tanaman dihaploid (DH) atau galur murni dalam pemuliaan padi. Penelitian ini bertujuan untuk mendapatkan galur-galur murni yang akan digunakan dalam perakitan padi berdaya hasil tinggi dan berumur sangat genjah. Bahan tanaman yang digunakan untuk kultur antera adalah malai dari tanaman F1 hasil persilangan Fatmawati/Kinamase, Inpari 1/Kinamase, Fatmawati/Waseaikoku, Inpari 1/Waseaikoku, Fatmawati/IR71146, Inpari 1/ IR71146, OM4495/Silugonggo, IR7146/Dodokan, dan IR71730/OM1490. Media kultur antera adalah N6 + NAA 2,0 mg/l + kinetin 0,5 mg/l untuk media induksi kalus, MS+ NAA 0,5 mg/l + kinetin 2,0 mg/l untuk media regenerasi, dan MS + 0,5 mg/l IBA untuk media perakaran. Putresine 10-3 M ditambahkan pada media induksi kalus dan regenerasi. Hasil penelitian menunjukkan bahwa kalus yang menghasilkan tanaman hijau (KMTH) berkisar antara 0,25-83,33%. Persilangan Fatmawati/ Kinamase memberikan KMTH tertinggi (245 kalus), diikuti oleh Inpari 1/Kinamase (78 kalus) dan Fatmawati/ Waseaikoku (68 kalus). Total tanaman hijau yang diperoleh adalah 2.038 planlet dihaploid, namun diperoleh 507 tanaman setelah planlet diaklimatisasi dan tanaman ditumbuhkan di rumah kaca. Evaluasi terhadap 100 DH dilakukan di lahan petani Ciranjang, Cianjur. Berdasarkan hari berbunga 50% (65 hari setelah semai), diperoleh 33 galur yang termasuk kategori sangat genjah (dipanen +100 hari). Galur-galur tersebut adalah 18 galur dari persilangan Fatmawati/Kinamase, 5 galur dari persilangan Inpari 1/Kinamase, 8 galur dari persilangan Fatmawati/ Waseaikoku, dan 2 galur dari persilangan Inpari 1/ Waseaikoku.
